        Two new species of Matileola (Diptera, Keroplatidae) from China

        Only three species belong to Matileola in previously described species of Keroplatidae from world. Here two species are added to the fauna of Chinese mainland, Matileola motuoensis sp. nov. and Matileola sejilaensis sp. nov., which are described and illustrated as new to science.         First record of the subgenus Tipula (Ramatipula) (Diptera, Tipulidae) from China, with description of two new species

        The subgenus Ramatipula Alexander, 1971 of the genus Tipula is recorded for the first time in China. Two new species, Tipula (Ramatipula) dolabcorolla, sp. nov., and Tipula (Ramatipula) spathulata, sp. nov. are described and illustrated.         Biomechanical Evaluation of 2 Endoscopic Spine Surgery Methods for Treating Lumbar Disc Herniation: A Finite Element Study

        Objective: This study aimed to evaluate the effects of 2 endoscopic spine surgeries on the biomechanical properties of normal and osteoporotic spines. Methods: Based on computed tomography images of a healthy adult volunteer, 6 finite element models were created. After validating the normal intact model, a concentrated force of 400 N and a moment of 7.5 Nm were exerted on the upper surface of L3 to simulate 6 physiological activities of the spine. Five types of indices were used to assess the biomechanical properties of the 6 models, range of motion (ROM), maximum displacement value, intervertebral disc stress, maximum stress value, and articular protrusion stress, and by combining them with finite element stress cloud. Results: In normal and osteoporotic spines, there was no meaningful change in ROM or disc stress in the 2 surgical models for the 6 motion states. Model N1 (osteoporotic percutaneous transforaminal endoscopic discectomy model) showed a decrease in maximum displacement value of 20.28% in right lateral bending. Model M2 (unilateral biportal endoscopic model) increased maximum displacement values of 16.88% and 17.82% during left and right lateral bending, respectively. The maximum stress value of L4–5 increased by 11.72% for model M2 during left rotation. In addition, using the same surgical approach, ROM, maximum displacement values, disc stress, and maximum stress values were more significant in the osteoporotic model than in the normal model. Conclusion: In both normal and osteoporotic spines, both surgical approaches were less disruptive to the physiologic structure of the spine. Furthermore, using the same endoscopic spine surgery, normal spine biomechanical properties are superior to osteoporotic spines.

      • Induction of Cytotoxicity and Apoptosis in Human Gastric Cancer Cell SGC-7901 by Isovaltrate Acetoxyhydrin Isolated from Patrinia heterophylla Bunge Involves a Mitochondrial Pathway and G2/M Phase Cell Cycle Arrest

        Background: Our previous study demonstrated cytotoxicity of a crude extract from Patrinia heterophylla Bunge (PHEB). In the present study, we aimed to investigate the effects of isovaltrate acetoxyhydrin (IA) isolated from PHEB on the gastric cancer cell SGC-7901, in order to explore a potential treatment for gastric cancer. Methods: MTT assays were employed to determine the effects of IA on cell vitality and proliferation, with monitoring of cell morphology changes and examination of apoptosis with Annexin V-PI staining. Flow cytometry was used to assess cell cycle progression and mitochondrial membrane potential. The activity of caspase 3, 9 was evaluated by spectrophotometry, and the protein levels of Bax, Bcl2 and Cyclin B1 were analyzed with Western blotting of total proteins extracted from cultured cells. Results: The results demonstrated direct toxicity of IA towards SGC-7901 cells. Evidence of apoptosis included blebbing and chromatin condensation. Annexin V-PI assays revealed early apoptosis, involving rapid depolarization of mitochondrial membranes and activity of caspase 3, 9 signaling pathways. Western blotting showed that Bcl2 and Bax proteins was down- and up-regulated, respectively, and cyclin B1 was up-regulated. Cell cycle analysis further indicated that IA could induce G2/M phase arrest in SGC-7901 cells. Conclusions: In conclusion, we believe that IA induces apoptosis of SGC-7901 cells, therefore providing a potential therapeutic agent for treatment of gastric cancer.

