      • DIF와 시토키닌의 상호작용이 포도의 기내생장에 끼치는 영향

        주야간 온도 변화(DIF)와 시토키닌의 상호작용이 포도의 기내생장에 끼치는 영향을 구명하여 포도의 기내 대량증식체계를 확립하기 위하여 유럽종 양조용 품종 'Cabernet Sauvignon'과 생식용 구미잡종 'Campbell Early' 포도를 공시하여 metal halide lamp와 형광등으로 210μmolㆍm-2ㆍs-1의 광량자속밀도, 상대습도 70±5%, 16시간 광주기로 조절된 생육상에서 일련의 시험을 수행하였다. 주야간 온도는 25/25, 25/18, 18/25℃로 설정하였고, 시토키닌은 zeatin, kinetin, 2ip, BA및 TDZ를 단용 처리하였다. 모든 시험은 3반복으로 실시하였으며, 4주 후 생장과 분지, 그리고 생체중을 조사하였다. Cabernet Sauvignon 에서는 DIF처리와 상관없이 대조구가 신초의 생장이 가장 좋았는데 -7DIF에서 생장이 전반적으로 나빴다. 그리고 BA와 TDZ처리는 DIF에 영향을 크게 받지 않은 것으로 나타났다. 0DIF는 시토키닌의 농도가 높을수록 생장이 다소 좋은 경향을 나타냈고, kinetin 10mM은 87mm로 대조구를 제외한 시토키닌 처리구 가운데에서 가장 높았다. Campbell Early 에서 -7DIF에서는 Zeatin 5mM처리가 162mm로 대조구의 112mm 보다 생장이 좋았으며 +7DIF에서 kinetin 5mM처리가 110mm로 가장 좋았다. 0DIF에서는 Zeatin 5mM 처리가 162mm로 대조구보다 약 2배 정도 높은 생장을 보였다. Cabernet Sauvignon에서 분지수는 DIF처리와 관계없이 대조구는 1개가 분지 되었으며 모든 DIF처리에서 신초의 생장이 적었던 BA와 TDZ처리에서 분지가 많았다. 0, +7DIF에서는 BA 10mM처리가 약 4개로 나타났으며 -7DIF처리는 TDZ 5mM처리에서 가장 많았다. Zeatin, 2ip와 kinetin 처리는 분지가 되지 않은 것으로 나타났다. Campbell Early 에서도 같은 결과였다. 총절편체수는 Cabernet Sauvignon에서는 모든 DIF처리에서 BA 10mM 처리가 가장 많았다. 신초생장이 좋았던 CT와 Zeatin, 2ip, kinetin 보다 생장은 적었지만 분지수가 많았던 BA와 TDZ처리에서 총절편체수가 많은 것으로 나타났다. Campbell Early 역시 Cabernet Sauvignon과 비슷한 경향을 나타냈으며, 0DIF처리가 -, +7DIF처리보다는 절편체수가 많은 것으로 나타났다. Cabernet Sauvignon에서는 -7DIF처리에서 대체로 200mg 이하로 적었으며 시토키닌의 종류에 차이도 크지 않았다. 0DIF처리는 2ip 10mM 처리가 가장 높았으며, +7DIF에서는 TDZ 5mM이 대조구와 비슷하게 380mg을 나타냈다. Campbell Early는 +7DIF처리에서 BA 5mM을 제외한 나머지 모든 처리구에서 대조구보다 높게 나타났고 2ip 10mM이 410mg으로 가장 높았다. 0DIF처리는 Zeatin 5와 2ip 10mM처리가 900, 840mg으로 대조구의 약 8배에 달했으며, kinetin 10mM처리가 가장 낮게 나타났다. -7DIF처리에서 모든 처리구가 대조구보다 높았으나 다른 DIF처리보다는 낮았다. Shoot growth, number of shoots and fresh weight of Vitis vinifera L.'Cabernet Sauvignon' and Vitis labruscana B. 'Campbell Early' explants under three DIF levels, -7, 0 and +7, 210μmolㆍm-2ㆍs-1 provided by metal halide and fluorescent lamps, under 16hㆍd-1 photoperiod were studied. Basal MS media were supplemented with 5 and 10 mM BA, TDZ, Zeatin, 2ip, kinetin, respectively. In all media, 2.1gㆍL-1 gelite and 3% sucrose were used and cultured for 4 weeks. Shoot growth was significantly increased by control regardless of DIF in Cabernet Sauvignon and was suppressed by -7DIF. Effects of BA and TDZ were not affected by DIF. 0DIF enhanced the growth with increasing cytokinin level. Kinetin at 10 mM was most effective for shoot growth with 87mm among cytokinins used except for control. Zeatin and kinetin each at 5 mM significantly increased shoot growth to respective 162 and 110mm. In 0DIF, double increase of 162 mm was obtained by 5 mM zeatin. In control, no further shoot proliferation was observed in Cabernet Sauvignon regardless of DIF. In 0 and +7DIF, 4 shoots were observed with 10 mM BA. Zeatin, 2ip, and kinetin resulted in poor shoot proliferation. BA at 10 mM resulted in profuse branching in all DIFs.

