        소양호에 서식하는 쏘가리 Siniperca scherzeri의 성숙과 산란 특성

        본 논문은 소양호에 서식하는 내수면어업의 주요 수산자원 중 하나인 쏘가리 (Siniperca scherzeri)의 성숙과 산란에 관하여 연구하였다. 본 논문에 사용된 쏘가리는 2014년 4월부터 11월까지 정치망과 자망을 통해 채집하였고, 암컷이 204개체로 전장범위는 113~365 mm이었고, 수컷은 197개체로 전장 범위가 140~342mm이었다. 산란기가 시작되는 5월의 수온은 15°C를 나타냈으며, 점진적으로 높아져 7월에 26°C를 나타내었다. 암컷의 생식소 숙도지수 (GSI)는 5월에서 7월까지 각각 2.3%, 4.6%, 3.1%로 나타났고, 수컷의 경우 8.0%, 6.6%, 4.5%로 나타났다. 난소 조직은 5월에 많은 수의 샘플이 중숙으로 관찰되었으며 6월에는 대다수가 성숙, 7월에는 완숙 및 방란으로 관찰되었다. 정소 조직의 경우에도 마찬가지로 5월에는 대다수가 중숙, 6월에는 성숙, 7월에는 방중으로 관찰되었다. 월별 성숙도와 생식소 숙도지수 (GSI), 생식소의 조직관찰을 통한 분석결과, 소양호에 서식하는 쏘가리의 산란기는 5월에서 6월로 추정되었다. 또한 군성숙도를 분석한 결과, 암컷 쏘가리의 50% 성숙체장은 245.16mm로 나타났다. The study about maturity and spawning of Korean Mandarin fish, Siniperca scherzeri collected using the set net and gill net in Soyangho Lake from April to November 2014 was conducted. The number of individuals was 401 (female: 204, male: 197). The total length of females ranged from 113 mm to 365 mm and that of males was from 140 mm to 342 mm. The water temperature in May which is the start of spawning season indicated about 15°C and gradually increased to July (26°C). Gonadosomatic index (GSI) of females indicated 2.3%, 4.6% and 3.1%, respectively, in May to July. Males’ value showed the similar pattern recording 8.0%, 6.6% and 4.5% in the same time. According to histological observations of gonadal tissue, the most of female in May had the gonadal tissue of maturing and mature stage. And the ovary of June was mostly in the stage of ripe and spawning and a number of individuals in July was recovery stage. In the case of male, a number of males in May showed in the stage of mature. The testis of June showed that mature sperm was releasing. And the testis of July was mostly in recovery stage. The total length at 50% group maturity was estimated 245.16 mm. As shown in the above, the main spawning period of S. scherzeri was May to June.

        쏘가리 (Siniperca scherzeri) 말초혈액 백혈구의 미세구조 분석

        광학현미경과 전자현미경을 이용하여 쏘가리(Siniperca scherzeri0의 말초혈액 세포의 형태적 특징을 조사하였다. 쏘가리 말초 혈구는 크게 적혈구, 트롬보사이트, 백혈구 등 3가지로 구분되었다. 적혈구는 성숙 적혈구, 미성숙 적혈구, 적혈 아세포 등 3가지로 구분되었다. 트롬보사이트는 둥근형에서 방추난형으로 구분되었고, 핵은 이질염색질이 풍부한 난형이었으며, 세포질에는 소포, 미세간질이 관찰되었다. 림프구는 소형 림프구(type Ⅰ), 중형 림프구(type Ⅱ), 대형 림프구(type Ⅲ)로 구분되었다. 과립백혈구는 핵과 과립의 형태에 따라 이질핵 과립백 혈구와 호산성 백혈구로 구분되었는데, 이질핵 과립백혈구는 다형립 핵과 세포질내 크고 작은 과립이 풍부했고, 호산성 과립백혈구는 둥근 핵과 크고 작은 둥근 과립이 풍부했다. 과립백혈구의 과립의 형태는 크고 작은 방추형과 원형, 침형 또는 난형으로 구분되었다. 쏘가리의 단핵구/대식세포는 형태적으로 포유동물과 유사하였다. The morphological characteristics of peripheral blood cells in the freshwater mandarin fish (Siniperca scherzeri) were investigated by light and electron microscopy. Erythrocytes, thrombocytes, lymphocytes, granulocytes and monocytes were identified as the peripheral blood cells. Erthrocytes were divided into three form types: mature, immature, and erthroblasts. Thrombocytes were round to long oval (fusiform), each containing vesicular and microtubular structures and an oval nucleus with abundant heterochromatins. Lymphocytes were divided into three types in size: small, medium, and large. Granulocytes were distinguished into three types (type Ⅰ, type Ⅱ, and type Ⅲ) according to the morphology of the granules and nucleus. Heterophilic granulocytes possessed lobulated nuclei and a large number of cytoplasmic granules, each of which was oval and contained electron-dense materials and a crystalloid. Acidophilic granulocytes possessed large and small granules, and round nuclei. The granules were filled with regularly arranged fibriform materials, some needle-like structures, and oval forms. Monocytes and lymphocytes in this fish species were morphologically similar to those of mammals.

