      • 참나리와 중나리의 종간교잡 화합성과 F₁ 후대의 생육 특성

        중국과 일본등지 에서 식용으로 널리 이용되고 있는 참나리는 경제적 가치가 높은 나리 속 식물 중의 하나이고, 중나리는 참나리의 근연종으로 형태적 특성이 참나리와 유사하나 주아가 착생되지 않는 특징을 가지고 있다. 오래전부터 식용, 약용, 화단용으로 이용되어 온 참나리와 중나리는 야생에서도 매우 강건하여 나리류의 육종을 위한 중요한 자원으로 활용되어왔다. 본 연구는 참나리와 중나리 간 종간 교잡 시 생육 및 생리학적 특성을 규명하고, 교잡조합에 대한 우량계통을 선발하여 식용나리 육성을 위한 육종 자료를 확보하고자 수행되었다. 2배체 참나리와 중나리의 정역 교잡에 참나리가 모본으로 사용되었을 경우 착과율이 높았고, 정상 종자 형성 율은 낮은 반면 이들 정상종자의 발아율은 높았다. 반면에 중나리를 모본으로 사용하였을 경우 착과 되지 않아 편 방향 화합성이 강하였다. 3배체와의 중나리 교배 조합에서는 정상 종자 형성 율은 높았지만 발아율은 극히 저조하였다. 참×중나리 종간 교잡 집단은 참나리나 중나리 모본보다 생육이 우수하였으며, F1 집단에서는 주아가 착생되지 않은 개체와, 주아가 착생된 개체가 가각 50%정도 발견되었다. 참나리 특성을 닮은 것은 주아가 착생되었고, 주아 착생되지 않는 계통은 중나리 특성을 많이 닮은 것으로 조사되었다. 종간 교잡화합성과 F1 후대의 생육 특성을 연구하기 위해 3년생 F1 종간 잡종을 주아 계통과 무 주아 계통으로 구분하여 개화기, 꽃의 형태, 초장, 경경, 엽장, 엽 폭, 구근 비대 등을 조사하였다. 또한, 개 화 후의 줄기의 잎을 상단, 중단, 하단별로 나누어 엽장과 엽 폭을 조사하였다. 주아 교배 조합은 상부에 있는 잎이 짧고 초장이 무 주아 교배 조합보다 작았다. 무주아 계통은 주아 계통보다 불화합성을 나타내는 것이 많았지만, 생육 속도 와 구근 비대정도가 우수 하였으므로, 식용나리 품종은 주아 계통보다 주아가 착생되지 않는 무 주아 계통으로 부터 육성하는 것이 바람직할 것으로 사료되었다. Tiger lily is the most popular lily in Korea and have been used as a garden plant and sometimes was grown for edible bulb. L. leichtlinii? var. maximowizii is almost same in morphology with Tiger lily except the absence of bulbil. These two lilies are broadly cultivated and used as an valuable economic plants in China and Japan but in Korea it is not cultivated for edible bulb and not bred. Lilium lancifolium has been often used as breeding parents for ornamental Asiatic lilies. It is a polyploid-complex species comprised of diploid and triploid, and the latter is common cytotype with relatively wide distribution in entire Japan, Korea, northeastern China, and southeastern Russia. This study was conducted to obtain fundamental data for breeding edible interspecific hybrids between materials Lilium lancifolium and L. leichtlinii? var. maximowizii and to understand the morphological charateristics of their progenies. Fruit set was 67% in crosses between diploid Lilium lancifolium × L. leichtlinii? var. maximowizii and 47% in diploid Lilium lancifolium × L. leichtlinii? var. maximowizii. Seed germination was 55% in former combinations and 0.3% in latter. F1 plants of interspecific crosses were more vigorous than their parent plants and separated into two groups of bulbil forming and non-bulbil forming plants. Their morphological characteristics of flowers, leaves, bulbs, plant heights, and flowering times were inspected.