        사회 변화에 따른 부름말의 용법 변화 - 친족관계말의 확대 현상을 중심으로 -

        국어 부름말은 말할이와 들을이의 사회적 관계에 따라 달리 실현된다. 그러나 요즘 사용되는 부름말의 양상을 보면 부름말이 말할이와 들을이의 사회적 관계와 일치하지 않는다. 이는 부름말의 용법이 변했음을 의미한다. 부름말은 대인 관계와 관련한 것이므로 상대를 대하는 태도와 관련이 있다. 상대를 대하는 태도에는 개인이 속한 사회나 집단이 지향하는 가치인 사회 규범이 반영되어 있다. 이러한 사회 규범은 시대와 더불어 변한다. 개인이 속한 사회에서 지향하는 가치가 변하면 사회 규범이 반영된 부름말의 용법도 변한다. 따라서 부름말의 용법 변화는 사회 규범의 변화와 관련지어 기술해야 한다. 산업화에 따라 남과 더불어 살게 되면서 상대를 친족인 자와 친족이 아닌 자로 구별하여 대하는 사회 규범은 약화한다. 이러한 사회 규범의 변화는 부름말에도 영향을 주어 남인 자에게도 친족어를 부름말로 한다. 산업화는 가정에도 변화를 주어 과거 대가족이던 가정은 핵가족으로 점차 변하였다. 핵가족화되면서 남의 부모에게도 자신의 부모처럼 친밀하게 대하고자 하는 말할이의 의도가 반영되어 남의 부모에게도 ‘아버지’, ‘어머니’를 부름말로 한다. 남녀를 구별하던 사회는 점차 남녀를 구별하지 않는 것으로 변하고 있다. 이러한 사회 규범의 변화는 부름말에도 반영이 되어 처부모에게도 ‘아버지’, ‘어머니’를 부름말로 한다. 남녀를 구별하던 사회는 점차 남녀를 구별하지 않는 것으로 변하고 있다. 이러한 사회 규범의 변화는 부름말에도 반영이 되어 처부모에게도 ‘아버지’, ‘어머니’를 부름말로 한다. In the Korean language, nouns of address can vary depending on the social relationships between the speaker and the addressee. However, the recent trend shows that there is a discrepancy between the nouns of address and the speaker"s and the addressee"s social relationships. It suggests that the usage of address words has changed. Since nouns of address are closely related to interpersonal relations, they are shaped by how individuals treat others. Moreover, the way individuals treat others reflects the norms of the society or group they belong to. Such norms evolve over time. When the social values change, the usage of address words changes accordingly, given that these words reflect such norms and values of society in which individuals are situated. Therefore, the changes in how the nouns of address are used in society should be understood in conjunction with the changes in social norms. Going through industrialization, it has become common to reside with others apart from one’s own family. This change has made it less relevant to distinguish people one has a relationship with, in terms of blood kinships nowadays. This norm change has influenced the way we address others accordingly. It has become common to call non-family members by using nouns that describe family members literally. Industrialization has transformed the family structure as well, gradually from extended family primarily in the past to nuclear family these days. Nuclearization of family has motivated individuals to treat others’ parents as closely as their own parents, and thus many people call them by using words such as ‘father’ and ‘mother.’ In addition, Korean society, which used to be strict about distinguishing gender in language, has changed to be less particular about gender differentiation over time. This trend has also been reflected in the use of address words such that a man addresses his parents-in-law as ‘father’ and ‘mother.’

