      • Association Between Single Nucleotide Polymorphisms in the XRCC1 Gene and Susceptibility to Prostate Cancer in Chinese Men

        Background: Prostate cancer (Pca) is one of the most common complex and polygenic diseases in men. The X-ray repair complementing group 1 gene (XRCC1) is an important candidate in the pathogenesis of Pca. The purpose of this study was to evaluate the association between single nucleotide polymorphisms in the XRCC1 gene and susceptibility to Pca. Materials and Methods: XRCC1 gene polymorphisms and associations with susceptibility to Pca were investigated in 193 prostate patients and 188 cancer-free Chinese men. Results: The c.910A>G variant in the exon9 of XRCC1 gene could be detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and DNA sequencing methods. Significantly increased susceptibility to prostate cancer was noted in the homozygote comparison (GG versus AA: OR=2.95, 95% CI 1.46-5.42, ${\chi}^2$=12.36, P=0.001), heterozygote comparison (AG versus AA: OR=1.76, 95% CI 1.12-2.51, ${\chi}^2$=4.04, P=0.045), dominant model (GG/AG versus AA: OR=1.93, 95% CI 1.19-2.97, ${\chi}^2$=9.12, P=0.003), recessive model (GG versus AG+AA: OR=2.17, 95% CI 1.33-4.06, ${\chi}^2$=8.86, P=0.003) and with allele contrast (G versus A: OR=1.89, 95% CI 1.56-2.42, ${\chi}^2$=14.67, P<0.000). Conclusions: These findings suggest that the c.910A>G polymorphism of the XRCC1 gene is associated with susceptibility to Pca in Chinese men, the G-allele conferring higher risk.

      • Human Papillomavirus Infection and Prognostic Predictors in Patients with Oropharyngeal Squamous Cell Carcinoma

        This study focused on infection rates and subtypes of human papillomavirus (HPV) in patients with oropharyngeal squamous cell carcinoma (OSCC), and the relationship between HPV status and prognosis of the disease. We evaluated sixty-six OSCC patients who met the enrollment criteria during the period from January 1999 to December 2009. The presence or absence of oncogenic HPV types in tumors was determined using the SPF10 LiPA25 assay. Overall survival (OS) and disease specific survival (DSS) for HPV positive and HPV negative patients were estimated using Kaplan-Meier analysis. The Cox regression model was applied for multivariate analysis. HPV-DNA was detected in 11(16.7%) of all specimens. Among them, 7 were type HPV-16, while other types were HPV-16/11, HPV-35, HPV-58/52, and HPV-33/52/54. Patients with HPV positive tumors were more likely to be female, non-smokers and non-drinkers (p=0.002, 0.001 and 0.001, respectively). After a median follow-up of 24.5 months, patients with HPV positive tumors had significantly better overall survival (HR=0.106[95%CI=0.014-0.787], p=0.016,) and disease specific survival (HR=0.121[95%CI=0.016-0.906], p=0.030). Patients with HPV positive OSCC have significantly better prognosis than patients with HPV negative tumors. HPV infection is an independent prognostic factor.

        Morphological defense of the egg mass of Spodoptera frugiperda (Lepidoptera: Noctuidae) affects parasitic capacity and alters behaviors of egg parasitoid wasps

        The fall armyworm, Spodoptera frugiperda, is a dangerous pest of cereals originating from the tropical and sub tropical parts of the Americas. It has invaded over a hundred countries and is spreading rapidly throughout East Asia. Biological control programs are a “model strategy” for the control of invasive pests. Egg parasitoids Tele nomus remus and Trichogramma pretiosum are viewed as candidates for the control of S. frugiperda. The aim of this study was to evaluate the behavioral patterns and parasitism efficacy of Te. remus, Tr. pretiosum, and Tricho gramma dendrolimi on S. frugiperda egg masses with and without scales. The proportion of parasitism by Te. remus was significantly higher than that by Tr. dendrolimi and Tr. pretiosum. The proportion of parasitism by Tr. pre tiosum and Tr. dendrolimi on egg masses with scales was significantly lower than that on egg masses without scales. However, Te. remus had a similar proportion of parasitism on egg masses with and without scales. Residence time, oviposition time, oviposition frequency, risk of host being found, and risk of parasitism by Te. remus were significantly higher than the corresponding parameters of Tr. pretiosum and Tr. dendrolimi. Tr. pre tiosum females exhibited a higher residence time, oviposition time, oviposition frequency, risk of host being found, and risk of parasitism than those of Tr. dendrolimi females. In addition, Te. remus females often crept into the scale layer covering the egg masses, whereas Tr. dendrolimi and Tr. pretiosum females did not; they could only parasitize the eggs located on the periphery of the egg mass. Both Te. remus and Tr. pretiosum females had similar proportions of superparasitism, which were significantly higher than that of Tr. dendrolimi. Therefore, Te. remus is the dominant egg parasitoid of S. frugiperda and this has important implications for developing augmentative biological control strategies for S. frugiperda.

