Endothelium, particularly pulmonary endothelium, is predisposed to injury by reactive oxygen species (ROS) and their derivatives. Heme oxygenase (HO) has been demonstrated to provide cytoprotective effects in models of oxidant-induced cellular and tissue injuries. In the present study, we investigated the effects of YS 49 against oxidant [tert-butylhydroperoxide (TBH)]-induced injury using cultured sheep pulmonary artery endothelial cells (SPAECs). The viability of SPAECs was determined by quantifying reduction of a fluorogenic indicator Alamar blue. We found that TBH decreased cell viability in a timeand concentration-dependent manner. YS 49 concentration- and time-dependently increased HO-1 induction on SPAECs. As expected, YS 49 significantly decreased the TBH-induced cellular injury. In the presence of zinc protophorphyrin, HO-1 inhibitor, effect of YS 49 was significantly inhibited, indicating that HO-1 plays a protective role for YS 49. Furthermore, YS 49 showed free radical scavenging activity as evidenced by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and inhibition of lipid peroxidation. However, YS 49 did not inhibit apoptosis induced by lipopolysaccharide (LPS) in SPAECs. Taken together, HO-1 induction along with strong antioxidant action of YS 49 may be responsible for inhibition of TBH-induced injury in SPAECs.

<P> Tetrahydroisoquinoline (THI) alkaloids can be considered as cyclized derivatives of simple phenylethylamines. Many of them, especially with 6,7-disubstitution, demonstrate a relatively high affinity for catecholamines. Present study examines the pharmacological action of limited series of THI, using rats isolated atria and aorta. In addition, a [<SUP>3</SUP>H] prazosin displacement binding study with THI compounds was performed, using rat brain homogenates to investigate whether these probes have -adrenoceptor affinity. We also compared the vascular relaxation potency of these probes with dobutamine. YS 49, YS 51, higenamine and dobutamine, concentration-dependently, relaxed endothelium-denuded rat thoracic aorta precontracted with phenylephrine (PE, 0.1 ㄍM) in which pEC<SUB>50</SUB> were 5.56⁑0.32 and 5.55⁑0.21, 5.99⁑1.16 and 5.57⁑0.34, respectively. These probes except higenamine also relaxed KCl (65.4 mM)-contracted aorta. In isolated rat atria, all THIs and dobutamine increased heart rate and contractile force. In the presence of propranolol, the concentration response curves of YS 49 and YS 51 shifted to the right and resulted in pA<SUB>2</SUB> values of 8.07⁑0.84 and 7.93⁑0.11, respectively. The slope of each compound was not deviated from unity, indicating that these chemicals are highly competitive at the cardiac -adrenoceptors. YS 49, YS 51, and higenamine showed -adrenoceptor affinity in rat brain, in which the dissociation constant (K<SUB>i</SUB>) was 2.75, 2.81, and 1.02 ㄍM, respectively. It is concluded, therefore, that THI alkaloids have weak affinity to <SUB>1</SUB>-adrenoceptors in rat aorta and brain, respectively, while these probes show relatively high affinity for cardiac -adrenoceptors. Thus, these chemicals may be useful in the treatment of congestive heart failure.

<P>Human demineralized bone matrix (DBM) containing bone morphogenetic proteins (BMPs) is naturally biocompatible and can be remodeled by patients’ own bone. The major shortcoming of many of the currently used DBM gel formulations is that they have a tendency to flow, particularly if there is continuous bleeding at the application site. In this study, the physicochemical properties of human DBM were examined to improve the efficiency of DBM formulations. DBM remarkably showed higher water absorption than nondemineralized bone powder after 150 min. Hydroxyl groups in DBM appeared in fourier transform infrared analysis, although hydroxyl band in nondemineralized bone powder was not observed. The results suggested that hydrogels such as CMC, hyaluronic acid, or poloxamer as carriers can be applied for injectable DBM products, such as gel or putty types.</P>

Saikosaponin productivity was examined in a Bupleurum falcatum L. BFHR2 hairy root culture in response to changes in the sucrose content $(2{\sim}8%)$, nitrogen content $(0{\sim}250mM\;NH_4NO_3)$, phosphate content $(0{\sim}12mM\;NaH_2PO_4)$, and the potassium content $(0{\sim}87.2 mM\; KCl)$ of the culture media. We found that the conditions for maximal saikosaponin production differed from those for optimal root growth. Highest saikosaponin yield was achieved for 8% sucrose, 62mM $NH_4NO_3$ 1.2 mM $NaH_2PO_4$, and 0.5mM KCl.

The basic and main nutritive ingredients of two temperature (Anguilla japonica and A. rostrata) and two tropical ( A. bicolor pacifica and A. marmorata) fresh water eel species that are farmed domestically were evaluated. With exception of A. rostrata, eels cultured at the same farm were used for analysis. The contents of crude protein were in the order A. marmorata (17.7%)> A. rostrata (17.5%)> A. bicolor pacifica (17.4%)> A. japonica (15.8%) and the contents of crude lipids were A. japonica (21.5%)> A. rostrata (15.4%)> A. bicolor pacifica (10.5%)> A. marmorata (8.9%). These values differed significantly even among the three species of eel farmed under identical culture conditions. In comparison, all four species of eel showed similar pattern in overall amino acid composition, although slight differences in the compositions of some amino acids were observed. The fatty acid compositions of muscle tissues were notably different among four species of eel, especially between the tropical and temperature eels. In a taste-test of the meat of the four eel species, which considered taste, flavor and texture, the overall preference was in the order A. japonica, A. marmorata, A. bicolor pacifica and A. rostrata.

Context: Based on fractional photothermolysis, fractional lasers have been developed to overcome the long downtime and possible complications of the traditional skin resurfacing technique, such as the pinhole method, which involves makin g multiple small holes using a carbon dioxide laser. Aims: As an extension of the pinhole method, we here introduce a fractional resurfacing technique that can deeply penetrate tissue to completely eliminate the lesion, while confining the treatment area to multiple lines or segments of various shapes in order to reduce complications. Settings and Design: The fractional resurfacing technique was used instead of surgery to treat small masses such as congenital pigmented nevi and nevi sebaceous, in addition, telangiectasia and cafe au lait macules. Treatment was performed using the Er:YAG laser and carbon dioxide laser. In treating our patients with the fractional resurfacing technique, we made line- or donut-shape holes surrounded by untreated area. The width of the treatment area was approximately 1 to 2 mm, while the depth depended on the depth of the lesion. Each lesion was treated in multiple sessions with 1-2-month intervals. Results: Patient``s lesions were successfully treated with this technique and there was no recurrence during the follow-up period. Conclusion: The fractional resurfacing technique can be used to completely remove a variety of lesions with a minimal downtime and few complications.