        Purpose: This study was performed to identify the current status of organization and education for evidence-based nursing (EBN) among acute care hospitals in Korea. Methods: Using a questionnaire, the data were collected from 60 acute general hospitals selected conveniently from October to November in 2016. Results: The response rate was 83.3% and the mean number of beds for responded hospitals were 977.83. Responded hospitals appeared to perceive the importance of EBN, though approximately 50% of hospitals showed insufficient status for EBN. EBN project implemented in accordance with institutional support. EBN education courses have operated within hospitals and also have attended at outside courses. Conclusion: The current status of EBN organization and education have identified and it is needed to monitor continuously to activate and expand EBN in clinical settings.

        Purpose: This study aims to identify current status of evidence-based nursing education to nursing students and strategies for dissemination. Methods: For investigation of current education status of evidence-based nursing, a searching for websites of 204 nursing colleges in Korea. And a focus-group interview was performed. Ten participants were faculties working at college of nursing. Interview data were analyzed using qualitative content analysis. Results: Nineteen (9.3%) colleges were running evidence-based nursing (EBN) course in the undergraduate course and 29 colleges opened EBN course in the graduate school. Five topics regarding experience on teaching EBN were as follows, 1) there are need to teaching EBN in nursing colleges 2) EBN education leads to improvement in professional perception and research, but low level of feed-back also indicated. 3) Barriers were lack of faculty ability, difficulty in course management, limitation of resource and gaps in practice. 4) Facilitators were changes in clinical nursing practice, teaching faculty and developing teaching program. 5) Educational strategies for dissemination were changes on external evaluation and strengthening of academic society activities. Conclusion: EBN education for nursing students are effective in improving critical thinking and nursing research. Developing strategies for dissemination of EBN to nursing students are needed.

      • Effect of microwave treatment on the food processing-related quality characteristics and antioxidant activity of red bean (Vigna angularis var. nipponensis)

        The application of the heat treatment method as one of various food processing technologies could contribute to the utilization of red beans. Microwave (MW) treatment is known to be a simple and efficient method, and to affect antioxidant properties of grains. The objective of this study was to characterize the food processing-related quality of red beans treated at different times of microwave. After microwave-treated, the a and b values decreased and the moisture content also decreased from 13.9 to 10.8%. Scanning electron microscope (SEM) images revealed that there was space between the seed coat and embryo due to loss of moisture. In hydration properties, the equilibrium moisture content of the control was 142.3%, but decreased to 122.2%, and 110.4%, respectively in the microwave-treated seeds. The DPPH and ABTS radical scavenging capacity of the control were 8.4 and 17.6 mg VCE/g, respectively, but increased to 9.3 and 18.9 mg VCE/g in the microwave treated. The TFC also changed from 5.0 to 6.2 mg catechin/g, but there was no significant difference in the TPC. Our results suggested that microwave treatment was effective to enhance the antioxidant activity of red beans.

        Identification of Coronavirus Isolated from a Patient in Korea with COVID-19

        Objectives Following reports of patients with unexplained pneumonia at the end of December 2019 in Wuhan, China, the causative agent was identified as coronavirus (SARS-CoV-2), and the 2019 novel coronavirus disease was named COVID-19 by the World Health Organization. Putative patients with COVID-19 have been identified in South Korea, and attempts have been made to isolate the pathogen from these patients. Methods Upper and lower respiratory tract secretion samples from putative patients with COVID-19 were inoculated onto cells to isolate the virus. Full genome sequencing and electron microscopy were used to identify the virus. Results The virus replicated in Vero cells and cytopathic effects were observed. Full genome sequencing showed that the virus genome exhibited sequence homology of more than 99.9% with SARS-CoV-2 which was isolated from patients from other countries, for instance China. Sequence homology of SARS-CoV-2 with SARS-CoV, and MERS-CoV was 77.5% and 50%, respectively. Coronavirus-specific morphology was observed by electron microscopy in virus-infected Vero cells. Conclusion SARS-CoV-2 was isolated from putative patients with unexplained pneumonia and intermittent coughing and fever. The isolated virus was named BetaCoV/Korea/KCDC03/2020.