        Liaohe National Park based on big data visualization Visitor Perception Study

        국립공원은 세계 자연 보존 연맹(WWF)이 수립한 보호지역 관리 체계의 중요 유형 중 하나이며, 또한 자연 및 문화 유산의 효과적인 보호와 지속적인 이용을 실현하는 세계 각국의 관리 모델이다. 이러한 공원은 보호, 과학 연구, 교육, 레크리에이션 및 지역 개발을 비롯한 중요한 역할을 담당하다. 대용량 데이터의 배경 아래, 본 연구는 전 세계 연안 습지의 대표적인 대상인 중국 랴오하 국립공원을 사례 지역으로 삼아 파이썬 기술을 사용하여 중국의 주요 관광 OTA 사이트 중 하나인 망픈웨이(Mafengwo), 셰어이(Gonglve), 큐난우(Chujingyou), 메이툰(Meituan) 및 대중점평넷(Dianping)의 관광객 여행기와 댓글을 데이터 소스로 수집하였다. 텍스트 시간 범위는 2015년부터 2022년까지이며, 총 2,998개의 댓글과 166,588개의 단어를 포함하다. ROST 콘텐츠 마이닝 및 Gephi 소프트웨어를 사용하여 랴오하 국립공원 방문객의 만족도, 인지 과정, 공선 네트워크, 감정 성향 등을 시각적 분석하였다. 결과는 다음과 같다. 야생 동물 및 식물 자원, 강과 바다가 결합 된 자연 경관, 습지 생태는 랴오하 국립공원 방문객의 인식에서 충분히 반영되었다. 방문객은 랴오하 국립공원에 대해 강한 긍정적인 감정을 가지고 있지만, 시설 서비스, 대중교육, 방문객 참여 경험 등에서 여전히 개선할 여지가 있다. National parks are one of the important types of protected area management systems established by IUCN and a management model for implementing effective conservation and sustainable use of natural and cultural heritage in countries around the world, and they assume important roles in conservation, scientific research, education, recreation and driving community development. In the context of big data, this study takes Chinas Liaohe National Park, a typical representative of global coastal wetlands, as a case study, and using Python technology to collect tourists travelogues and reviews from major OTA websites in China as a source. The text spans from 2015 to 2022 and contains 2998 reviews with 166,588 words in total. The results show that wildlife resources, natural landscape, wetland ecology and the fishing and hunting culture of northern China are fully reflected in the perceptions of visitors to Liaohe National Park; visitors have strong positive feelings toward Liaohe National Park, but there is still much room for improvement in supporting services and facilities, public education and visitor experience and participation.

        Radon-induced demethylation of Cdk2 CpG island in the rat lung

        Radon exposure has been linked to lung carcinogenesisin both human and animal studies. The identificationof sequential changes in DNA methylation duringtumour progression and the elucidation of their interplaywith genetic changes will broaden our molecular understandingof this disease. Rats were exposed to 120 or 400working level months (WLM) of radon, lung pathologicalchanges were examined by haematoxylin and eosin staining,lung single cell suspension cell cycles were detected byflow cytometry, lung cell cycle regulated gene (Cdkn2a,P53, Cdk4/2, Mdm4/2 and Rb1 genes) expression wasquantified by real-time PCR and methylation of CpG islandsin the promoters of cell cycle-regulated genes were detectedby bisulfite sequencing PCR. The alveolar walls of rat lungsafter exposured to radon exhibited papillae and the lungbronchial epithelial cells stained positively for proliferatingcell nuclear antigen. The bronchial epithelial cells displayedsome hyperplasia after challenged by 400 WLM of radon. Moreover, G1 arrest decreased; Rb1, Mdm2/4, and Cdk2/4expression decreased and Cdk2 was demethylated at thesecond and sixth CpG loci from base pairs 3092704 to3092953 of chromosome 7. Cdk2 demethylation may beapplicable as a biomarker of early lung damage that wasinduced by radon and other environmental carcinogens.

        Engineering of a Bacillus subtilis Strain Deficient in Cellulase: Application in Degumming of Ramie

        Degumming is the most important step before ramie fibers are used in textile industry. Conventional chemical degumming technology with alkaline treatment at high temperature causes critical problems in environment. In addition to multiple degumming enzymes (e.g. pectinases and xylanases), Bacillus subtilis strain 168 can also produce cellulase which brings irreversible damage to ramie fibers. In this study, a strain deficient in cellulase was constructed by insertional inactivation of gene eglS to make it suitable for microbial degumming. Results showed that the cellulase activity was not detected in strain ΔeglS, after 25 h of incubation with strain ΔeglS, the weight loss of the ramie fibers was 20.20 %. Though the residual gum content of ramie fibers obtained in microbial degumming process was 19.67 %, microbial intervention followed by diluted alkaline solution treatment showed lower residual gum content (2.73 %) than chemical degumming (2.96 %). The single fiber breaking strength value of ramie fibers from bio-chemical degumming process reached 27.19 cN, which was the highest of three samples. Moreover, bio-chemical degumming process resulted in a 75 % decrease of alkali dosage as compared with chemical degumming process. This study provides a suggestion to genetically modify wild-type strain in order to protect ramie fibers from cellulase damage, and indicates a sustainable alternative for the traditional chemical degumming in the textile industry.

