담배거세미나방에서 분리된 항균펩티드 유전자의 대량 발현에 관한 연구

The antibacterial gene isolated from the common cutworm. Spotoptera litura showed high degree of homologies in its nucleotide sequence to those of the cecropin genes from Hyalophora cecropia, implying that it may encode the antibacterial peptide. In this experiment, the antibacterial gene from S. litura was subcloned to the expression vector pGEX-1λT and overexpressed in BL21. The antibacterial gene is composed of total 114 nucleotides which encodes the relatively small peptide of 37 amino acid residues. This gene was designed to make a fusion protein with glutathione-S-transferase in response to the addition of IPTG. The total proteins from the cell extract were separated by SDS-PAGE and the overexpressed fusion protein was detected at the right positon in terms of its molecular weight. The fusion protein was also purified by use f the affinity column chromatography technique with the resin in which glutathione was cross-linked to 4% agarose bead.
사람 혈청 알부민에 대한 단세포군 항체의 제조와 특성 연구

Serum albumin은 혈청 단백질 중 50-60%를 차지한다. 알부민은 순환계 내에서 삼투압을 유지시켜 세포내외의 체액을 유지 및 지용성 물질의 이동에 관여한다. 신사구체와 기저막의 투과성이 증가하거나 세뇨관의 흡수감소 등으로 단백질의 요배설량이 하루에 150mmg 이상되는 단백뇨는 신장내의 병변이 있음을 나타내는 중요한 지표이다. 이러한 단백뇨는 크게 사구체성 단백뇨, 세뇨관성 단백뇨, 혼합성 단백뇨로 나눌수 있는데 이중 사구체 기저막의 손상으로 발생되는 사구체성 단백뇨는 알부민과 같은 고분자량의 단백질이 소변 내로 배설되는 현상이다. 일반적으로 알부민이 정상치 이상으로 소변을 통하여 배설되는 것을 'microalbuminuria'라고 하며 당뇨병으로 인한 신장병변을 예견하는데 중요한 지표로 알려져 있다. Microalbuminuria는 방사 면역확산법, 방사성 면역측정법, 면역혼탁측정법, 비탁측정법, 효소결합면역측정법(ELISA) 등으로 겸출할 수 있으나 최근에는 안전하면서도 민감도가 높고 측정이 용이한 방법인 ELISA를 이용한 면역분석방법이 많이 사용되고 있다. 면역특이성이 떨어지는 다세포군 항체를 사용한 방법의 문제점을 극복하기 위해서 세포융합기술을 이용한 단세포군항체를 생산하는 세포주를 개발하였고 그 특성을 규명하였다. 따라서 본 연구에서 세포융합기술을 도입하여 개발된 인간혈청 알부민에 대한 단세포군항체는 glycohemoglobin에 대한 단세포군항체와 더불어 당뇨병의 진행 정도를 진단할 수 있는 키트의 원료로 사용될 수 있다.

This study was carried out to investigate the purification effect of wastewater included LAS by electrolysis treatment system. Wastewater included LAS treated electrolysis treatment process, temperature of inner treated system were high than those of this raw wastewater, temperature change was electrolysis oxidation of LAS wastewater both cathode and anode. Electrolysis oxidation was oxidized LAS by oxidants of chlorine and ozone, on the time, according to the result development of treated efficiency on electrolysis contact time.

In this paper, the effect of shot peening on the fatigue characteristics have been investigated. Two types of specimens were used to obtain the experimental results. One type of specimen was made by machining and heat-treating SCM415 steel. The other type of specimen was made by machining, heat-treating, and shot peening SCM415 steel. The hydraulic testing machine was used to do the fatigue test (R=0.1, 10Hz). The experimental results show that the fatigue life and strength of shot peened gear were largely improved compared with the unpeened gear.