      • IEC 기술표준 개정에 따른 가스폭발위험범위 계산 및 대응방안에 관한 연구

        The occupational safety and health act evaluates and explosion hazardous areas according to Korean Standard(KS C IEC 60079-10-1) and proper explosion-proof constructions with electric machines and apparatus have to be installed for flammable gases or liquids. Now, Current KS follows IEC 1st edition and there has been no change considering revisions since 2008, thus it is expected that KS will be updated based on IEC 2nd edition, revised in 2015. In this study, IEC 60079-10-1 1st Edition is compared with IEC 2nd edition. Total 112 case studies, four chemical compounds(methane, propane, benzene, methanol)are selected, are evaluated based on IEC 1st and 2nd edition and explosion hazardous ranges of them are analyzed according to various leakage pressures and hole sizes. In order to verify the results calculated by IEC, PHAST, which is one of the most representative consequence analysis programs, is employed to calculate the explosion hazardous ranges. As a result, it is concluded that there are many differences between IEC 1st and 2nd edition due to the discharge coeffcient and the ventilation parameters. In general, the explosion hazardous ranges evaluated by 1st edition are larger than them calculated by 2nd edition and PHAST. However, in the case of propane, the explosion hazardous ranges calculated according to the 2nd edition are larger if the leakage hole size is larger than 2.5㎟ and a leakage pressure is bigger than 0.1MPa. Therefore, it is worth to note that the large value between the explosion hazardous ranges value calculated by the IEC technical standard(IEC 60079-10-1) 1st Edition and 2nd Edition should be selected until KS is revised in future. Also, future studies for IEC 2nd should be performed to evaluate the accuracy.

      • 향사육군자탕이 식욕증진과 위장관 호르몬에 미치는 영향

        Background : Hyangsayukgunja-tang is a traditional Korean medicine that is used to treat various gastrointestinal tract disorders such as anorexia, functional dyspepsia and gastric ulcer. Our purpose is to clarify that Hyangsayukgunja-tang increases appetite and affects gut hormones . Gut hormones act peripherally to modulate digestion and absorption of nutrients. However they also act as neurotransmitters within the central nervous system to control food intake. Peptide YY(PYY) supress appetite, oppositely ghrelin increases appetite through afferent vagal fibers to the brain stem or directly to the hypothalamus. Methods : We administered high and low dose of Hyangsayukgunja-tang(High dose; 1g/kg Low dose; 200mg/kg) for 2 weeks. During 2 weeks, body weight and food intake were measured daily. 2weeks later we weighed epididymal and retroperitoneal white adipose tissues and liver. We examined the changes in plasma peptide level(ghrelin, amylin, PYY, leptin) and mRNA expression levels of ghrelin and Ghrelin o-acyltransferase(GOAT). Result : 1. The weight & rate of body weight gain showed significant different patterns between administration of Hyangsayukgunja-tang and control by times(P<0.005, Generalized Estimating Equation). The rate of body weight gain in rats was significantly increased at 8, 12, 14 days after administration of a high dose of Hyangsayukgunja-tang(P<0.025, Mann-Whitney U test). 2. Feed efficiency showed significant different patterns between administration of Hyangsayukgunja-tang and control by times(P<0.005, Generalized Estimating Equation). 3. The plasma ghrelin level in rats was significantly increased for 14days after administration of a low dose of Hyangsayukgunja-tang(P<0.005, Mann-Whitney U test). 4. The plasma PYY level in rats was significantly decreased for 14days after administration of a high dose of Hyangsayukgunja-tang(P<0.005, Mann-Whitney U test). 5. The mRNA expression levels of ghrelin and GOAT was up-regulated. Conclusion : In rats, Hyangsayukgunja-tang increased the mRNA expression levels of ghrelin and GOAT, thus Hyangsayukgunja-tang increases appetite by enhance the plasma ghrelin level, oppositely inhibit the plasma PYY level.