        한국산 쏘가리의 기원과 분자계통진화적 위치

        To explain the origin of the Korean mandarin fish (Siniperca scherzeri), phylogenetic relationships and DNA polymorphism among Siniperca fishes have been investigated based on mitochondrial cytochrome b DNA sequences. As a result, S. roulei were firstly differentiated early in the evolution of Siniperca fishes and the other six species (S. schezeri, S. undulata, S. fortis, S. obscura, S. knerii and S. chuatsi) were evolved slightly later. However, the order of species differentiation among six species was not clear because the nodes of their phylogeny were poorly resolved. The constructed molecular phylogeny revealed three genetically distinct groups of local populations of S. scherzeri. The first group (group 1) is the local populations of Korean peninsula and northern China including Lioaning and Henan. The second one (group 2) is the local populations of Anhui, Fujian and Guangxi. The third one (group 3) is the local population of Zhejiang. The number of nucleotide differences in base pairs were 31~43 between group 1 and 2; 37~44 between group 2 and 3; 27~29 between group 1 and 3; and 1~5 within group 1. Thus, the Korean mandarin fish was likely to be originated from the northern China local population which was isolated from the middle or southern China local populations during the Cenozoic Pliocene. Low level of sequence divergence between Korean mandarin fish populations and northern China population indicated a recent expansion of distribution ranges from northern China to Korean peninsula.

        To induce synchronized maturation of mandarin fish, Siniperca scherzeri by controlling photoperiod and water temperature, experimental fishes were maintained in filtered recirculating tanks and exposed to three experimental condition, control, 23℃-16L8D. Mean gonadosomatic index (GSI) of the fishes held at 23℃-16L8D was much highest 10.85 than the GSI of the control, 1.88. For sexual maturation of mandarin fish, photoperiod was believed to be a prime factor and water temperature was a compensative factor.

        사육 수온에 따른 쏘가리(Siniperca scherzeri) 치어의 성장 및 체조성 변화

        This study was conducted to investigate the effects of different water temperatures on growth, survival, biochemical composition, and blood physiological parameters of juvenile mandarin fish, Siniperca scherzeri. Feed-trained juvenile fish were subjected to four water temperatures (20, 23, 26 and 29℃) with two replicate groups. The fish were fed to apparent satiation twice daily using a formulated diet containing 55% crude protein and 6% crude lipid. After the 8-week feeding trial, survival was >96% in all groups. Weight gain and feed efficiency of fish reared at 26 and 29℃ were higher than those reared at 20℃. The protein efficiency ratio, daily feed intake, and whole body proximate composition were not affected by water temperature. These results indicate that a suitable water temperature range for optimal growth and feed efficiency of juvenile mandarin fish is 26-29℃ under these experimental conditions.

