Interval-censored observations are common in medical and epidemiologic studies; however, limited studies exist due to the complexity and special structure of interval-censoring. This paper introduces the imputation method and the self consistency method in the interval-censored data. We propose a new method of generating random numbers under an interval-censoring set-up. Through simulation studies we compare two methods under various simulation schemes in the sense of the mean squared error for estimating the median survival time and the mean integrated squared error for estimating the survival function. Under a moderate censoring percentage, the mean imputation method showed a better performance than the self-consistency method in estimating the median survival time and the survival function.

Al₂O₃ thin films were deposited on the moth eye anti-reflective nanostructured polycarbonate films by atomic layer deposition (ALD) techniques. Without ALD-Al₂O₃ thin films, moth eye anti-reflective nanostructured films had a high optical transmittance of 95.47% at a wavelength of 550 nm and a very poor hardness of 0.1381 GPa. With increasing the thickness of Al₂O₃ thin films from 5 to 25 nm, the transmittance of moth eye anti-reflective nanostructured films was gradually decreased from 94.94 to 93.12% . On the other hand, the hardness of the films was greatly increased from 0.3498 to 0.7806 GPa with increasing the thickness of Al₂O₃ thin films. This result shows that ALD thin films can be applied to improve mechanical properties with an adequate optical transmittance of the conventional moth eye anti-reflection nanostructure films.

본 연구는 곤충 유전자를 이용한 항세균성 펩타이드 생산 및 농업용 소재로서의 응용에 관한 연구로서 항세균성 단백질 누에신 유전자를 베큘로 바이러스 발현계(BEVS)를 이용하여 곤충세포주에서 발현한 후 누에신 단백질의 농업용 소재로서의 가능성을 모색하기 위해 농작물을 가해하는 감자 고추의 무름병을 일으키는 Pectobacteriun carotovorum subsp. carotovorum, 가지 및 고추의 풋마름병을 일으키는 Ralstonia solanacearum, 양송이 버섯의 세균성 갈색 무늬병을 일으키는 Pseudomonas tolaasii 및 무와 배추의 검은썩음병을 일으키는 Xanthomonas campestries pv. campestris에 대해서 항세균 활성을 관찰하였다. 그 결과 Pectobacterium carotovorum subsp. carotovorum, Ralstonia solanacearum 및 Pseudomonas tolaasii에 대해 높은 활성을 나타태었으며 Xanthomonas campestris pv. campestris에는 활성을 나타내지 않았다. Ion exchange 및 gel filtration chromatography를 수행하여 약 20 kDa의 성숙 누에신 단백질을 순수 분리하여 pH 및 온도에 대한 안정성을 조사한 결과, pH 2∼12 완충액에서 30분간 처리하였을 때에도 항세균 활성이 그대로 유지되었고 100℃에서 2시간 처리시에는 활성이 안정되었으며 4시간 처리시에도 80% 정도로 유지됨을 확인함으로써 누에신 단백질은 pH 및 온도에 대한 안정성이 있음을 확인할 수 있었다. The antibacterial protein gene, nuecin was expressed in Sf9 cells using baculovirus expression vector system (BEVS). The antibactetial activity of mature nuecin against Pectobacterium carotovorum subsp. carotovorum,k Ralstonia solanacearum and Pseudomonas tolaasii was significantly high,k demonstrating that nuecin had a wider antibacterial spectrum on gram vegabtive and positive bacteria. The result appears to be superior to other antibacterial peptide, attacin. The nuecin was purfied by SP- sepharose and Mono Q HR ion-exchange chromatography, and then by Superdex 200 HR 10/30 column. The purified nuecin is quite stable at 80°C and 100°C for several hours of incubation and in a wide pH range (pH 2-12).

Primary hepatic neuroendocrine carcinoma (PHNEC) is rare and its origin is not clearly understood. The coexistence of PHNEC and hepaotcellular carcinoma has been reported in only a few cases. We report a rare case of combined PHNEC and hepatocellular carcinoma in a patient with liver cirrhosis caused by chronic hepatitis B that resulted in aggressive behavior and poor prognosis. The purpose of this study is to understand and examine child care center teachers` awareness and management of free play activity, and to provide fundamental data for more effective future management of free play activity. For this study, 240 surveys were distributed to teachers in child care centers to explore their awareness and management of free play activity. The results of this study are as follows: First, most child care center teachers were positively aware of the purpose of free play activity as introducing specific concepts to children. There were differences in teachers` overall awareness of the purpose of free play activity by a type of center, teaching experiences, and ages, while there was no difference by teachers` educational background. Most teachers were positively aware of their role in free play activity as solving a problem through intervention in fights between children. There were differences in teachers` overall awareness of their role in free play activity by a type of center and educational background, while there was no difference by teaching experiences and ages. Second, most teachers were highly aware of a problem in free play activity as not knowing how to guide children properly in various problem situations. Third, schedule of free play activity for most child care center teachers was 50~60 minutes and twice a day. Most teachers make an arrangement of free play activity alone, and they don`t choose an area of free play activity in an established way. Most teachers allow children to choose the next activity for themselves in general. Teachers were aware that the most active area in free play activity is the black area because children prefer to the black area itself. On the other hand, teachers were aware that the most inactive area in free play activity is the number/science area because children dislike this area itself. Most teachers set up free play activity according to section, season, and term. And they limit the number of children when a certain area is crowded. After free play activity, most teachers evaluate the activity.
누에 배양세포(Bm5) 및 생체에서 베큘로바이러스 발현계를 이용한 누에신 단백질 발현 특성

