CRISPR-Cas9-generated mouse model of neurofibromatosis type 1

Background To date, no experiments have been conducted to generate a neurofi bromatosis type 1 (NF1) mouse model using the clustered regularly interspaced short palindromic repeats (CRISPR)-CRISPR associated protein 9 (CRISPR-Cas9) embryo editing system. Objective This study was to deliver ribonucleoprotein (RnP) via electroporation in various formats and delivery methods of the CRISPR-Cas9 system for genetic modifi cation of mouse embryonic fi broblast NIH3T3 cells and embryos. The insertion– deletion (indel) effi cacy and pattern of NF1 were analyzed using next-generation sequencing (NGS). Results We established four candidate single-guide RNAs (sgRNAs) and transfected them with Streptococcus pyogenes Cas9 (SpCas9) protein (RnP) via electroporation into NIH3T3 cells to analyze the indel effi cacy and pattern of NF1. Two of the four candidates with a 50% indel effi cacy were selected for embryo editing; however, without an appropriate sgRNA concentration, biallelic mutants were generated in 60–80% of the cases. Thus, by fi nding an appropriate concentration for RnP, it was possible to increase the rate of monoallelic mutant generation. Finally, we successfully produced an NF1 heterozygote mouse model, and the mutant sequence was confi rmed using NGS. Conclusion Our study showed that the CRISPR-Cas9 embryo editing method is an effi cient tool for creating NF1 heterozygous ( NF1 +/− ) animal model.

Shin Suk-jung is one of the poets who have made an effort to explorer the poetic beauty of our pure Korean language. We could infer that his intention is covered in the repetition of specific poetic dictions as he chooses. This thesis scopes out the peculiarities and transitional aspects in all of Shin Suk-jung’s poetry quantitatively based on the poetic words that the previous studies described significantly.Two poetic words, ‘far image’ and ‘green image’ are selected as experimental materials. These are symbols of utopian and natural consciousness that have come to an agreement by all the researchers going deep into the poetry of Shin Suk-jung. With this research findings, internal significance in the same poetic words had been changed in his poetics. In addition to, it has come out into the open that ‘far image’ and ‘green image’ have an mutual complementary interrelationships. The modern history of Korea had effects on the periodical frequencies and changing significances of his poetic words. It is a achievement to have investigated the changing process of his poetic consciousness in identical poetic words as a poet had written. It was for his activity-oriented attitude that Shin Suk-jung had regarded ‘far image’ as something to designate Utopia.A romanticist usually makes it through the challenging life with his eye gazing into the distant goal. He tried to realize it in the real world so far from dreaming Utopia. Ordinary things could have their beings in Utopia as they should be precious. ‘Green image’ insinuates one’s practical methodology for it.Utopia like a mountain or a sky is far from here. However, Utopia’s ‘green’ color can be modeled in our real life. Shin Suk-jung holds on to his poetic consciousness-living on aiming for Utopia at all times by being alive to construe his ‘green image’ as a look of practical ethics.

본 연구에서는 직류 서보전동기의 위치, 속도 및 토오크 등의 제어기법중 외란에 대해 강인한 것으로 평가되고 있는 기존의 가속도 제어기법을 해석하고 그 결과 적용상의 문제점으로 판단되는 계통 파라미터 변화에 대한 강인성을 개선하기 위한 대책으로서 변경된 가속도제어 루프를 포함한 위치제어계를 제안하고 이를 직류전동기의 위치제어게 적용하였다. 제안된 제어계는 가속도제어기와 자기종조 퍼지 PID 제어기로 구성되어 있으며 제안된 제어계와 기존의 가속도제어계와의 근복적 차이점은 이 제어기법의 핵심 요소라 할 수 있는 가속도 기준입력 정정량의 발생기구로서 고전적 PID제어기를 자동조정하는 자기동조 PID 제어방식을 채택하였다는 점이다. 이 기준입력 발생기구를 도입한 직류전동기 위치제어계의 성능을 검토하기 위하여 그 응답특성을 기존의 가속도제어 위치제어계의 응답특성과 비교한 결과 파라미터 변화에 대한 강인성, 과도 특성 및 정상상태 특성면에서 제안된 제어계가 매우 우월한 성능을 가짐을 확인하였다. 따라서 제안된 제어기법은 기존의 가속도제어에 근거한 위치제어계의 성능을 개선시켜줌으로써 이 제어기법의 적용범위 확대에 기여하며 특히 모델의 불확정성이 크거나 부하등 운전환경이 크게 변화하는 악조건 하에서의 정밀한 위치제어에 적용될 수 있다. In this paper, a DC servo motor position control system based on acceleration control is proposed. The proposed control system consists of an acceleration controller and an auto-tuqing fuzzy PID controller. The auto-tuning fuzzy PID controller provides corrections for an acceleration reference to remove the effect of parametric uncertainties. And it comprises of the expert system which performs the automatic tuning of the PID controller parameters and the conventional PID controller. Expermental results demonstrate strate thi~tth e proposed overall control system has robust properties and good control performances with regard to unmeasurable disturbances and parameter variations. Therefore, the proposed control scheme enhances the applicability of an acceleration control approach and especially performs accurate position control under such an operating environment that model uncertainties exist and/or load, etc. change significantly.
