      • The Research of Data Mining Based on Application Data Pool

        Today, people use various kinds of information technology applications to deal with applications in our daily life, which generates lots of information. However,most of the informationis just stored in many Distributed Heterogeneous Databases (DHDs) as log records, instead of being used abundantly and effectively. In this paper, we mainly discusses about how to use these data in useful ways by Data Mining (DM). Relative to thetraditional Data Mining based on Data Warehouse (WD),we propose a definition named Application Data Pool (ADP)replacing WD in this paper. And we design a KnowledgeDiscovery in Databases (KDD) model with ADP to use these data more efficiently. At last we use an application in Shanghai University ID Card designed with the ADP to prove the effectiveness and feasibility in KDD.

        Growth, reproduction and recruitment of Silvetia siliquosa (Fucales, Phaeophyceae) transplants using polyethylene rope and natural rock methods

        Silvetia siliquosa is an ecologically and commercially important brown alga that is harvested from its natural habitats,but its population has recently been diminishing along the Korean coast. To develop new techniques for algal populationrestoration, we tested two newly developed transplantation methods (using polyethylene ropes and natural rock pieces)at two study sites, Gwanmaedo and Yeongsando, on the southwest coast of Korea, from May to November 2014. Thetransplants on polyethylene ropes showed significantly greater survival, maturity, and growth than those on natural rocksat both study sites. Newly recruited juveniles (<3 cm) of S. siliquosa increased remarkably from May to December near thetransplants on polyethylene ropes and natural rocks.Therefore, we suggest that transplantation using polyethylene ropesis more effective than using natural rocks to restore the population of S. siliquosa in Korea.

        Non-isothermal Decomposition Kinetics of a New High-energy Organic Potassium Salt: K(DNDZ)

        A new high-energy organic potassium salt, 2-(dinitromethylene)-1,3-diazepentane potassium salt K(DNDZ), was synthesized by reacting of 2-(dinitromethylene)-1,3-diazepentane (DNDZ) and potassium hydroxide. The thermal behavior and non-isothermal decomposition kinetics of K(DNDZ) were studied with DSC, TG/DTG methods. The kinetic equation is $\frac{d{\alpha}}{dT}$ = $\frac{10^{13.92}}{\beta}$3(1 - $\alpha$[-ln(1 - $\alpha$)]$^{\frac{2}{3}}$ exp(-1.52 ${\times}\;10^5$ / RT). The critical temperature of thermal explosion of K(DNDZ) is $208.63\;{^{\circ}C}$. The specific heat capacity of K(DNDZ) was determined with a micro-DSC method, and the molar heat capacity is 224.63 J $mol^{-1}\;K^{-1}$ at 298.15 K. Adiabatic time-to-explosion of K(DNDZ) obtained is 157.96 s.

      • Luciferase Assay to Screen Tumour-specific Promoters in Lung Cancer

        Objective: Specific promoters could improve efficiency and ensure the safety of gene therapy. The aim of our study was to screen examples for lung cancer. Methods: The firefly luciferase gene was used as a reporter, and promoters based on serum markers of lung cancer were cloned. The activity and specificity of seven promoters, comprising CEACAM5 (carcinoembryonic antigen, CEA), GRP (Gastrin-Releasing Peptide), KRT19 (cytokeratin 19, KRT), SFTPB (surfactant protein B, SP-B), SERPINB3 (Squamous Cell Carcinoma Antigen, SCCA), SELP (Selectin P, Granule Membrane Protein 140kDa, Antigen CD62, GMP) and DKK1 (Dickkopf-1) promoters were compared in lung cancer cells to obtain cancer-specific examples with strong activity. Results: The CEACAM5, DKK1, GRP, SELP, KRT19, SERPINB3 and SFTPB promoters were cloned. Furthermore, we successfully constructed recombinant vector pGL-CEACAM5 (DKK1, GRP, SELP, KRT19, SERPINB3 and SFTPB) contained the target gene. After cells were transfectedwith recombinant plasmids, we found that the order of promoter activity from high to low was SERPINB3, DKK1, SFTPB, KRT19, CEACAM5, SELP and GRP and the order for promoters regarding specificity and high potential were SERPINB3, DKK1, SELP, SFTPB, CEACAM5, KRT19 and GRP. Conclusion: The approach adopted is feasible to screen for new tumour specific promoters with biomarkers. In addition, the screened lung-specific promoters might have potential for use in lung cancer targeted gene therapy research.