        Ethosomes, Binary Ethosomes and Transfersomes of Terbinafine Hydrochloride: A Comparative Study

        The aim of this study was to compare the skin permeation of ethosomes, binary ethosomes and transfersomes of Terbinafine Hydrochloride (TH) under non-occlusive conditions. These lipid vesicles were prepared and characterized for shape, size, zeta-potential and entrapment efficiency. Franz diffusion cells and confocal laser scanning microscopy (CLSM) were used for the percutaneous absorption studies. The quantity of drug in the skin from ethosomes, binary ethosomes (the weight ratio of ethanol to propylene glycol 7:3, ethanol-PG = 7:3, w/w), and transfersomes was 1.26, 1.51 (p <0.05), 1.56 (p <0.01) times higher than that of TH from traditional liposomes (control). The skin deposition of the applied dose (DD%) of TH from ethosomes, binary ethosomes, and transfersomes was 3.34 (p < 0.05), 9.88 (p < 0.01), 2.52 times higher than that of TH from control. The results of CLSM experiments showed that penetration depth and fluorescence intensity of Rhodamine B from binary ethosomes was much greater than that from ethosomes and transfersomes. These results indicated the binary ethosomes (ethanol-PG = 7:3, w/w) most effectively permitted drug penetration through skin; transfersomes made drug easiest to accumulate in the skin. Ethosomes improved drug delivery with greater improvement in skin permeation than improvement in skin deposition.

        An Experimental Study on the Correlation between Composite Lining Stiffness and Bearing Capacity in Soft Ground

        The composite lining is intended to be stiff and strong, based on the current design guidance, to carry the designated load alone in the long-term operation. However, according to the latest research, the temporary support is supposed to carry load even under the undesired circumstance of serious deterioration, and it is contentious whether thicker lining is favorable for tunnel durability or not. A series of model tests were conducted in this study, therefore, to develop the correlation between lining stiffness and tunnel durability. In the tests, the test box, consisting of surrounding earth, temporary support and lining, is considered as an integrated structure to carry the load transferred from the upper, through which composite lining is represented. Eight conditions were designed by changing the stiffness of the temporary support and the lining. The test results from experimental data analysis (e.g. failure process, lining displacement and internal force) show that the more flexible the lining is and the stiffer the temporary support is, the more durable the condition becomes At the same time, the imperfect contact between the temporary support and the lining greatly weakens the carrying capacity of the whole structure.

        ROOM-TEMPERATURE SOLID-STATE REACTION TO TREE-LIKE FRACTAL NANO-CdS AND THIRD-ORDER NONLINEAR OPTICAL PROPERTIES

        A novel tree-like nano-cadmium sulfide (CdS) with the fractal feature is synthesized by solid-state reaction at room temperature from complex precursor with aminotrimethylenephosphonic acid (ATMP) as ligand. The obtained sample is the crystalline cubic beta cadmium sulfide. The tree is composed of nanorods with an average diameter of ca. 95 nm and a length of up to 100–650 nm. The nanorods grow in the asymmetrical "Y" shape. The amount of ATMP plays an important role in the formation of fractal structure. Nonlinear optical (NLO) measurements by the Z-scan technique exhibit that the tree-like fractal nano-CdS has the third-order nonlinear optical properties of both NLO absorption and NLO refraction with self-focusing effect and the optical limiting performance.

        Preparation and In Vitro Release of Ramose Chitosan-Based-5-Fluorouracil Microspheres

        In order to construct a controlled release system of drugs and to reduce toxic side effects of 5-fluorouracil, the novel ramose chitosan-based-5-fluorouracil microspheres (CS-FU-MS) were prepared. Firstly, using chitosan (CS) as carriers and 5-fluorouracil (5-FU) as a model drug, ramose chitosan-based-5-fluorouracil (CS-FU) was efciently synthesized by chemical crosslinking method through microwave irradiation, drug loading was 10.6%; Secondly, CS-FU-MS were prepared by CS-FU self-assembled under the dialysis conditions and the free 5-FU was encapsulated further at the same time. The size dispersivity of particles is uniform, and the average diameter of the CS-FU-MS was $4{\mu}m$. The drug encapsulation efficiency was 76.1%, and the drug loading was increased to 26.22%. CS-FU-MS maintain the zero-order release time in PBS (pH = 7.4) and HCl/KCl (pH = 1.2) dialysis medium was 40h and 34h respectively, and the cumulative release were 58.89% and 79.33% in 182 h. The results showed that CS-FU-MS have excellent sustained release properties.