Study on Method of Sperm Aspiration and Injection into an Oocyte in Intracytoplasmic Sperm Injection(ICSI) Immobilization of spermatozoa prior to intracytoplasmic sperm iniection(ICSI) sometimes results in crooked tail and this makes it difficult to aspirate sperm into an injection pipette tail first. Head-first sperm aspiration into an injection pipette avoid this problem due to the bigger size of the sperm head. The effect of head or tail-first sperm injection into an oocyte on fertilization cleavage, percentage of grade I embryos and development to blastocyst stage in ICSI program has been studied. A single living immobilized spermatozoa from oligoasthenozoospermic patient was injected into an oocyte head-first or tail-first according to the treatment. Eighteen hours after microinjection, oocytes ware inspected for survival and fertilization Fertilized oocytes with two pronuclei were cultured in 30μl drop of mHTF supplemented with 10% heat-inactivated follicular fluid(FF) at 37℃. On day 2. embryo transfer was performed with cleaved embryos. The remaining 2-8 cell stage embryos were co-cultured with BRL cells in mHTF + 10% FF for 72 hours and the developmental stage was observed. The data were analyzed by Analysis of Variance. A total of 164 oocytes from 36 cycles were assigned to earth treatment and ICSI was performed(88 head-first, tail-first). The rates of normal fertilization were 81.8% and 76.3% for head-first and tail-first, respectively. Of the fertilized oocytes, the percentage of cleaved embryos and the percentage of grade 1 embryo among cleaved embryos were 88.9% and 68.8%, 93.1% and 74.1% for head-first and tail-first, respectively. Of the 2-8 cell embryos cultured, 44.4%(16/36) and 50.0%(10/20) for head first and tail first, respectively developed to blastocyst stage. There were no differences in fertilization, cleavage, rates of grade 1 embryos, and development to blastocyst stage. In conclusion, head-first or tail-first sperm injection into an oocyte in ICSI program does not affect fertilization and subsequent embryo development to blastocyst stage in vitro.

From the panax ginseng chloroplast, the psbE and psbF genes, encoding the α- and β-subunits of cytochrome b-559 of the photosystem Ⅱ reaction center, respectively, were cloned and characterized. The psbE and psbF genes were composed of 252 and 117 nucleotides, respectively. The deduced amino acid sequence of the α-subunit showed 95%, 93%, and 91% homology to monocots, dicots, and liverwort, respectively, whereas the β-subunit showed approximately 98% to 95% homology to the same species. Southern blot analysis revealed that a single copy of the psbEF gene exists in the chloroplast plastid. Northern blot analysis indicated that the psbE and psbF genes are cotranscribed as a polycistron.

The cDNA clone encoding the antibacterial peptide (SL-1) was isolated from the fat body of the common cutworm, Spodoptera litura, immunized with E. coli K12. The primary structure analysis revealed that its deduced amino acid sequence showed the characteristics of the cecropin family antibacterial peptides and that the amino acid residues highly conserved in the antibacterial peptides from moths and flies were also conserved, implying that SL-1 was a cecropin-like, and especially cecropin B-like, peptide. The predicted secondary structure of the mature SL-1 consists of three domain: (i) an amphiphilic α-helical domain (Ile-4 to Gly-18); (ii) the hinge region (Gly-23 and Pro-24); and (iii) a hydrophobic domain (Ala-25 to Ile-38).

E. coli K12로 면역시킨 배추흰나비 유충의 지방체로부터 항균 펩티드와 유사한 단백질을 암호하는 3종류의 cDNA를 부분적으로 클로닝하였다. 이들 유전자의 염기서열은 서로 매우 유사하였으며 일차구조 분석 결과 연역된 아미노산 서열은 cecropin류의 항균단백질의 특성을 나타내었다. 그리고, 나방류와 파리류에서 밝혀진 항균단백질에서 잘 보존된 모든 아미노산이 새로이 클로닝된 단백질들에서도 모두 잘 보존되어 있음이 확인되었다. Three cDNA clones encoding the partial sequence of the antimicrobial factor-like peptide were isolated from the fat body of the cabbageworm, Artogeia rapae immunized with E. coli K12. Nucleotide sequences of three cloned genes have a high degree of homology one another. The primary structure analysis revealed that their deduced amino acid sequences showed the characteristics of the cecropin family antibacterial peptides and that the amino acid residues which are highly conserved in the antibacterial peptides from moths and flies are also conserved in butterfly, implying that these clones are encoding the antibacterial peptides.
