        In order to study the roole of residue in the active site of glutathione S-transferase(GST), Arg13 residue in human GST P 1-1 was replaced with alanine, leucine or lysine by site-directed mutagenesis to obtain mutants R13A, R13L and R13K. These mutants were expressed in Escherichia coli and purified to electrophoretic homogeneity by affinity chromatography on immobilized GSH. The specific activities were determined by measuring the initial rates of the enzymes-catalyzed conjugation of GSH towards electrophilic substrates. Our results suggest that Arg13 in human GST P1-1 contributes to the binding of electroophilic substrate, but it is not needed for the glutathione peroxidase activity and the steroid isomerase activity of human glutathione S-transferase p1-1.

      • Aspergillus niger 글루코오스 산화효소의 유도 발현

        Glucose oxidase catalyzes the oxidation of β-D-glucose to gluconic acid. It is necessary to be obtained maximum glucose oxidase activity because the enzyme is used commercially for various applications, in particular extracelluar glucose oxidase. For high activity of extracellular glucose oxidase, we used the enzyme from Aspergillus niger and YEP medium. Extracellular glucose oxidase activity as high as 7.5Uml^-1 was obtained with sucrose as carbon source and peptone as nitrogen source. The result of testing a various of medium condition was obtainable the highest enzyme activity in condition of culture medium of 1% sucrose(w/v), 2% peptone(w/v) and 1% yeast extract(w/v) at 25℃. Our work indicate an economically attractive process for enzyme production. In addition Aspergillus niger glucose oxidase was purified by DEAE-Sephacel chromatography. The activity of the enzyme proceeded at pH 5.5 and the effective substrate of the enzyme was only glucose.

        물리치료사와 물리치료과 학생의 성별에 따른 HIV/AIDS에 대한 지식과 태도

        The purpose of this descriptive study is to investigate the general knowledge and attitude about HIV/AIDS among gender in physical therapists and physical therapy students. AIDS a clinical syndrome that involves progressive immune deficiency and consequent development of opportunistic infection, tumors, neurologic disease and systemic wasting with chronic trends. Many medical specialists are afraid of AIDS patients because they have a little information of AIDS and fear of HIV infection from AIDS patients at work site. This study surveyed 649 physical therapists and physical therapy students by means of a self-administered questionnaire. This study was carried out from April, 2003 to October, 2003 for collecting data. The findings of this study can be summarized as follows. 1) The male and female physical therapist and physical therapy students who have an experience of educational program for HIV/AIDS show higher general knowledge about HIV/AIDS. 2) The male physical therapist and physical therapy students were conscious of the need for physical therapeutic intervention for the complication of HIV/AIDS. 3) The male who has higher general knowledge about HIV/AIDS are conscious of the need for educational program about HIV/AIDS. 4) The male and female want to have the choice to accept or reject the HIV/AIDS patients for treatment. 5) The knowledge of transmission mode in sexual intercourse of HIV/AIDS influence the attitude of male and female to patients with HIV/AIDS. 6) The knowledge of prevention of HIV/AIDS influence the attitude of male and female to patients with HIV/AIDS. 7) The knowledge of regarding symptom of HIV/AIDS influence the attitude of female to patients with HIV/AIDS. 8) The knowledge of general transmission mode of HIV/AIDS influence the attitude of male and female to patients with HIV/AIDS. 9) The knowledge of transmission risk factors in work site of HIV/AIDS influence the attitude of male and female to patients with HIV/AIDS. 10) The knowledge of needs for prevention in work site of HIV/AIDS influence the attitude of male and female to patients with HIV/AIDS. In conclusion, male and female with more knowledge of HIV/AIDS could positively treat HIV/AIDS patients.

        정중운동신경과 척골운동신경의 전기생리학적 연구

        The determination of peripheral nerve conduction velocity is and important part to electrodiagnosis. Its value as neurophysiologic investigative procedure has been known for many years but normal value of median and ulnar motor nerve was poorly reported in Korea. To evaluate of median and ulnar motor nerve terminal latency, amplitude of CMAP(compound muscle action potential), conduction velocity and F-wave latency for obtain clinically useful reference value. 71 normal volunteers(age,19-65 years;142 hands) examined who has no history of peripheral neuropathy, diabetic mellitus, chronic renal failure, endocrine disorders, anti-cancer medicine, anti-tubercle medicine, alcoholism, trauma, radiculopathy. Nicolet Viking Ⅱ was use for detected terminal latency, amplitude of CMAP, conduction velocity and F-wave latency of median and ulnar motor nerve. Data analysis was performed using SPSS. Descriptive analysis was used for obtain mean and standard deviation, independent t-test was used to compare between Rt and Lt side also compare between different in genders. The results are summarized as follows: 1. Median motor nerve terminal latency was right 3.00ms, left 2.99ms and there was no significantly difference between right and left side and genders. 2. Median motor nerve amplitude of CMAP was right 17.26mV, left 17.50mV and there was no significantly differences between right and left side and genders. 3. Median motor nerve conduction velocity was right 57.89m/sec, left 58.03m/sec and there was no significantly difference between right and left side and genders. 4. Median motor nerve F-wave latency was right 25.74ms, left 25.59ms and there was significantly differences between genders. 5. Ulnar motor nerve terminal latency was right 2.38ms, left 2.45ms and there was significantly differences between right and left side. 6. Ulnar motor nerve amplitude of CMAP was right 15.99mV, left 16.02mV and there was no significantly differences between right and left side and genders. 7. Ulnar motor nerve conduction velocity was right 60.35m/sec, left 59.73/sec and there was no significantly differences between right and left side and genders. 8. Ulnar motor nerve F-wave latency was right 25.53ms, left 25.57ms and there was significantly differences between genders.