      • Aconiti tuber attenuates oxaliplatin-induced mechanical and cold allodynia in rats by suppression of spinal astrocyte activation

        Oxaliplatin, a widely used anticancer drug against metastatic colorectal cancer, can induce acute peripheral neuropathy, which is characterized by cold and mechanical allodynia. Activation of glial cells (e.g. astrocytes and microglia) and increase of pro-inflammatory cytokines (e.g. IL-1β and TNF-α) in the spinal cord play a crucial role in the pathogenesis of neuropathic pain. Our previous study demonstrated that Gyejigachulbu-Tang (GBT), a herbal complex formula, alleviates oxaliplatin-induced neuropathic pain in rats by suppressing spinal glial activation. However, it remains to be elucidated whether and how Aconiti Tuber, a major ingredient of GBT, is involved in the efficacy of GBT. Cold and mechanical allodynia induced by an oxaliplatin injection (6 mg/kg, i.p.) in Sprauge-Dawley rats were evaluated by a tail immersion test in cold water (4°C) and a von Frey hair test, respectively. Aconiti Tuber (300 mg/kg) was administrated orally for five consecutive days following the oxaliplatin injection. Glial activation in the spinal cord was quantified by immunohistochemical staining using GFAP (for astrocytes) and Iba-1 (for microglia) antibodies. The amount of spinal pro-inflammatory cytokines, IL-1β and TNF-α, were measured by ELISA. Significant behavioral signs of cold and mechanical allodynia were observed three days after an oxaliplatin injection. Oral administration of Aconiti Tuber significantly alleviated oxaliplatin-induced cold and mechanical allodynia by increasing the tail withdrawal latency to cold stimuli and mechanical threshold. Immunohistochemical analysis showed the activation of astrocytes and microglia and the increase of IL-1β and TNF-α levels in the spinal cord following an oxaliplatin injection. Administration of Aconiti Tuber suppressed the activation of spinal astrocytes without affecting microglial activation and down-regulated both IL-1β and TNF-α levels in the spinal cord. These results indicate that Aconiti Tuber has a potent anti-allodynic effect in a rat model of oxaliplatin-induced neuropathic pain, which is associated with the inhibition of activation of astrocytes and release of pro-inflammatory cytokines in the spinal cord. Thus, these findings suggest that administration of Aconiti Tuber could be an alternative therapeutic option for the management of peripheral neuropathy, a common side-effect of oxaliplatin. 연구 배경 및 목적 Oxaliplatin은 전이성 대장직장암에 널리 사용되는 항암제이나, 차가운 온도에 대한 자극이나 기계적 자극에 대한 이질통과 같은 말초 신경병증을 유발한다는 부작용이 있다. 이러한 신경통의 병리기전으로 척수내의 염증유발 cytokine들 (e.g. IL-1β and TNF-α)의 증가와 아교세포 (e.g. astrocytes and microglia)의 활성화로 볼 수 있다. 이전의 연구 결과에서는 계지가출부탕이 쥐 모델에서 척수의 아교세포의 활동을 억제하여 oxaliplatin에 의하여 유발된 신경통을 억제한다고 보고되었으나, 한의학적으로 온리(溫裏)약으로 분류가 되어있으며, 진통 작용이 있는 것으로 알려진 부자가 실제로 oxaliplatin에 의하여 유발된 신경성 이질통에 대하여 어떤 기전을 통해 진통 작용을 하는 것에 대해서는 연구된 바는 없었다. 연구방법 Sprauge-Dawley rat에 oxaliplatin을 투여하여 (6 mg/kg, i.p.) 한성 자극과 기계적 자극에 대한 이질통을 유발하였으며, 차가운 물에(4°C) 접촉시 꼬리를 드는 시간을 측정하여 한성 이질통을, von Frey hair test 로 기계적 자극에 대한 이질통을 매일 측정하였다. 부자는(300 mg/kg) oxaliplatin 주입 다음날부터 5일 연속으로 경구 투여되었다. 척수에서의 아교세포의 활성화는 by immunohistochemical (IHC) 염색방법 이후 관찰했으며, astrocyte 염색에는 GFAP 항체, microglia 염색에는 Iba-1 항체를 이용해 각각 염색하였다. 염증유발 cytokine들인 IL-1β 와 TNF-α는 ELISA를 통하여 관찰하였다. 연구결과 Oxaliplatin 주입을 한 이후 3일만에 기계적 자극에 대한 쥐의 역치의 변화는 물론 (P<0.01), 한랭자극에 대한 역치의 변화 (3일째에 P<0.05 , 5일째에 P<0.01) 를 보여 부자의 경구투여가 oxaliplatin에 의해 유발된 신경통을 완화하였다. Oxaliplatin 투여 이후 Immunohistochemical 분석상 척수에서의 성상교세포와 미세아교세포의 활성화와 IL-1β, TNF-α의 수적 증가가 관찰되었으며(P<0.01), 부자의 투여로 인해 염증유발 cytokine은 (IL-1β, TNF-α) 둘 다 유의하게 억제되었으며(P<0.01),, 척수에서의 성상교세포의 활성은 억제되었으나(P<0.01), 미세아교세포의 활성에는 영향을 주지 않았다. 결론 이러한 연구결과들은 부자가 쥐 모델에서 oxaliplatin에 의해 유발된 신경통을 완화해 준다고 볼 수 있으며, 그 기전은 척수에서의 성상교세포의 억제와 염증유발 cytokine 분비감소로 보여진다.

