탈지대두박에서의 Isoflavone의 분리를 위한 흡착제의 조건

탈지대두박을 이용한 isoflavone의 분리 및 정제를 위한 최적 조건을 찾고자 absorption resin인 Amberlite XAD-1180를 사용하여 흡착제의 활성화용매의 농도, 흡착된 isoflavone의 용출용매의 농도, 용출용매의 양과 용출속도, 시료와 흡착제의 비율 그리고 수지에 미치는 pH와 ion의 영향을 조사하였다. 수지의 활성화와 용출에는 Amberlite XAD-1180의 경우 methanol 75%로 활성화시키기 ethanol 80%로 용출시키는 경우가 가장 높은 값을 보였으며, 이때 사용하는 absorption resin은 isoflavone 흡착을 위해 사용한 단백질제거 탈지대두박용액양의 약 0.5배로, ethatnol의 양은 단백질제거 탈지대두박용액 양의 4배를 사용하여 flow rate 12mL/min의 경우가 적절한 조건임이 밝혀져 흡착제 사용이 isoflavone 함량을 최고 2~3배까지 향상시키는 결과를 보였다. 이때 분리된 isoflavone의 순도는 최고 40%까지 얻을 수 있었다. pH와 ion의 영향에서는 pH는 5.0,NaCl은 0.25N 이하에서 isoflavone의 유출량이 높았다. Conditions for sorption and elution of isoflavone with using amberlite XAD-1180 were studied to separate Isoflavone from defatted soybean flour (DSF). Isoflavone analyzed were daidzin, genistin, glycitin, daidzein, genistein and glycitein by HPLC. The result showed that washing Amberlite XAD-1180 with 75% methanol and then elution of isoflavone with 80% ethanol resulted the highest amounts of isoflavone recovered from the absorption resin. The most proper eluent factor was 4(eluent/sample volume), the sample factor was about 2(the volum of sample/the weight of resin) ad flow rate 12 mL/min. The pH 5.0 and less than 0.25 N NaCl were found to be proper for seperation of isoflavone. The isoflavone contents after seperation with absorption resin were about 40% in dry weight basis.

In order for a novel test method to be applied for regulatory purposes, its reliability and relevance, I.e., reproducibility and predictive capacity, must be demonstrated, Here, we examine the predictive capacity of a novel non-radioisotopic local lymph node assay, LLNA:BrdU-FCM (5-bromo-2`-deoxyuridine-flow cytome-try), with a cutoff approach and inferential statistics as a prediction model. 22 reference substances in OECDTG429 were tested with a concurrent positive control. Hexylcinnamaldehyde 25%(PC), and the stimulation index(SI) representing the fold increase in lymph node cells over the vehicle control was obtained. The optimal cutoff SI(2.7≤cutoff<3.5), with respect to predictive capacity, was obtained by a receiver operating character-istic curve, which produced 90.9% accuracy for the 22 substances. To address the inter-test variability in respon-siveness, SI values standardized with PC were employed to obtain the optimal percentage cutoff(42.6≤Cutoff<57.3%of PC), which produced 86.4% accuracy. A test subastance may be diagnosed as a sensitizer if a statistically significant increase in Si is elicited. The parametric one-sided t-test and non-parametric Wilcoxon rank-sum test produced 77.3% accuracy. Similarly, a test substance could be defined as a sensitizer if the SI means of the vehicle control, and of the low, middle, and high concentrations were statistically significantly different, which was test-ed using ANOVA or Kruskal-Wallis, with post hoc analysis, Dunnett, or DSCF(Dwass-Steel-Critchlow-Fligner), respectively, depending on the equal variance test, producing 81.8% accuracy. The absolute SI-based cutoff approach produced the best predictive capacity, however the discordant decisions between prediction models need to be examined further.

Objective The cognitive, Affective, and Somatic Empathy Scale (CASES) suggests novel three components structure of empathy. Although CASES developed to assess the trait empathy of both children and adult, the validation in adult sample is not yet conducted. This study developed Korean version of CASES (CASES-K) and examined the psychometric properties in young adult for first time. Methods Confirmatory factor analysis was conducted to test whether the factor structure suggested in initial development study is valid in Korean young adult population. 350 young adults participated, and 44 of them also answered to other empathy scales for validation. The construct validity was ascertained through a series of correlational analysis with widely used trait empathy scales. Results The result of confirmatory factor analysis supports the three-factor model of empathy consists of affective, cognitive and somatic facets. The result of correlational analysis supports the validity of CASES-K, and especially the separation of somatic empathy from affective empathy. Conclusion The current study provides the first evidence of the validity and utility of the CASES-K in young adult population. The result also supports the commonality of three factor structure of empathy in both East Asian and Caucasian, from the children to adults.
Enhanced anti-scratch performance of nanopatterned anti-reflective polymer films

<P><B>Abstract</B></P> <P>A heat treatment process was proposed to enhance the anti-scratch performance of nanostructured anti-reflection (AR) PMMA films fabricated by thermal nanoimprint lithography (t-NIL). Nanostructured AR films were fabricated using the high-temperature/high-pressure flat pressing (FP) to control free volume and rapid cooling (RC) to induce a compressive residual stress, which are compatible with t-NIL. The effects of the heat treatment process on the optical and the mechanical properties of the nanostructured films were examined by evaluating the optical properties, such as reflectance and transmittance, and measuring the mechanical properties with a pencil hardness test and a nanoindentation. It was found that the anti-scratch performance of the film was significantly improved while the optical properties are degraded slightly by a combination of FP and RC processes. It was further confirmed that the enhanced anti-scratch performance is due to the improved elastic modulus of the film by the optimized sequence of the process consisting of FP, RC and t-NIL. Antiadhesive effect and safety of oxidized regenerated cellulose after thyroidectomy

Purpose: To evaluate the antiadhesive effects and safety of an oxidized regenerated cellulose (Interceed) after thyroidectomy. Methods: Seventy-six thyroidectomized patients were prospectively randomized into two groups with regard to the use of Interceed. We evaluated each group for their adhesive symptoms using four subjective and four objective items at the 2nd week, 3rd and 6th month after thyroidectomy. All patients were examined for vocal cord motility by indirect laryngoscope at each period. Results: Total adhesion scores at each postoperative follow-up period decreased with time, but were not significantly different in each group. The median score for swallowing discomfort for liquid was significantly lower in the Interceed group than in the control group 2 weeks after surgery. In addition, the severity of skin adhesion to the trachea was reduced in the Interceed group compared with the control group 6 months after surgery. During the study, there were no adverse effects or significant differences in postoperative complications between the groups. Conclusion: Interceed appeared to be safe and effective in improving neck discomfort at early postoperative periods and preventing skin adhesion to the trachea 6 months after thyroidectomy.
Spontaneous Splenic Rupture as a Paradoxical Reaction during Treatment for Splenic Tuberculosis

This report describes a rare case of a patient with splenic tuberculosis (TB) who developed spontaneous splenic rupture after 10 weeks of antituberculous chemotherapy. The patient responded well to the antituberculous regimen prior to the spontaneous splenic rupture. We considered a paradoxical reaction as a cause of the splenic rupture. The patient underwent splenectomy and continuously received initial antituberculous drugs without change. To the best of our knowledge, this is the first report of spontaneous splenic rupture as a paradoxical reaction to antituberculous chemotherapy in an immunocompetent host with splenic TB.
