Proceedings of international Symposium on East-West Medicine, Seoul. 44-64, 1999.-Cordyceps sinensis is a major parasitic fungus on larva of Lepidoptera. It is one of the well know fungi used in traditional Chinese medicine for treatment asthma, lung and kideney diseases. Many fungi belonging to the genus Cordyceps have been demonstrated to produce natural products with various biological activities. Reported biological activities of genus cordyceps include:(a)inhibition of DNA and RNA synthesis;(b)enhancement of cell differentiation;(c)restruction of cytoskeleton;(d)inhibition of protein kinase activity;(e)antitumor activity on bladder, colon, lung carcinoma as well as fibroblastoma;(f)inhibition of the infection and revers transciptase activity of human immunodeficiency virus type 1; and(g) inhibition of methylation of nucleic acid. Thus, it is suggested that the extracts of Cordyceps sp. Contain antitumor, antiviral, and many other biological activities. In our previous studies, we proved that C. sinensis contains the antitumor and immunomodulatory agents. In the present studies, effects of C. sinensis on bronchoalveolar lavage fluids(BAL)cells were demonstrated. The crude methanolic extracts of C. sinensis were fractionated into 15 fractions by silica gel column chromatography. Effects of various fraction on BAL cells proliferation and lipopolysaccharide(LPS) stimulated interleukin-1β(IL-8),platelet activating factor(PAF), interleukin-6(IL-6), tumor necrosis factor-α(TNF-α),and interleukin-8(IL-8) production on BAL cells were determined. The result indicated that the CS-19-22 fraction suppressed BAL cells proliferation activated by LPS. The median inhibitory concentration(IC50) was 6㎍/ml. The CS-19-22 fraction also decreased IL-1β,PAF,IL-6, TNF-αand IL-8 production.The results of revers transcription-polymerase chain reaction(RT-PCR) demonstrated that the CS-19-22 fraction did not affect IL-1β,PAF,IL-6, TNF-α,and IL-8 mRNAs expression in BAL cells activated by LPS. By contrast, it inhibited the IL-10 mRNA expression but enhanced IFN-γand IL-12 mRNAs expression in activated BAL cells. Moreover, the CS-19-22 fraction blocked PAF-induced platelet aggregation. It is unlikely that cytotoxicity was involved. Because no cell deths were observable. We hypothesize that inhibitory mechanisms of CS-19-22 on BAL cells proliferation may be related to the impairments of gene expression and production of cytokines in BAL cells. The explanation of C. sinensis antiasthma function may involved the following(1)C.sinenesis contained immunomodulatory agents that stimulated IFN-γand IL-12 expression in TH1cells.(2)Enhanced secretion of IFN-γand IL-12 will inhibit the Tγcells immune responses Ex. Decreasing of IL-6 and IL-10 production.(3) The impairments of cytokines production in TH2 cells suppressed th BAL cells proliferation then more cytokines production Ex.IL-1β,PAF,IL-6,TNF-αand IL-8 in activated BAL cells were decreased. (4)The suppression of TH2cells immune responses will inhibit the growth and differentiation of B cells then IgE production will be decreased. (5)Reduced production of IgE will decreased asthma attack occurring in individuals. However, the results of the present study suggested that C. sinensis may also have acted to treat asthma in part by inhibiting BAL cells proliferation and cytokine gene expression and production. Future, experiments with treatment of animals with asthma with C. sinensis will be necessary to defined whether C. sinensis can reduce asthma injury. Plans are underway for the isolation of pure principal growth inhibitors and their mechanisms of action are subjected for further study.

        The Effect of Agomelatine in Behavioral and Psychological Symptoms of Dementia

        Objective: Moderate and severe behavioral and psychological symptoms of dementia (BPSD) often need medical treatment to improve symptoms. Agomelatine is a selective melatonergic (MT1/MT2) agonist that has normalizing effects on disturbed circadian rhythms and disrupted sleep−wake cycles. Its activity of 5HT-2C receptor antagonism is associated with lessening depression and anxiety and increasing slow-wave sleep. Based on past clinical records and current findings it suggests that agomelatine can improve BPSD for patients. This retrospective cohort study was designed to compare the BPSD before and after using agomelatine. Methods: Records of dementia cases who had ever received agomelatine treatment for BPSD in a general hospital setting during the past 2.5 years were identified and reviewed. Scores from before and after 3 months of treatment with agomelatine were collected for Neuropsychiatric Inventory (NPI), Brief Psychiatric Rating Scale (BPRS), and Clinical Global Impression (CGI) to compare and analyze the difference of psychological and behavioral symptoms pre- and post-agomelatine used. Results: Records of 144 cases of dementia with BPSD who had ever used agomelatine from January 2015 to June 2017 were collected. All of the 112 cases had BPRS and CGI scores, of which 75 cases had additional NPI scores. Among these 112 cases, the BPRS and CGI scores were significantly improved in all types of dementia. NPI scores indicated that the use of agomelatine alleviated obvious symptoms and decreased overall distress, especially in the depression/poor mood, anxiety, and sleep/night behavior. Conclusion: It is consistent with an effective result of agomelatine in improving BPSD.