본 연구는 누에신 단백질의 대량발현을 통해 농업용 소재로서 이용하고자 하는 연구의 일환으로 누에 핵다각체병 바이러스 유래의 pBm10pol-Xa 벡터에 누에신 유전자를 도입하여 누에 배양세표(Bm5) 및 누에 생체에서 발현한 결과 누에신전사체는 바이러스 접종 후 1일째부터 발현되기 시작하여 5일째에 최대로 발현되었음을 확인할 수 있었고, 누에신 단백질은 3일째부터 발현량이 증가하여 5일째까지 계속 지속적으로 발현되었으나 그 발현량은 전사체에서처럼 많지 않음을 확인할 수 있었으며, 누에신 단백질의 발현량은 누에 세포에 비해 누에 생체에서 기대만큼 그 발현량이 많지 않았다. 또한 누에신의 베큘로바이러스 발현계(baculovirus expression vector system, BEVS)를 이용하여 세포내 및 번역후 변형과정을 통하여 세포외로 분비된 누에신의 발현양상을 확인한 결과 세포내에 비해 세포외로 분비시 그 발현량이 현저히 줄어들었음을 확인할 수 있었다. 따라서 베큘로바이러스 발현계를 이용하여 외래 단백질을 생산할 경우 정확한 메카니즘은 밝혀지지 않았으나 강력한 프로모터에 의해 세포내 단백질 생산량은 많은데 비해 정확한 단백질 고차구조 형성을 도와주는 foldase 및 chaperon의 양은 한정되어져 있으므로 정확한 고차구조를 형성하여 세포외로 분비되는 생물학적 활성을 띠는 단백질은 매우 적은 양만이 발현됨을 확인할 수 있었다. 그러므로 추후 누에신 단백질 대량생산을 위해서는 분비 프로세싱의 해명 및 번역 후 변형과정의 개선을 통한 발현계의 개량이 시급히 요구된다. For the practical use of nuecin protein, we tried to overexpress nuecin using Brn5 inserted nuecin cDNA into pBm10pol-Xa vector derived from B. mori nuclear polyhedrosis virus (BmNPV), and expressed in Bm5 cells and B. mori respctively. SDS-PAGE and Northern blot analysis showed an expressed of the protein when baculovirus expression vector system (BEVS) was used. The amount of intracellular protein is abundant, but the amount of extracellular protein is poor. The results suggest that the bilogically active nuecin protein produced by using BEVS is poor because incresed level of misfolded nuecin by the strong promoter, polyhedrin and p10 of BEVS, decrease the level of free chaperons and foldases by binding with them.
결핵균과 비결핵성항산균 검출에 Real-time PCR의 유용성

Background: The purpose of this study was to evaluate recently developed real-time polymerase chain reaction (PCR) assay kit to detect Mycobacterium tuberculosis (MTB) and nontuberculous mycobacteria (NTM) in respiratory specimens. Methods: We assessed the positive rate of the real-time PCR assay to detect MTB and NTM in 87 culture-positive specimens (37 sputum, 50 bronchial washing), which were performed real-time PCR by using Real-QTM MTB&NTM Kit from January 2009 to June 2009, at Gyeongsang University Hospital. To compare the efficacy with the TB-PCR assay, we evaluated 63 culture-positive specimens (19 sputum, 44 bronchial washing) for MTB or NTM, which were performed TB-PCR by using ABSOLUTETM MTB II PCR Kit from March 2008 to August 2008. Results: Among 87 specimens tested using real-time PCR, MTB and NTM were cultured in 58 and 29, respectively. The positive rate of real-time PCR assay to detect MTB was 71% (22/31) and 92.6% (25/27) in AFB stain-negative and stain-positive specimens. For NTM, the positive rate of real-time PCR was 11.1% (2/18) and 72.7% (8/11) in AFB stain-negative and stain-positive specimens. Among 63 specimens performed using TB-PCR, MTB and NTM were cultured in 46 and 17, respectively. The positive rate of TB-PCR was 61.7% (21/34) and 100% (12/12) in AFB stain-negative and stain-positive specimens. TB-PCR was negative in all NTM-cultured 17 specimens. Conclusion: TB/NTM real-time PCR assay is useful to differentiate MTB and NTM in AFB stain-positive respiratory specimens and it is as effective in detecting MTB with TB-PCR.

The reasons for selecting insects as future food are high growth and feed conversion rates, reproduce quickly, low environmental footprint, valuable source of nutrients, and source of a various undeveloped functional materials. Since 2014, Tenebrio molitor larva, Protaetia brevitarsis larva, Allomyrina dichotoma larva, and Gryllus bimaculatus were registered as new food in Korea because it has been scientifically proven that they are not harmful to eat and nutritious. Therefore they can be legally produced and sold as food. Accordingly, there are 7 species of edible insects including grasshopper, silkworm pupa, and Baekgangjam in Korea. To improve aversion to edible insects, using their powder, chop, and gravy hidden their morphology, we developed more than 100 different kinds of recipes for general food menu, held several times tasting events to be familiar with edible insect food, and published cookbooks. Moreover, we developed more than 50 kinds of recipes for patients. To eat the insect food more and more people, we have been analyzed various function of insects. If health food based on the results of functional analysis is developed, edible insect’s value will be raised. If various insect foods are developed and consumed by people, edible insect market will grow up more than 100 billion won in 2020.