      • DNA Repair Gene Polymorphisms Do Not Predict Response to Radiotherapy-Based Multimodality Treatment of Patients with Rectal Cancer: a Meta-analysis

        Background: A number of association studies have been carried out to investigate the relationship between genetic polymorphisms in DNA repair genes and response to radiotherapy-based multimodality treatment of patients with rectal cancer. However, their conclusions were inconsistent. The objective of the present study was to assess the role of DNA repair gene genetic polymorphisms in predicting genetic biomarkers of the response in rectal cancer patients treated with neoadjuvant chemoradiation. Materials and Methods: Studies were retrieved by searching the PubMed database, Cochrane Library, Embase, and ISI Web of Knowledge. We conducted a meta-analysis to evaluate the association between genetic polymorphisms and the response in rectal cancer treated with neoadjuvant chemoradiation by checking odds ratios (ORs) and 95% confidence intervals (CIs). Results: Data were extracted from 5 clinical studies for this meta-analysis. The results showed that XRCC1 RS25487, XRCC1 RS179978, XRCC3 RS861539, ERCC1 RS11615 and ERCC2 RS13181 were not associated with the response in the radiotherapy-based multimodality treatment of patients with rectal cancer (p>0.05). Conclusions: This study shows that DNA repair gene common genetic polymorphisms are not significantly correlated with the radiotherapy-based multimodality treatment in rectal cancer patients.

        Causal Relations between Exposome and Stroke: A Mendelian Randomization Study

        Background and Purpose To explore the causal relationships of elements of the exposome with ischemic stroke and its subtypes at the omics level and to provide evidence for stroke prevention. Methods We conducted a Mendelian randomization study between exposure and any ischemic stroke (AIS) and its subtypes (large-artery atherosclerotic disease [LAD], cardioembolic stroke [CE], and small vessel disease [SVD]). The exposure dataset was the UK Biobank involving 361,194 subjects, and the outcome dataset was the MEGASTROKE consortium including 52,000 participants. Results We found that higher blood pressure (BP) (systolic BP: odds ratio [OR], 1.02; 95% confidence interval [CI], 1.01 to 1.04; diastolic BP: OR, 1.03; 95% CI, 1.01 to 1.05; pulse pressure: OR, 1.03; 95% CI, 1.00 to 1.06), atrial fibrillation (OR, 1.18; 95% CI, 1.13 to 1.25), and diabetes (OR, 1.13; 95% CI, 1.07 to 1.18) were significantly associated with ischemic stroke. Importantly, higher education (OR, 0.69; 95% CI, 0.60 to 0.79) decreased the risk of ischemic stroke. Higher systolic BP (OR, 1.06; 95% CI, 1.02 to 1.10), pulse pressure (OR, 1.08; 95% CI, 1.02 to 1.14), diabetes (OR, 1.28; 95% CI, 1.13 to 1.45), and coronary artery disease (OR, 1.58; 95% CI, 1.25 to 2.00) could cause LAD. Atrial fibrillation could cause CE (OR, 1.90; 95% CI, 1.71 to 2.11). For SVD, higher systolic BP (OR, 1.04; 95% CI, 1.00 to 1.07), diastolic BP (OR, 1.06; 95% CI, 1.01 to 1.12), and diabetes (OR, 1.22; 95% CI, 1.10 to 1.36) were causal factors. Conclusions The study revealed elements of the exposome causally linked to ischemic stroke and its subtypes, including conventional causal risk factors and novel protective factors such as higher education.

      • A Tracking Control Method for a PPR Underactuated Manipulator

        A nonlinear output feedback controller is developed for an Underactuated manipulator in this paper. The PPR planar Underactuated manipulator consists of two active joints and one passive joint. The dynamic constraint on the free link is 2nd-order non-holonomic. Firstly, the motion equations are transformed into a special form using some global coordinate and input transformations. Secondly, a time-varying feedback controller is developed using Lyapunov’s direct method and an extension of the current popular backstepping technique. The control method is able to force the end-effector of a three DOF planar underactuated manipulator to globally asymptotically track a reference trajectory. And the stability of the control method is proved. Simulation results are provided to demonstrate the effectiveness of the proposed control laws.