        <B>Abstract</B><P>We present a measurement of the anisotropic and isotropic Baryon Acoustic Oscillations (BAO) from the extended Baryon Oscillation Spectroscopic Survey Data Release 14 quasar sample with optimal redshift weights. Applying the redshift weights improves the constraint on the BAO dilation parameter α(zeff) by 17 per cent. We reconstruct the evolution history of the BAO distance indicators in the redshift range of 0.8 &lt; z &lt; 2.2. This paper is part of a set that analyses the eBOSS DR14 quasar sample.</P>

        This paper presents our new intelligent interactive robot, which is constructed to eagerly provide multi-functional services in an office environment. In order to endow a full interactive capability of our robots for realizing so-called human-robot interaction (HRI), we propose sensor fusion based human detection and tracking system and human pose estimation to deal with a number of situations which may take place in the office environment. Not only by these perceptions, human interact with the robot also by some natural way, such as touching the interface screen and talking with the robot through microphone. Finally, the effectiveness of the proposed work is tested and validated by some of experiments.

        Analysis of Factors Influencing the Willingness of Taiwanese Students to Work on Mainland China: An Example of “31 Preferential Policies for Taiwan”

        In the Xi Jinping era, a key focus of the Communist Party of China (CPC), in terms of exchanges with Taiwan, is to attract young Taiwanese people to mainland China, and various policies exist to encourage their willingness to integrate. The “31 Preferential Policies for Taiwan” (“31 Measures”) is an influential guiding policy for this focus. Given that the negative impact of China's “sharp power” on liberal democracy has become an international point of focus, issues such as the impact of these policies on Taiwanese youths' willingness to seek employment (or entrepreneurship) opportunities on mainland China, and factors influencing this willingness, have become major concerns for supporters of democracy globally. In order to address these questions, this study proposes four hypotheses based on theories of rational choice and political socialization. An analysis of surveys of Taiwanese students found that the mean value of students' willingness to work on mainland China was 3.22, indicating that the overall willingness of students had shifted from “neutral” towards “willing.” Students perceived favor‐granting benefits and comparative benefits significantly enhanced their willingness to work on mainland China, while the life risks involved in doing so significantly reduced such willingness.

        Cognitive Dissonance and Changes in External Political Efficacy: A Panel Study of Chinese Students Studying in Taiwan

        Does studying in a democratic country elicit cognitive dissonance in students from communist countries and lead to changes in their political attitudes? This study recruited subjects from among Chinese students studying in Taiwan and adopted a panel study to explore the changes in the external political efficacy of these students. On the basis of cognitive dissonance theory, rational choice, social identity, and political socialization theories were combined to create a theoretical model. The results showed that the mean external political efficacy of all students was significantly lowered by 0.18 units on a scale of 1 to 4. The proposed regression model explained 26.87 percent of the variance in the degree of changes in external political efficacy. Four hypotheses were empirically supported.

      • Synthesis of Nesseria meningitidis W135 capsular oligosaccharide with various lengths for immunogenicity comparison

        Nesseria meningitidis is a type of bacteria which causes meningococcal meningitis and meningococcal septicemia. It often infects children under two years old and causes infant dead. Meningococcal diseases are prevalent in developing countries, like China and Africa. Based on their surface capsular polysaccharides, they are classified into 13 serogroups, while group A, B, C, Y, W135 are the major epidemic strains in the world. Though, some efforts have devoted to develop polysaccharide vaccines against N. meningitidis, but these vaccines often elicit weak immune responses and short immune memories. Therefore, polysaccharide-protein conjugate vaccines were developed to tackle the problem and the immunogenicity improved greatly. However, the compositions of commercial available vaccines are very heterogeneous and the quality is hard to control. In this study, we synthesized the N. meningitidis W135 capsular oligosaccharide in different lengths and acquired the homogeneous form to detect the immunogenicity. Because the N. meningitidis W135 capsular oligosaccharide is silalic acid-galactose repeating structure, we used sialyl phosphate as a donor to conjugate thiol galactoside to form the basic repeat unit. By linking the repeat units, we have obtained various length of oligosaccharide from disaccharide to decasaccharide. We conjugated these different length of oligosaccharides to carrier protein CRM197 (a mutated diphtheria toxin) and injected these conjugates to mice to search for the best vaccine composition.

      • The Different Expression of Gene Profiles on Hepatocellular Carcinoma Cells with Different Intracellular Hepatitis C Viral Load

        Aims: The different hepatitis C virus (HCV) replication has been reported among individual hepatocytes in chronic HCV infection by identifying hepatocytes with different HCV RNA levels. We have previously established a fluorescence-activated cell sorting (FACS) protocol to study the effects of different intracellular viral loads in HCV-infected cells. The present study aimed to further study the gene expression on different hepatocellular carcinoma (HCC) cells with different HCV viral load. Methods: The JFH1-EYFP viral florescence intensity was used to sort the high and low viral load cells after 5 days infection in vitro which has been shown in our previous study that infected cells efficiently and accurately discriminated between high- and low-viral load cell populations. The next generation sequence-RNA sequence was used to clarify the mRNA and miRNA gene network between HCV-high and HCV-low infected cells of the HCC cell line. Venn diagram summarizing the probe sets that were differentially expressingbetween the Huh7.5.1 versus each differential viral load cell population and miRDB and miRTar databases were used to predict HVL and LVL/S2 unique miRNA target genes. Results: By analyzing the NGS dataset and miRNA microarray dataset, of the significant transcripts, three miRNA were unique for the LVL/S2 cells and nine miRNA unique for the HVL. Twenty-three miRNA were common for all 3 viral load groups. We verified them by q-PCR and data confirmed the array data expression level. We found that high viral loads were associated with cell inflammation- and cell death-associated pathway; and the low viral loads were associated many stress response- and cell adhesion molecular (CAMs)-related genes. Conclusions: With the established cell sorting protocol, we have demonstrated that different gene network between HCV-high and HCV-low infected cells in JFH1-EYFP infectious cells exists. Our results may provide a boarder gene regulation map between high and low viral load cell populations.