        The purpose of this study was to investigate the effects of sports club Activities on mental health of elementary students and also to provide a justification that the improvement of mental health required continuous efforts and attention. The subjects of this study were 200 elementary students studying at Gwangyang city, that is, 100 students who were participating in sports activity and another 100 students not participating in. MMPI(Minnesota Multiphasic Personality Inventory) which was revised by Wang Il(1997) was used to measure mental health. The collected data were analyzed via repeated measures of ANOVA with before- and after-measurements of each group using SPSS 16.0. And the following conclusions were made. Firstly, according to the pre- and post-test of mental health by gender, it showed significant differences in depression, hypomania and obsession by gender and measurement timing and it featured significant differences in introversion and hysteria by measurement timing, respectively. Secondly, according to the pre- and post-test of mental health by whether or not participating, it showed significant differences in depression and obsession by whether or not participating and measurement timing, and it featured significant differences in hypomania, introversion and hysteria by measurement timing, respectively.

      • 비폐색성 위 전정부 확장이 있는 흰쥐의 위 수축력, 체중 및 위 형태 복원에 대한 苓桂朮甘湯과 補中益氣湯 의 효능

        Objectives : The aims of this study were to evaluate whether rats with non obstructive antral dilation could be a useful tool resembling functional dyspeptic patients. Also we investigated the effect of Bojoongikki-tang(BJ), and Youngkaechulgam-tang(YK) in antral dilated rats. Methods : Non obstructive antral dilation was performed by first wrapping a non-absorbable rubber ring (D:6mm, W:4mm, T:1mm) around the 1st portion of the duodenum causing pyloric obstruction(PO). After 12weeks, except for some PO group rats used for control group, the rubber ring was removed by performing another operation. The antral dilated rats (AD) were then divided into three groups, a none treatment group(AD-NT), and two herbal medicine groups each given an extract solution each containing 125mg/kg of Youngkaechulgam-tang (AD-YK), and Bojoongikki-tang (AD-BJ) for 4 weels. Then the changes of gastric contractility, weight, as well as the morphologic changes of the stomach were evaluated for each groups including the normal intact group(NI). Results : Loss of weight and enlargement of the stomach surface area was seen in the PO group. Decrease of gastric motility index was observed in the AD-NT group, also the increased surface area of the stomach was not significantly different from the PO group. Youngkaechulgam-tang showed to increase gastric contraction, whereas Bojoongikki-tang showed no effect. Weight gain of rats were observed in both AD-YK and AD-BJ groups, but there seemed to be no change of the dilated stomach surface area. Conclusions : The non obstructive antral dilated rat seems to be an experimental pathologic model that reflects the gastric dysmotility similar to functional dyspeptic patients with antral dilation. Youngkaechulgam-tang is shown to increase both gastric contraction and weight in antral dilated rats, therefore it may be used for treating functional dyspepsia, however, Bojoongikki-tang should be used with caution in patients with gastric dysmotolity.