        Gut Microbiota Dysbiosis Correlates With Long COVID-19 at One-Year After Discharge

        Background: Long coronavirus disease 2019 (COVID-19) in recovered patients (RPs) is gradually recognized by more people. However, how long it will last and the underlining mechanism remains unclear. Methods: We conducted a prospective follow-up study to evaluate the long-term symptoms and clinical indices of RPs at one-year after discharge from Union Hospital, Wuhan, China between December 2020 to May 2021. We also performed the 16S rRNA sequencing of stool samples from RPs and healthy controls (HCs) and analyzed the correlation between the gut microbiota and long COVID-19. Results: In total, 187 RPs were enrolled, among them, 84 (44.9%) RPs reported long COVID-19 symptoms at one-year after discharge. The most common long-term symptoms were cardiopulmonary symptoms, including chest tightness after activity (39/187, 20.9%), palpitations on exercise (27/187, 14.4%), sputum (21/187, 11.2%), cough (15/187, 8.0%) and chest pain (13/187, 7.0%), followed by systemic symptoms including fatigue (34/187, 18.2%) and myalgia (20/187, 10.7%), and digestive symptoms including constipation (14/187, 7.5%), anorexia (13/187, 7.0%), and diarrhea (8/187, 4.3%). Sixty-six (35.9%) RPs presented either anxiety or depression (42/187 [22.8%] and 53/187 [28.8%] respectively), and the proportion of anxiety or depression in the long symptomatic group was significantly higher than that in the asymptomatic group (41/187 [50.6%] vs. 25/187 [24.3%]). Compared with the asymptomatic group, scores of all nine 36-Item Short Form General Health Survey domains were lower in the symptomatic group (all P < 0.05). One hundred thirty RPs and 32 HCs (non-severe acute respiratory syndrome coronavirus 2 infected subjects) performed fecal sample sequencing. Compared with HCs, symptomatic RPs had obvious gut microbiota dysbiosis including significantly reduced bacterial diversities and lower relative abundance of short-chain fatty acids (SCFAs)-producing salutary symbionts such as Eubacterium_hallii_group, Subdoligranulum, Ruminococcus, Dorea, Coprococcus, and Eubacterium_ventriosum_group. Meanwhile, the relative abundance of Eubacterium_hallii_group, Subdoligranulum, and Ruminococcus showed decreasing tendencies between HCs, the asymptomatic group, and the symptomatic group. Conclusion: This study demonstrated the presence of long COVID-19 which correlates with gut microbiota dysbiosis in RPs at one-year after discharge, indicating gut microbiota may play an important role in long COVID-19.

        JCAD deficiency attenuates activation of hepatic stellate cells and cholestatic fibrosis

        Background/Aims: Cholestatic liver diseases including primary biliary cholangitis (PBC) are associated with active hepatic fibrogenesis, which ultimately progresses to cirrhosis. Activated hepatic stellate cells (HSCs) are the main fibrogenic effectors in response to cholangiocyte damage. JCAD regulates cell proliferation and malignant transformation in nonalcoholic steatoheaptitis-associated hepatocellular carcinoma (NASH-HCC). However, its participation in cholestatic fibrosis has not been explored yet. Methods: Serial sections of liver tissue of PBC patients were stained with immunofluorescence. Hepatic fibrosis was induced by bile duct ligation (BDL) in wild-type (WT), global JCAD knockout mice (JCAD-KO) and HSC-specific JCAD knockout mice (HSC-JCAD-KO), and evaluated by histopathology and biochemical tests. In situ-activated HSCs isolated from BDL mice were used to determine effects of JCAD on HSC activation. Results: In consistence with staining of liver sections from PBC patients, immunofluorescent staining revealed that JCAD expression was identified in smooth muscle α-actin (α-SMA)-positive fibroblast-like cells and was significantly up-regulated in WT mice with BDL. JCAD deficiency remarkably ameliorated BDL-induced hepatic injury and fibrosis, as documented by liver hydroxyproline content, when compared to WT mice with BDL. Histopathologically, collagen deposition was dramatically reduced in both JCAD-KO and HSC-JCAD-KO mice compared to WT mice, as visualized by Trichrome staining and semi-quantitative scores. Moreover, JCAD deprivation significantly attenuated in situ HSC activation and reduced expression of fibrotic genes after BDL. Conclusions: JCAD deficiency effectively suppressed hepatic fibrosis induced by BDL in mice, and the underlying mechanisms are largely through suppressed Hippo-YAP signaling activity in HSCs.
