        야생동충하초로부터 우량균주선발 (I)

        1999년 5월 중순부터 10월까지 완주군 일대의 연석산, 모악산, 대아리 수목원의 해발 약 400m의 그늘지고 습한곳에서 약 20여종의 동충하초를 채집하였다. 이 중 2속 5종을 확인하였고 이것들에서 10균주를 분리하여 9균주를 자실체유도 실험에 이용하였다. 그결과 Cordyceps militaris (99-0007) 1균주, Isaria japonica (99-0001, 99-0002, 99-0003) 3균주, Cordvceps soborifera (99-0008) 1균주, C.kanzashiana (99-0009) 1균주는 이용가치가 충분히 있다고 사료되어 우량균주로 선발하였다. Many Cordyceps rpp were collected for excellent strains selection at Mts. Moak, Younsuk and Daea-ree herbarium near Wanju-kun from mid May to October,1999. They were identified as the resulting 2 genera and 5 species, 10 strains were got through 5 species culture. Among them 4 species used for induction of artifitial fruiting body and 6 strains of them were selected to excellent strains: Cordyceps militaris (99-0007), Isaria japonica (99-0001, 99-0002, 99-0003), Cordyceps soborifera (99-0008), Cordyceps kanzashiana(99-0009).

        동충하초에서 분리된 acetyepoxyltrichothecenediol에 의한 인간 백혈병 HL-60 세포의 세포고사 연구

        N/A Objectives : It has been reported that the trichothecene mycotoxin 4-acetyl-12,13-epoxyl-9-trichothecene-3,15-diol (AETD) from the fruiting bodies of Isaria japonica Yasuda is a potent inducer of apoptosis in human promyelocytic HL-60 cells. Methods : The present study aims to characterize the molecular events leading to AETD-induced apoptosis in HL-60 cells. Results : The percentage of apoptotic cells (annexin V-positive cell population) increased dose- and time-dependently after AETD exposure. Apoptosis of HL-60 cells by AETD was associated with the formation of intracellular reactive oxygen species (ROS), the depletion of intracellular glutathione (GSH), and the activation of caspase-3. Pre-treating the cells with the antioxidant N-acetyl-L- cystein (NAC) and the caspase-3 inhibitor Z-DEVD-fmk abrogated AETD-induced apoptosis and caspase-3 activation. NAC blocked intracellular ROS formation and GSH depletion, but Z-DEVD-fmk did not. Conclusions : These results indicate that AETD induces apoptosis in HL-60 cells by causing intracellular ROS formation and GSH depletion followed by the downstream event of caspase-3 activation.

        Anti-inflammatory Effects of Various Mushrooms in LPS-stimulated RAW264.7 Cells

        Mushrooms have been widely cultivated and consumed as foods and herbal medicines owing to their various biological properties. However, few studies have evaluated the anti-inflammatory effects of mushrooms. Here, we investigated the effects of mushroom extracts (MEs) on lipopolysaccharide (LPS)-induced inflammation in macrophages (RAW264.7 cells). First, we extracted MEs with either water or ethanol. Using LPS-treated RAW264.7 cells, we measured cell proliferation and NO production. Gene expression of tumor necrosis factor-α (TNF-α), interleukin (IL)-6 (IL-6), and IL-1β was assessed by RT-PCR, and protein abundance of inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) and phosphorylation of p65 were determined by immunoblotting. MEs prepared using both water and ethanol inhibited LPS-induced inflammation in RAW264.7 cells. Nitric oxide (NO) levels induced by LPS were reduced by treatment with MEs. Isaria japonica Yasuda water extracts and Umbilicaria esculenta (Miyoshi) Minks ethanol extracts significantly decreased the mRNA expression of inflammation-related cytokine genes including TNF-α, IL-6, and IL-1β. Similarly, the protein abundance of iNOS and COX-2 was also decreased. The phosphorylation of p65, a subunit of nuclear factor-κB was at least partly suppressed by MEs. This study suggests that mushrooms could be included in the diet to prevent and treat macrophage-related chronic immune diseases.

      • 국내 자생 눈꽃동충하초 균의 명명 고찰

        눈꽃동충하초는 국내에서 몇 가지 학명으로 불려지고 있으나 그 중 오명이 포함되어 있음으로 인해 학자들간에 논란이 되고 있으며, 균학도들에 의해서 잘못 인용되는 사례가 빈번함에 따라 본 시험에서는 균을 재 동정코자 하였다. 수집균은 담황색 분생자병속을 1∼5여개 형성하며 그 끝마디에는 다량의 분생포자가 발생되었다. MA 배지 상에서는 25℃, 14일간 직경 2.1㎝로 더딘 생장을 보이며, colony의 색상은 앞면은 옅은 담황색, 뒷면은 연노랑 색이며, 분생포자는 타원형 혹은 난형으로 2.9∼6.6×1.5∼2.5 ㎛이며 분생포자를 지탱하고 있는 phialide는 부풀고 팽창한 형태로 3∼4개가 형성되며 4.0∼6.6×2.0∼2.2 ㎛이다. 본 균주는 Samson 분류에 의거하여 주요 특성이 일치하여 Paecilomyces tenuipes(Peck) Samson 균과 동일 종으로 인정되었다. Paecilomyces tenuipes (Nunkkot Dongchunghacho) was erroneously treated in three different scientific names: Paecilomyces japonica,k Isaria japonica and Paecilomyces tenuipes. Since the fungus has been widely used in various fields of research and industry, its nomenclatural uncertainty caused many problems. Therefore, morphological studies of korean isolate were carried out and compared with published data on other isolates of P. tenuipes to establish its proper taxonomec position.
