본 연구는 치아우식증에 관련된 Mutans streptococci 표준균주 및 임상분리 균주의 성장억제를 유도할 수 있는 천연 생약추출물을 검색하기 위하여 실시되었다. 녹차추출물은 CHMC-2032를 사용하였으며, 결명자 추출물은 50% 에탄올을 이용하여 얻었다. 이들 추출물의 Mutans streptococci 표준균주 및 교정환자들에서 브라켓과 치면 사이의 경계부에서 분리된 각각 10 균주씩의 S. mutans 및 S. sobrinus에 대한 최소성장억제농도를 액체배지 희석법으로 구하였다. 그 결과 CHMC-2032의 S. mutans 및 S. sobrinus의 표준 균주, S. sobrinus의 대부분 임상분리 균주에 대한 최소성장억제농도는 5 mg/ml이었다. 그러나 결명자 추출물에 의한 S. mutans 및 S. sobrinus에 대한 세균 성장억제 효과는 미미하였다. 본 연구 결과 치아우식증의 예방적 측면에서 결명자차보다는 녹차를 마시는 것이 유리하며, CHMC-2032를 이용하여 구강양치용액을 제조하여 사용할 경우 교정환자를 포함하여 대부분의 사람에게서 치아우식증 예방 효과가 있으리고 추정된다. Mutans streptococci is the major causative factor in dental caries. Especially, orthodontic patients with fixed appliance are a risk group for dental caries. Because fixed appliances attached on teeth may change the environment of dental plaque, the enamel decalcification or dental caries around the bracket and band is a major side effect of orthodontic treatment. The aim of this study was to search plant extracts that have antimicrobial effect on mutans streptococci. Seed-extract of Casio tora were prepared with ethanol and CHMC-2032, the leaf-extracts from Camellia sinensis extract, was obtained extract, 2 type strains and 20 clinical isolates of mutans streptococci isolated from the interface between orthodontic brackets and tooth surfaces in the orthodontic patients were used in this study. The minimal inhibitory concentration of CHMC-2032 was 5 mg/ml on the S. mutans KCTC 3065, S. sobrinus KCTC 3088, and 8 clinical isolates of S. sobrinus. However, there was no antibacterial effect of seed-extract of C. tora on mutans streptococci. These data suggest that green tea may be more effective than the tea prepared from C. tora in the prevention of enamel decalcification or dental caries around brackets.

The purpose of this study is to confirm the influences of learner-centered instruction on learners’ achievement and reason ability. In order to accomplish them, the fraction unit and the polygonal unit in the fourth grade were implemented with teaching methods and materials suitable for learner-centered mathematics instruction. Some conclusions could be drawn from the results as follows: First, learner-centered mathematics instruction has a more positive effect on learning of learned knowledge and generating unlearned knowledge in the experimental period than teacher-centered instructions. Second, learner-centered instruction makes an influence of low learning ability on getting achievement positively. Third, as the experimental treatment is repeated, learner-centered instruction has a positive effect on students’ reasoning ability. The reasoning ability of students showed a difference in the comparison between the experimental group and the comparative group, and within the experimental group, there was a positive effect of the extension of the positive reasoning ability. Fourth, it can be estimated that the development of students' reasoning ability interchangeably affected their generation test results.

본 연구는 전국의 646세대 가정주부를 대상으로 김장김치 제조시 사용하는 부재료 및 젓갈의 종류, 계절별 가정에서 주로 애용하는 김치류, 일일김치 소비량 등을 조사함으로써 지역적으로 다양한 겨울철 김장김치 맛에 영향을 줄 수 있는 요인을 분석하여 지역적 특성을 살린 시판김치의 산업화를 위한 기초자료로 이용되고자 실시하였다. 김장김치 담금시 고춧가루, 마늘, 생강, 파, 깨, 설탕, 풀(죽) 등 7종의 부재료의 사용빈도가 지배적이었으며, 굴, 새우 등의 해산물은 충청북도를 제외한 모든 지역에서 사용빈도가 높았다. 젓갈류에서 새우젓이 전국적으로 높은 사용빈도를 보였고, 멸치젓과 멸치액젓이 그 다음으로 많이 이용되는 것으로 나타났다. 계절별 애용되는 김치로는 봄철에는 파김치, 부추김치, 여름에는 열무, 오이소박이, 가을에는 총각김치와 가을배추겉절이, 겨울철에는 동치미가 있었으며, 남부지역은 파, 부추김치 등의 자극성이 강한 김치류가, 중부지역은 자극성이 적고 담백한 나박김치가 선호되었다. 대부분의 가정에서 김장김치를 직접 담그고 있었고, 봄에는 파김치와 무말랭이를, 여름에는 오이소박이와 깻잎김치, 가을에는 고구마줄기, 겨울에는 고들빼기와 같이 한번씩 입맛을 돋울 수 있는 김치를 사먹는 것으로 나타났다. 20~30대에서는 6~10포기의 김치를 담는 경우가 33.3~35.8%로 가장 많았고, 40~50대는 16~20포기, 50대 이상에서는 40포기 이상을 담는 경우가 많았다. 그리고 가정에서 1일 김치소비량은 1/8~1/4포기가 가장 많았고(42.7%), 개인당 1일 소비량으로 환산하면 80 g미만이 33.1%로 가장 많았다. This study was attempted to investigate preference characteristics affecting for winter kimchi. Questionnaire were collected by 646 nationwide households during 3 months from December 2000 in Korea, and the data were analyzed by SPSS program. The results were as follows: (1) Seven minor ingredients including red pepper, garlic, ginger, green onion, paste (e.g. glutinous rice, rice and/or wheat flour paste), sugar and sesame were used in high preference for making winter kimchi in that odor. Seafoods (oyster, shrimp, Alaska pollack, yellow corvenia, squid and hairtail) were also utilized for making winter kimchi related with regional characteristics. (2) Two jeotkals, anchovy and anchovy juice, were popular ingredients for winter kimchi in southern region, while shrimp jeotkal in middle region including Seoul, Chungnam, Chungbuk, Daejeon, Daegu, Gwangju and Jeju, northern sand lance juice jeotkal in western region, respectively. (3) Two kimchi, green onion and leek kimchi were high favored by people in spring regardless of region, and baby radish kimchi and cucumber kimchi in summer, pony-tail kimchi and gutjuli (fresh kimchi) in autumn, respectively. Except for winter cabbage kimchi, however, dongchimi and godulbaegi (Korean lettuce kimchi) were high favorite kimchi in winter. It was estimated that Koreans consume about 80 g of kimchi per person per day.