        사료 공급 횟수가 쏘가리(Siniperca scherzeri) 치어의 성장 및 체조성에 미치는 영향

        A feeding trial was conducted to investigate the effect of feeding frequency on the growth performance and body composition of juvenile mandarin fish Siniperca scherzeri. Triplicate groups of fish (initial fish weight, 7.5 g/fish) were fed to apparent satiation at one, two, three or four meals per day for 8 weeks. The results showed that weight gain of fish fed one meal per day was significantly (P<0.05) lower than those fish fed two, three or four meals per day. Feed efficiency of fish fed two meals per day was significantly (P<0.05) higher than other experimental groups. The plasma cholesterol content was significantly lower in fish fed one meal per day than those fed two, three or four meals per day. Consequently, we can conclude that feeding juvenile mandarin fish twice daily to apparent satiation is acceptable and sufficient to achieve good growth and feed efficiency, as fish performance was not remarkably enhanced when feeding increased from two to four times per day.

        Description of Nocardioides piscis sp. nov., Sphingomonas piscis sp. nov. and Sphingomonas sinipercae sp. nov., isolated from the intestine of fish species Odontobutis interrupta (Korean spotted sleeper) and Siniperca scherzeri (leopard mandarin fish)

        A polyphasic taxonomic approach was used to characterize three novel bacterial strains, designated as HDW12AT, HDW- 15BT, and HDW15CT, isolated from the intestine of fish species Odontobutis interrupta or Siniperca scherzeri. All isolates were obligate aerobic, non-motile bacteria, and grew optimally at 30°C. Phylogenetic analysis based on 16S rRNA sequences revealed that strain HDW12AT was a member of the genus Nocardioides, and closely related to Nocardioides allogilvus CFH 30205T (98.9% sequence identities). Furthermore, strains HDW15BT and HDW15CT were members of the genus Sphingomonas, and closely related to Sphingomonas lutea JS5T and Sphingomonas sediminicola Dae 20T (97.1% and 97.9% sequence identities), respectively. Strain HDW12AT contained MK-8 (H4), and strains HDW15BT and HDW15CT contained Q-10 as the respiratory quinone. Major polar lipid components of strain HDW12AT were diphosphatidylglycerol, phosphatidylglycerol, and phosphatidylinositol, and those of strains HDW15BT and HDW15CT were sphingoglycolipid, diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, and phosphatidylcholine. The G + C content of strains HDW12AT, HDW15BT, and HDW15CT were 69.7, 63.3, and 65.5%, respectively. The results of phylogenetic, phenotypic, chemotaxonomic, and genotypic analyses suggest that strain HDW12AT represents a novel species within the genus Nocardioides, and strains HDW15BT and HDW15CT represent two novel species within the genus Sphingomonas. We propose the names Nocardioides piscis for strain HDW12AT (= KACC 21336T = KCTC 49321T = JCM 33670T), Sphingomonas piscis for strain HDW15BT (= KACC 21341T = KCTC 72588T = JCM 33738T), and Sphingomonas sinipercae for strain HDW15CT (= KACC 21342T = KCTC 72589T = JCM 33739T).

        Effect of dietary protein and lipid level on growth, feed utilization, and muscle composition in golden mandarin fish Siniperca scherzeri

        A feeding trial was designed to assess the effects of dietary protein and lipid content on growth, feed utilization efficiency, and muscle proximate composition of juvenile mandarin fish, Siniperca scherzeri. Six experimental diets were formulated with a combination of three protein (35, 45, and 55%) and two dietary lipid levels (7 and 14%). Each diet was fed to triplicate groups of fish (8.3 ± 0.1 g) to apparent satiation for 8 weeks. The results showed that growth performance in terms of weight gain (WG) and specific growth rate (SGR) increased with increasing dietary protein level from 35 to 55% at the same dietary lipid level. At the same dietary lipid content, WG and SGR obtained with diets containing 55% protein was significantly higher than those obtained with diets containing 45 and 35% protein. No significant effect on growth rate was found when the dietary level of lipid was increased from 7 to 14%. While the levels of protein and lipid in the diets had no significant effect on feed intake, other nutrient utilization efficiency parameters including daily protein intake (DPI), feed efficiency (FE), and protein efficiency ratio (PER) showed a similar trend to that of growth rates, with the highest values obtained with diets containing 55% protein. Muscle chemical composition was not significantly affected by the different dietary treatments for each dietary lipid or protein level tested. These findings may suggest that a practical diet containing 55% protein and 7% lipid provides sufficient nutrient and energy to support the acceptable growth rates and nutrient utilization of mandarin fish juveniles.
