혈청이 마우스 간 세포주 BNL CL.2의 Nitric Oxide 생성에 미치는 영향

마우스 간 세포주인 BNL CL.2의 시험관내 배양에서 혈청과 IFN-γ가 세포주의 nitric oxide(NO) 생성과 세포 손상에 미치는 영향을 알아보기 위한 실험을 하였다. 혈청이 공급된 배양에서 IFN-γ에 의한 세포 생존율은 거의 변동이 없었으나, 혈청을 제거한 배양에서는 약 65%의 생존율이 유지되었으며, NO 생성 억제제인 N -monomethy-L-arginine (NMA)의 첨가는 농도 의존적으로 세포의 생존율을 감소시켰다. 혈청이 제거된 BNL CL.2세포주는 IFN-γ단독 처리에서도 NO 를 생성할 수 있었으며, IFN-γ와 lipopolysaccharide (LPS)의 복합 처리는 세포주의 NO 생성을 상승적으로 증가시켰다. 또한 protein tyrosine kinase (PTK) inhibitor인 herbimycin A와 genistein에 의해서 NO 생성이 억제되어 PTK의 활성이 혈청이 고갈된 BNL CL.2 세포에서 NO의 생성에 중요한 역할을 담당하고 있기 때문으로 판단된다. IFN-γ의 독성은 혈청을 제거시킬 때 NO 생성 억제제에 상승적으로 간 세포를 손상시키며, 이때 NO가 IFN-γ에 의해 유도된 손상을 어느 정도 억제시키는 것을 알 수 있었다. Nitric oxide (NO) plays an important role in immunologic defense, and influences upon the functioning of secretory tissues and cells. It also exhibits cytotoxic/cytostatic activity as one of major operating effectors of the cellular immunity system. We investigated the effects of serum on the cell damages and NO production in the mouse liver cell line BNL CL.2 to establish the role of NO. We observed that, when BNL CL.2 cells were cultured in serum-free medium, they were induced to cell damage by the stimulation of IFN-γalone or IFN-γplus LPS. Serumstarved cells showed large amount of nitrite accumulation and NO synthase (NOS) expression in response to IFN-γalone in dose-and time-dependent manners, but serum-supplied cells did not. The production of NO was blocked by protein tyrosine kinase (PTK) inhibitors, genistein and herbimycin. These results suggest that the deprivation of serum in the BNL CL.2 cell culture medium might primed with the cells to produce NO when the cells are triggered by IFN-γ and the involvement of PTK signal transduction pathway in the expression of NOS gene in murine hepatocytes.

A numerical study is conducted for the secondary side screw-type tube inlet orifice design of a once-through steam generator. Various tube plugging conditions and power levels are considered, and the secondary coolant inlet temperature is adjusted to maintain a constant thermal power. Comprehensive numerical solutions are acquired to evaluate the minimum orifice length under various operating conditions, and the required minimum orifice length to suppress the flow oscillation below the allowable level is evaluated. The results obtained show that the lowest power level results in the highest minimum orifice length and non-plugging condition provides a limiting case for the orifice length criterion.
Papermaking Characteristics of Kudzu Root Fibers with Different Morphology

Kudzu kills and damages neighboring plants by smothering or encircling them. Kudzu was divided into male and female in appearance. At the utilizing standpoint of kudzu, the leftovers from the roots extraction were pulped by two different pulping methods. Soda pulping was individually applied for male and female root leftovers, and kraft pulping was done for their mixed leftovers. Soda pulps made from the separate male and female root leftovers did not show any big difference in the contents of both extractives and lignin before bleaching. However, after bleaching, the decrease in extractives and lignin of each pulp occurred. Kraft pulping made under strong alkaline treatment contributed to the higher reduction in the contents of extractives and lignin even after bleaching. Starch content harmful in white water during papermaking was higher in soda pulp than in kraft pulp. Even though 10% of kudzu pulps was mixed in a papermaking stock, most of properties of the sheets containing bleached soda pulp and kraft pulp were exceedingly weaker than those containing bleached SW-BKP. In conclusion, irrespective of pulp types, kudzu root leftovers could be a minor resource to produce paper with special functionality when weaker fading resistance and lower strength were considered.

Background: Atopic dermatitis is a chronic recurrent inflammatory skin disease that often that affects up to 20% of children. The pathogenesis of atopic dermatitis is not fully understood, but newly identified immune cells such as ILC2s have been suggested to play an important role. Food allergy is a known provoking cause of AD, but the exact relationship between oral tolerance to food antigen and development of AD is still unknown. Objectives: To show 1) oral tolerance can hindered the development of AD-like skin lesions in mouse model by epicutaneous sensitization with OVA. 2) ILC2s play acrucial role in oral tolerance-induced epicutaneous sensitization mouse model. Methods: BALB/C female mice were sensitized with OVA by epicutaneous application. Experimental group was feed with OVA by oral route before treatment. We analyzed changes of antibodies, cells and cytokines in MLN andskin of each group. Results: Epicutaneous sensitization increased level of anti-OVA IgE and IgG1, while oral tolerance decreasedlevel of anti-OVA IgE and IgG1. In oral tolerance group, increased DCs, iTregs, TGF-モ in MNL and decreased IL-5, IL-13, Ccr3 in skin were shown. Also, ILC2s were decreased in oral tolerance group. Conclusion: 1. Oral tolerance induced by OVA hindered the development of AD-like skin lesions in epicutaneous sensitized mice with OVA. 2. MLN-induced Tregs plays main role in induction of oral tolerance. 3. When AD-like skin lesions occur after oral tolerance, ILC2s are increased. They are con