        Performance and Spatial Succession of a Full-Scale Anaerobic Plant Treating High-Concentration Cassava Bioethanol Wastewater

        A novel two-phase anaerobic treatment technology was developed to treat high-concentration organic cassava bioethanol wastewater. The start-up process and contribution of organics (COD, total nitrogen, and NH4+-N) removal in spatial succession of the whole process and spatial microbial diversity changing when sampling were analyzed. The results of the start-up phase showed that the organic loading rate could reach up to 10 kg COD/m3d, with the COD removal rate remaining over 90% after 25 days. The sample results indicated that the contribution of COD removal in the pre-anaerobic and anaerobic phases was 40% and 60%, respectively, with the highest efficiency of 98.5%; TN and NH4+-N had decreased to 0.05 g/l and 0.90 g/l, respectively, and the mineralization rate of total nitrogen was 94.8%, 76.56% of which was attributed to the anaerobic part. The microbial diversity changed remarkably among different sample points depending on the physiological characteristics of identified strains. Moraxellaceae, Planococcaceae, and Prevotellaceae were dominant in the pre-anaerobic phase and Bacteroidetes, Campylobacterales, Acinetobacter, Lactobacillus, Clostridium, and Bacillus for the anaerobic phase. Methanosarcinaceae and Methanosaeta were the two main phylotypes in the anaerobic reactor.

        Remarkable impact of steam temperature on ginsenosides transformation from fresh ginseng to red ginseng

        Background: Temperature is an essential condition in red ginseng processing. The pharmacological activities of red ginseng under different steam temperatures are significantly different. Methods: In this study, an ultrahigh-performance liquid chromatography quadrupole time-of-flight tandem mass spectrometry was developed to distinguish the red ginseng products that were steamed at high and low temperatures. Multivariate statistical analyses such as principal component analysis and supervised orthogonal partial least squared discrimination analysis were used to determine the influential components of the different samples. Results: The results showed that different steamed red ginseng samples can be identified, and the characteristic components were 20-gluco-ginsenoside Rf, ginsenoside Re, ginsenoside Rg1, and malonyl-ginsenoside Rb1 in red ginseng steamed at low temperature. Meanwhile, the characteristic components in red ginseng steamed at high temperature were 20R-ginsenoside Rs3 and ginsenoside Rs4. Polar ginsenosides were abundant in red ginseng steamed at low temperature, whereas higher levels of less polar ginsenosides were detected in red ginseng steamed at high temperature. Conclusion: This study makes the first time that differences between red ginseng steamed under different temperatures and their ginsenosides transformation have been observed systematically at the chemistry level. The results suggested that the identified chemical markers can be used to illustrate the transformation of ginsenosides in red ginseng processing.

        BeMicroarray Analysis of the Major Depressive Disorder mRNA Profile Data

        ObjectiveaaMajor depressive disorder (MDD) is a common mood disorder associated with several psychophysiological changes like disturbances of sleep, appetite, or sexual desire, and it affects the patients’ life seriously. We aimed to explore a genetic method to investigate the mechanism of MDD. MethodsaaThe mRNA expression profile (GSE53987) of MDD was downloaded from Gene Expression Omnibus database, including 105 samples of three brain regions in post-mortem tissue suffered from MDD and unaffected controls. Differentially expressed genes (DEGs) in MDD were identified using the Limma package in R. Gene Ontology functions and Kyoto Enrichment of Genes and Genomes pathways of the selected DEGs were enriched using Database for Annotation, Visualization and Integrated Discovery. Protein-protein interactive network of DEGs was constructed using the Cytoscape software. ResultsaaTotally, 241 DEGs in MDD-hip group, 218 DEGs in MDD-pfc group, and 327 DEGs in MDD-str group were identified. Also, different kinds of biological processes of DEGs in each group were enriched. Besides, glycan biosynthesis of DEGs in MDD-str group, RIG-I-like receptor signaling and pyrimidine metabolism of DEGs in the MDD-hip group were enriched, respectively. Moreover, several DEGs like PTK2, TDG and CETN2 in MDD-str group, DCT, AR and GNRHR in MDD-pfc group, and AKT1 and IRAK1 in MDD-hip group were selected from PPI network. ConclusionaaOur data suggests that the brain striatum tissue may be greatly affected by MDD, and DEGs like PTK2, GALNT2 and GALNT2 in striatum, AR in prefrontal cortex and IRAK1 and IL12A in hippocampus may provide novel therapeutic basis for MDD treatment.

        Parameter Optimization for the Thickness and Hydraulic Conductivity of Tunnel Lining and Grouting Rings

        Water inrush is a serious geological hazard in the deep buried tunnel construction in karst areas. Lining and grouting ring are often built to prevent tunnel from water inrush. The estimation of external water pressure and water inflow is of great significance to the structure design of lining and grouting ring. In this paper, the axisymmetric analytic solutions of external water pressure and water inflow were derived for deep karst tunnels under saturated steady-state flow conditions. A simplified formula for the reduction coefficient of external water pressure was established. This paper analyzed the changing rule of water inflow coefficient and the reduction coefficient of external hydraulic pressure on linings, which varies with the parameters of lining and grouting circle. The rational and available coefficients of grouting circle in karst deep buried tunnel construction are drawn: the grouting ring thickness is 6 m and the hydraulic conductivity ratio of surrounding rock and lining is 100. In addition, the accuracy of the simplified formula was verified by the comparison of analytical solution and numerical analysis. This study provides a reliable calculation method for water inflow and lining water pressure of the deep buried tunnel and have certain reference value for the structural design of deep buried tunnel to reduce water inflow and ensure construction safety.