The anti-fingerprint coating agents, consist of a fluorinated macromer(F-macromer) were prepared by marcromonomer technique and hydrosilation reaction. The products were characterized by ¹H-NMR, differential scanning calorimetry. The surface properties of the anti-fingerprint coating agents were investigated by the contact and sliding angle measurement. The modified surface with the anti-fingerprint coating agents showed the extremely low surface free energy of a 10.09 dyn/cm and the sliding angle was smaller than that of commercial products. That is, The synthesised coating agents can be the good candidate for the water and oil repellent, soil release and anti -fingerprint application coating agent.
<i>Marinoscillum</i> <i>luteum</i> sp. nov., isolated from marine sediment

<P>A novel strain, designated SJP7<SUP>T</SUP>, was isolated from sediment of the Tofua Arc of the Tonga Trench. The 16S rRNA gene sequence of the isolate showed the highest similarity to that of <I>Marinoscillum pacificum</I> KCCM 42325<SUP>T</SUP> (95.9 %). Phylogenetic analysis revealed that the isolate formed a distinct phyletic line with <I>Marinoscillum pacificum</I> KCCM 42325<SUP>T</SUP> and <I>Marinoscillum furvescens</I> LMG 13023<SUP>T</SUP> within the family <I>Cytophagaceae</I>. Cells of strain SJP7<SUP>T</SUP> were Gram-stain-negative and appeared as long rods that were motile by gliding. Growth was observed at 15–40 °C (optimum, 30 °C), at pH 5.5–9.0 (optimum, pH 7.5–8.0) and in the presence of 0.5–7.0 % (w/v) NaCl (optimum, 2.5–3 %). The major respiratory quinone was MK-7. The dominant fatty acids were summed feature 3 (comprising C<SUB>16 : 1</SUB>ω7<I>c</I> and/or C<SUB>16 : 1</SUB>ω6<I>c</I>), iso-C<SUB>15 : 0</SUB> and C<SUB>16 : 1</SUB>ω5<I>c</I>. The DNA G<I>+</I>C content was 43.5 mol%. These properties support the affiliation of strain SJP7<SUP>T</SUP> with the genus <I>Marinoscillum</I>. Further phenotypic differentiation of strain SJP7<SUP>T</SUP> from other species of the genus <I>Marinoscillum</I> was indicated by the results of physiological and biochemical tests. On the basis of evidence from our polyphasic taxonomic study, strain SJP7<SUP>T</SUP> represents a novel species of the genus <I>Marinoscillum</I>, for which the name <I>Marinoscillum</I> <I>luteum</I> sp. nov. is proposed. The type strain of <I>Marinoscillum</I> <I>luteum</I> is SJP7<SUP>T</SUP> ( = KCTC 23939<SUP>T</SUP> = NCAIM B02491<SUP>T</SUP>).</P>
Ultra-fast Generic LC-MS/MS Method for High-Throughput Quantification in Drug Discovery

An ultra-fast generic LC-MS/MS method was developed for high-throughput quantification of discovery pharmacokinetic (PK) samples and its reliability was verified. The method involves a simple protein precipitation for sample preparation and the analysis by ultra-fast generic LC-MS/MS with the ballistic gradient program and selected reaction monitoring (SRM) mode. Approximately 290 new chemical entities (NCEs) (over 10,000 samples) from 5 therapeutic programs were analyzed. The calibration curves showed good linearity in the concentration range of 1, 2 or 5 to 2000 ng/mL. No significant ion suppression was observed in the elution region of all the NCEs. When approximately 300 plasma samples were continuously analyzed, the peak area of internal standard was constant and reproducible. In the repeated analysis of samples, the plasma concentrations and the area under the curve (AUC) were consistent with the results from the first analysis. These results showed that the present ultra-fast generic LC-MS/MS method is reliable in terms of selectivity, sensitivity, and reproducibility and could be useful for high-throughput quantification and other bioanalysis in drug discovery.

After success of sildenafil for the treatment of erectile dysfunction, a large number of its analogues have been approved from FDA. Recently, the illegal dietary supplements which include sildenafil, vardenafil, tadalafil, or analogues of these drugs as ingredient have been widely distributed. Therefore, the determination of the residue of synthetic phosphodiesterase- 5 (PDE-5) inhibitors in dietary supplements is highly required due to indiscriminate and unintentional overdose caused nausea, chest pains, fainting and irregular heartbeat. In this paper, we report a rapid and sensitive analytical method for the simultaneous determination of nine phosphodiesterase-5 inhibitors by liquid chromatography-high resolution mass spectrometry. The present method was found to be accurate and reproducible with 40 μg/g of the limit of quantification for the nine PDE-5 inhibitors. The developed method can be successfully applied to the analysis of the seven illegal dietary supplements.
