        A new coccolith modified electrode-based biosensor using a cognate pair of aptamers with sandwich-type binding

        <P><B>Abstract</B></P> <P>In this study, we report a cognate pair of the aptamer-based sandwich-type electrochemical biosensor for type 2 diabetes biomarker (Vaspin) using coccolith modified electrodeposited on the screen-printed gold electrode (CME-SPGE). The coccolith derived from E. huxleyi used in this study were known to be highly-structured microparticles with many nano-sized pores. The CME-SPGE was successfully fabricated by drop-casting coccoliths, followed by Au sputtering and electrodeposition of Au. On this CME-SPGE electrode, the sandwich-type electrochemical aptasensor was fabricated by using a cognate pair of aptamers. The morphological, electrochemical characteristics and the performances of both the CME-SPGE and the completely fabricated sandwich-type aptasensor were investigated by SEM, EDAX, cyclic voltammetry, and chronoamperometry. Due to the synergic effect of a cognate pair of aptamers on CME-SPGE, this newly developed sandwich-type electrochemical biosensor for Vaspin showed high specificity, and good sensitivity with a limit of detection (LOD) of 298 pM, along with more widen the linear range. To the best of our knowledge, this is the first report about the use of a coccolith modified electrode with a cognate pair aptamer resulting in sandwich-type binding in an electrochemical biosensor. With the advantages of using highly-structured biomineral microparticles and a cognate pair of aptamers, this new study may pave the innovative way to design a novel sandwich-type electrochemical aptasensor platform.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The original SPGE surface was successfully modified uniformly with the highly-structured coccolith microparticles. </LI> <LI> A cognate pair of aptamers was applied to fabricate sandwich-type electrochemical aptasensor successfully. </LI> <LI> This newly developed sandwich-type electrochemical biosensor was highly sensitive to type 2 diabetes biomarker, Vaspin. </LI> </UL> </P>

        샌드위치 타입의 세라믹-세라믹 관절면을 이용한 일차성 무시멘트 고관절 전치환술 (최소 10년 추시 결과)

        목적: 세라믹 라이너에 폴리에틸렌을 부착한 샌드위치형 라이너를 이용한 일차성 고관절 전치환술의 최소 10년 이상 추시 결과를 평가하였다. 대상 및 방법: 1998년 11월부터 2000년 12월까지 샌드위치형 라이너를 이용한 고관절 전치환술을 시행하고 최소 10년 이상 추시 가능하였던 34명(40예)을 대상으로 하였다. 평균 추시 기간은 134개월(120~145개월), 수술 시 평균 연령은 47.1세(24~65세)이었다. 임상적 평가는 Harris hip score, 대퇴부통증 등을 조사하였으며, 방사선학적 평가는 골내막 신생골, 방사선 투과성선, 삽입물의 이동 등을 관찰하였다. 결과: Harris hip score는 최종 추시에서 평균 90.9점으로 향상되었다. 전례에서 골성 고정을 얻었으며 삽입물의 이동이나 해리는 관찰되지 않았다. 합병증으로 수술 시 대퇴골의 비전위 골절이 2예(5%), 후방 탈구가 1예(2.5%), 세라믹 라이너 파손이 1예(2.5%)가 있었다. 결론: 샌드위치형 라이너를 이용한 고관절 전치환술의 최소 10년 이상 추시 생존율은 양호한 결과를 보였으나, 세라믹의 골절은 여전히 중요한 문제점으로 판단되며 비구컵의 정확한 고정과 지속적인 세라믹 부품의 재질 개선이 필요할 것으로 사료된다. Purpose: To evaluate the minimum 10-year follow up results of primary total hip arthroplasty (THA) performed using a sandwich-type (alumina-polyethylene-titanium) ceramic bearing. Materials and Methods: Thirty four patients (40 hips) who underwent a THA with sandwich typed liners from November 1998 to December 2000 were analyzed. Among the 34 patients, 25 were men and nine were women. Mean follow-up was 134 months (range, 120~145 months) and mean patient age at the time of THA was 47.1 years (range, 24~65 years). The clinical results were evaluated using the Harris hip score and level of thigh pain with a limping gait. The radiographic evaluation was done in terms of the endosteal new bone formation, radiolucent line, subsidence of the stem, migration of the acetabular cup, and proof of loosening. Results: The mean preoperative Harris hip score of 50.2 points (range, 31~87 points) was improved to 90.9 points (range, 75~99 points) at the final follow-up and thigh pain with limping gait in one case. All cases had fixation by bony ingrowth. No radiographically detectable loosening was observed in any hip. Fracture of ceramic liner in one case required change of the polyethylene liner. Conclusion: At the minimum 10-year follow up, survival rate as the end-point was favorable. However, fracture of the ceramic is still a major problem. Satisfactory results can be obtained by more precise surgical technique for acerabular inclincation and anteversion, and by improving the quality of the ceramic.

