Purpose: To evaluate the cysticidal effect of 5 kinds of commercially available contact lens disinfectants against 2 clinical isolates of Acanthamoeba. Methods: Five kinds of commercially available contact lens disinfectants were soaked with cysts of Acanthamoeba ludgdunesis and castellanii at the concentration of 103, 104, and 105 cells/ml for 1 and 4 or 6 hours. Cysts which were not excysted in 7 days after treatment were recognized to be killed. Morphologic changes were evaluated by electron microscopic observation. Results: Contact lens disinfectants which contain myristamidopropyl dimethylamine (MAPD) showed the best cysticidal effect. These disinfectants demonstrated a cysticidal effect on both Acanthamoeba species of all concentrations in 6-hour treatment. Contact lens disinfectants which contain polyhexamethylene biguanide (PHMB) did not demonstrate cysticidal effect, except for Acanthamoeba castellanii at the concentration of 103 cells/ml, in either 4- or 6-hour treatment. Separation of plasma membrane from endocyst and damage of organelles were prominent in cases showing a cysticidal effect. Conclusions: Contact lens disinfectant which contains MAPD may be helpful in preventing the Acanthamoeba keratitis. A higher concentration of PHMB is required to be effective in preventing Acanthamoeba keratitis.

Alterations in mitophagy have been increasingly linked to aging and age-related diseases. There are, however, no convenient methods to analyze mitophagy in vivo. Here, we describe a transgenic mouse model in which we expressed a mitochondrial-targeted form of the fluorescent reporter Keima (mt-Keima). Keima is a coral-derived protein that exhibits both pH-dependent excitation and resistance to lysosomal proteases. Comparison of a wide range of primary cells and tissues generated from the mt-Keima mouse revealed significant variations in basal mitophagy. In addition, we have employed the mt-Keima mice to analyze how mitophagy is altered by conditions including diet, oxygen availability, Huntingtin transgene expression, the absence of macroautophagy (ATG5 or ATG7 expression), an increase in mitochondrial mutational load, the presence of metastatic tumors, and normal aging. The ability to assess mitophagy under a host of varying environmental and genetic perturbations suggests that the mt-Keima mouse should be a valuable resource.
Design of highly uniform spool and bar horns for ultrasonic bonding

<P>Although the groove and slot have been widely utilized for horn design to achieve high uniformity, their effects on uniformity have not been analyzed thoroughly. In this work, spool and bar horns for ultrasonic bonding are designed in a systematic way using the design of experiments (DOE) to achieve high amplitude uniformity of the horn. Three-dimensional modal analysis is conducted to predict the natural frequency, amplitude, and stress of the horns, and the DOE is employed to analyze the effects of the groove and slot on the amplitude uniformity. The design equations are formulated to determine the optimum dimensions of the groove and slot, and the uniformity is found to be influenced most significantly by the groove depth and slot width. Displacements of the spool and bar horns were measured using a laser Doppler vibrometer (LDV), and the predicted results are in good agreement with the experimental data.</P>

<P>Efficient isolation of circulating tumor cells (CTCs) from whole blood is a major challenge for the clinical application of CTCs. Here, we report an efficient method to isolate CFCs from whole blood using highly dense and transparent silica microbeads. The surfaces of silica microbeads were fully covered with an antibody to capture CFCs, and blocked by zwitterionic moieties to prevent the non-specific adsorption of blood cells. Owing to the high density of the silica microbeads, the complexation of CTCs with silica microbeads resulted in the efficient sedimentation of CTC-microbead complexes, which enabled their discrimination from other blood cells in density gradient media. Model CFCs (MCF-7, HCC827, and SHP-77) with various levels of epithelial cell adhesion molecule (EpCAM) were isolated efficiently, especially those with low EpCAM expression (SHP-77). Moreover, the transparency of silica microbeads enabled CTCs to be clearly identified without interference caused by microbeads. The improved sensitivity resulted in increased CTC recovery from patient samples compared with the FDA-approved CellSearch system (14/15 using our method; 5/15 using the CellSearch system). These results indicate that the isolation method described in this report constitutes a powerful tool for the isolation of CTCs from whole blood, which has important applications in clinical practice. Genomic Alterations in Biliary Tract Cancer Using Targeted Sequencing <sup>1</sup>

<P><I>Background:</I> Biliary tract cancers (BTCs) are rare and heterogeneous group of tumors classified anatomically into intrahepatic and extrahepatic bile ducts and gallbladder adenocarcinomas. Patient-derived tumor cell (PDC) models with genome analysis can be a valuable platform to develop a method to overcome the clinical barrier on BTCs. <I>Material and Methods:</I> Between January 2012 and June 2015, 40 BTC patients’ samples were collected. PDCs were isolated and cultured from surgical specimens, biopsy tissues, or malignant effusions including ascites and pleural fluid. Genome analysis using targeted panel sequencing as well as digital multiplexed gene analysis was applied to PDCs as well as primary tumors. <I>Results:</I> Extrahepatic cholangiocarcinoma (<I>N</I> = 15, 37.5%), intrahepatic cholangiocarcinoma (<I>N</I> = 10, 25.0%), gallbladder cancer (<I>N</I> = 14, 35.0%), and ampulla of Vater cancer (<I>N</I> = 1, 2.5%) were included. We identified 15 mutations with diverse genetic alterations in 19 cases of BTC from primary tumor specimens. The most common molecular alterations were in TP53 (8/19, 42.1%), including missense mutations such as C242Y, E285K, G112S, P19T, R148T, R248Q, and R273L. We also detected two NRAS mutations (G12C and Q61L), two KRAS mutations (G12A and G12S), two ERBB2 mutations (V777L and pM774delinsMA) and amplification, and three PIK3CA mutations (N345K, E545K, and E521K). PDC models were successfully established in 27 of 40 samples (67.5%), including 22/24 from body fluids (91.7%) and 5/16 from tissue specimens (31.3%). <I>Conclusions:</I> PDC models are promising tools for uncovering driver mutations and identifying rational therapeutic strategies in BTC. Application of this model is expected to inform clinical trials of drugs for molecular-based targeted therapy.</P>

The bar horn is designed to increase uniformity of the displacement on the output face through simulation and experiments. Three-dimensional modal analysis is conducted using the finite element method to calculate the vibration mode and displacement on the output face, and the design of experiment (DOE) technique is employed to determine the optimum dimensions of the groove and slot so that the high amplitude uniformity of the bar horn is produced. Displacement of the bar horn was measured using the Laser Doppler Vibrometer (LDV), and the experimental results show good agreements with the predicted results. High uniformity of the bar horn is achieved with the dimensions of the groove and slot determined using the design equations.