        전경골근 등척성 수축에 의한 경직성 뇌졸중 환자의 비복근 α-운동 신경원 흥분 변화

        Spasticity has been defined as "a motor disorder characterized by a velocity-dependent increased in tonic stretch reflexes with exaggerated tendon jerks, resulting from hyperexcitability of the stretch reflex, as one components of the upper motorneuron syndrome". Spasticity is one of the common symptoms of stroke patients and frequently interferes with the motor functions such as gait, posture and activities of daily living. Therefore, its management is becoming a major issue in physical therapy. The purpose of this study was to determined the effects of reciprocal inhibition by isometric contraction of pre-tibia muscle on spasticity in hemiplegic patients through Hoffmann reflex. The subjects were consisted 45 patients who had hemiplegia due to stroke. All subjects randomly assigned to 3 group: manual reciprocal inhibition program group(manual group), neuromuscular electrical stimulation group(NMES group) and control group. The manual group received voluntary isometric contraction of pre-tibia muscle. The NMES group received neuromuscular electrical stimulation on tibialis anterior. The control group was not received any therapeutic intervention. Before and after experiments, Hoffmann reflex, M-wave and Modified Ashworth scale was measure in all patients. The data of 30 patients who complete experimental course were statistically analysed. Modified Ashworth scale were significantly decreased after experiment in manual group(p<.01). The Hmax/Mmax ratios were significantly decreased after experiment in manual group(p<.01). There were no statistical difference between pre-test and post-test with modified Ashworth scal in NMES group(p>.01). There were no statistical difference between pre-test and post-test with Hmax/Mmax ratios in NMES group(p>.01). There were no statistical difference between pre-test and post-test with modified Ashworth scale in control group(p>.01). There were no statistical difference between pre-test and post-test with Hmax/Mmax ratios in control group(p>.01). The present results revealed that reciprocal inhibition which produced by voluntary isometric contraction of pre-tibia muscle can be reduce spasticity of gastrocnemius. Therefore, reciprocal inhibition is useful improve functional activities in hemiplegic patient. Further study should be done to analyse the effects of intervention duration of reciprocal inhibition, appropriate muscle contraction, optimal time to apply the reciprocal inhibition in more long period.

        척골 지단 신경의 전기생리학적 연구

        The ulnar nerve extends down the arm, across the elbow, and into the hand. It provides sensation to the little and ring fingers and activates many of the small muscles in the hand. The determination of peripheral nerve conduction velocity is an important part of ulnar nerve evaluation. The electrodiagnostic value as neurophysiologic investigative procedure has been known for many years but normal value of digital nerve was not reported in korea. The purpose of this investigation was to measure the digital nerve conduction velocity of ulnar nerve for obtain clinically useful reference value and compare difference in each fingers and then compare with the other countries. 71 normal Korean volunteers (age, 19-65 years; 142 hands) examined who has no history of peripheral neuropathy, diabetic mellitus, chronic renal failure, endocrinedisorders, anti-cancer medicine, anti-tubercle medicine, alcoholism, trauma, radiculopathy. Nicolet Viking Ⅱ(EMG machine) was use for detected conduction velocity and amplitude of digital nerve in ulnar nerve. Data analysis was performed using SPSS. Descriptive analysis was used for obtain mean and standard deviation and independent t-test was used to compare with ring and little finger. Conduction velocity of the right ring finger was 57.44m/sec and little finger was 55.32msec. The left ring finger was 55.55msec and little finger was 54.11msec. Amplitude of the right ring finger was 30.28µV and little finger was 48.36µV. The left ring finger was 30.67µV and little finger was 57.76µV. There were significantly difference between ring and little in amplitude (p<.05) but there were no statistically difference between conduction velocity of ring and little finger (p>.05). The amplitude of little finger are greater than ring finger. The present results revealed that electodiagnosis can easily perform in little finger for digital nerve of ulnar nerve study.

        We performed the cross-cultural adaptation process directed at the translation into Korean of the Pain Visual Analogue Scale, the Self-Report of Improvement and the Beacke Physical Activity Questionnaire for Older Adults. This project will help ensure that the Korean versions of these measures retain their meaning and measurement properties when applied to the Korean language and culture. This will help reduce measurement error in the RCT. This research should be of value to other researchers who wish to study older adults with rheumatological conditions.