        Specific detection of avian influenza H5N2 whole virus particles on lateral flow strips using a pair of sandwich-type aptamers

        <P><B>Abstract</B></P> <P>We report a selection of a cognate pair of aptamers for whole avian influenza virus particles of H5N2 by using graphene-oxide based systemic evolution of ligands by exponential enrichment (GO-SELEX), and the application of a pair of sandwich-type binding aptamers on the lateral flow strips. The aptamers were characterized by GO-FRET assay, and Kd values of the selected aptamers were estimated to be from 6.913 × 10<SUP>5</SUP> to 1.27 × 10<SUP>6</SUP> EID<SUB>50</SUB>/ml (EID<SUB>50</SUB>/ml: 50% egg infective dose). Based on the evidence from confocal laser scanning microscope (CLSM), surface plasmon resonance (SPR), and circular dichroism (CD) spectrum analysis, the aptamers, J<SUB>3</SUB>APT and JH<SUB>4</SUB>APT, were found to be working as a cognate pair that binds to the target virus at the different sites simultaneously. This cognate pair of aptamers then was successfully applied on the lateral flow strips, clearly showing sandwich-type binding images with the presence of the certain numbers of H5N2 virus particles. On the newly developed lateral flow strips, the target virus was detectable down to 6 × 10<SUP>5</SUP> EID<SUB>50</SUB>/ml in the buffer and 1.2 × 10<SUP>6</SUP> EID<SUB>50</SUB>/ml in the duck's feces, respectively, by the naked eye. By using the ImageJ software, the LOD was found to be 1.27 × 10<SUP>5</SUP> EID<SUB>50</SUB>/ml in the buffer and 2.09 × 10<SUP>5</SUP> EID<SUB>50</SUB>/ml in the duck's feces, respectively. Interestingly, on the lateral flow strips, enhanced specificity towards the target virus (H5N2) appeared over other subtypes of H5Nx. To the best of our knowledge, this is the first report about the application of the cognate pair of aptamers for the detection of influenza virus on the lateral flow strips. This study shows the promising perspective of a cognate pair of aptamers for the on-site detection system which could be useful for rapid detection of avian influenza viruses for preventing the pandemic influenza viruses from spreading.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A cognate pair of aptamers binding to avian influenza H5N2 whole virus particles in a sandwich-type was successfully screened for the first time. </LI> <LI> This sandwich-type cognate pair of aptamers was successfully applied on lateral flow strips for the first time. </LI> <LI> This aptamer-based sandwich-type lateral flow strip biosensor is sensitive and specific to avian influenza H5N2 whole virus. </LI> </UL> </P>

        Recently, electrochromic devices (ECDs) have gathered increasing attention owing to their high color contrast and memory effect, which make them highly applicable to smart windows, auto-dimming mirrors, sensors, etc. Traditional ECDs have a sandwich structure that contains an electrochromic layer between two ITO substrates. These sandwich-type devices are usually fabricated through the lamination of two electrodes and followed by the injection of a liquid electrolyte in the inner space. However, this process is sometimes complex and time consuming. In this study, we fabricated ECDs with a lateral electrode structure that uses only an ITO substrate and an all-in-one electrochromic gel, which is a mixture of electrolyte and electrochromic material. Furthermore, we investigated the EC properties of the lateral-type device by comparing it with a sandwich-type device. The lateral-type ECD shows strong blue absorption as the applied voltage increases and has a competitive coloration efficiency compared to the sandwich-type device.

        나노구조체 에너지 하베스팅 소자와 IoT 센서 네트워크의 융합 연구

        We have demonstrated a sandwich-type ZnO-based piezoelectric energy harvesting nanogenerator, namely ZCZ-NG device, composed of symmetrically stacked layers of ZnO/carbon tape/ZnO structure. Especially, we have adopted a conductive double-sided adhesive carbon tape in an effort to fabricate a high-quality ZCZ-NG device, leading to its superior output performance in terms of the peak-to-peak output voltage. Effects of the device size, ZnO layer thickness, and bending strain rate on the device performance have been investigated by measuring the output voltage. Moreover, to evaluate the effectiveness of the fabricated ZCZ-NG devices, we have experimentally implemented a sensor network testbed which can utilize the output voltages of ZCZ-NG devices. This sensor network testbed consists of several components such as Arduino-based transmitter and receiver nodes, wirelessly transmitting the sensed information of each node. We hope that this research combining the ZnO-based energy harvesting devices and IoT sensor networks will contribute to the development of more advanced energy harvester-driven IoT sensor networks in the future. 본 논문에서는 산화아연/탄소 테이프/산화아연의 대칭 구조를 갖는 ZCZ-NG라는 샌드위치형 산화아연(ZnO) 압전 에너지 하베스팅 소자를 제시한다. 고품질의 ZCZ-NG 소자를 제작하기 위해 전도성 양면 접착 탄소 테이프를 활용하였으며, 이는 매우 높은 피크 투 피크 전압(Vpp)을 발생시키는 ZCZ-NG 소자의 개발로 이어졌다. ZCZ-NG 소자의 크기, 산화아연 층의 두께 그리고 벤딩 변형률 변화에 따른 ZCZ-NG 소자의 출력 성능 변화를 측정, 분석하였다. 또한 제작된 ZCZ-NG 소자의 실효성 및 응용 가능성을 검증하기 위한 실험적인 센서 네트워크 테스트베드를 구축하였다. 상용 아두이노를 기반으로 한 송신, 수신 노드들로 이루어진 테스트베드에서 노드들은 각 노드에서 감지한 정보들을 무선으로 송수신한다. 본 연구에서 사용된 대칭 구조의 샌드위치형 ZCZ-NG 소자 제작 기술과 센서 네트워크와의 융합 연구가 앞으로 더 발전되어 사물인터넷 구현을 위한 자가발전 센서 네트워크 연구에 도움이 되길 바란다.
