        Cytoskeletal changes during nuclear and cell division in the freshwater alga Zygnema cruciatum (Chlorophyta, Zygnematales)

        Cytoskeletal changes were observed during cell division of the green alga Zygnema cruciatum using flourescein isothiocynate (FITC)-conjugated phallacidin for F-actin staining and FITC-anti-α-tubulin for microtubule staining. Z. cruciatum was uninucleate with two star-shaped chloroplasts. Nuclear division and cell plate formation occurred prior to chloroplast division. Actin filaments appeared on the chromosome and nuclear surface during prophase, and the F-actin ring appeared as the cleavage furrow developed. FITC-phallacidin revealed that actin filaments were attached to the chromosomes during metaphase. The F-actin ring disappeared at late metaphase. At telophase, FITC-phallacidin staining of actin filaments disappeared. FITC-anti-α-tubulin staining revealed that microtubules were arranged beneath the protoplasm during interphase and then localized on the nuclear region at prophase, and that the mitotic spindle was formed during metaphase. The microtubules appeared between dividing chloroplasts. The results indicate that a coordination of actin filaments and microtubules might be necessary for nuclear division and chromosome movement in Z. cruciatum.

      • Improvement of saccharification process for bioethanol production from <i>Undaria</i> sp. by gamma irradiation

        <P><B>Abstract</B></P><P>Recently, many research works have reported on improvements to the saccharification process that increase bioethanol production from cellulosic materials. Gamma irradiation has been studied as an effective method for the depolymerization of complex polysaccharides. In this study, the effect of gamma irradiation on saccharification of <I>Undaria</I> biomass for bioethanol production was investigated. The <I>Undaria</I> biomass was irradiated at doses of 0, 10, 50, 100, 200 and 500kGy and then hydrolyzed using sulfuric acid. The effects of gamma irradiation were measured through microscopic analysis to determine morphological changes and concentration of the reducing sugar of hydrolysates. Microscopic images show that gamma irradiation causes structure breakage of the <I>Undaria</I> cell wall. The concentration of reducing sugar of hydrolysates significantly increased as a result of gamma irradiation, with or without acid hydrolysis. These results indicate that the combined method of gamma irradiation with acid hydrolysis can significantly improve the saccharification process for bioethanol production from marine algae materials.</P>

        Conjugation Process in Spirogyra varians Monitored with FITC-lectins (Zygnemataceae, Chlorophyta)

        The conjugation processes of a filamentous freshwater green alga Spirogyra varians were examined using FITClectins. Conjugation comprised five steps: 1) aligning with adjacent filaments, 2) formation of conjugation protrusion (papilla), 3) fusion of the protrusions, 4) formation of conjugation tube, and 5) formation of zygotes. Three lectins, ConA, RCA and UEA, showed considerable labeling during the progression of conjugation. FITC-ConA labeled the surfaces of filaments throughout the whole conjugation processes. FITC-RCA labeling was observed at the conjugation protrusions only after the papilla formation. Strong labeling continued until formation of zygotes at the contacting area where the conjugation tube developed, but no labeling was detected on the surface of vegetative filaments. The labeling decreased gradually over time and disappeared when zygotes were formed. FITC-UEA showed similar labeling pattern with FITC-RCA except that weak labeling remained after zygote formation. Inhibition experiments using RCA, UEA which are complementary to sugars L-fucose and D-galactose, showed considerable decrease of conjugation (< 32% vs. 70% in control). These results suggested that the lectin-carbohydrate recognition system might be involved in the conjugation of Spirogyra varians.

      • Effect of gamma irradiation on Korean traditional multicolored paintwork

        <P><B>Abstract</B></P> <P>Gamma irradiation can destroy fungi and insects involved in the bio-deterioration of organic cultural heritages. However, this irradiation procedure can alter optical and structural properties of historical pigments used in wooden cultural heritage paintings. The crystal structure and color centers of these paintings must be maintained after application of the irradiation procedure. In this study, we investigated the effects of gamma irradiation on Korean traditional multicolored paintwork (Dancheong) for the preservation of wooden cultural heritages. The main pigments in Korean traditional wooden cultural heritages, <I>Sukganju</I> (Hematite; Fe<SUB>2</SUB>O<SUB>3</SUB>), <I>Jangdan</I> (Minium; Pb<SUB>3</SUB>O<SUB>4</SUB>), <I>Whangyun</I> (Crocoite; PbCrO<SUB>4</SUB>), and <I>Jidang</I> (Rutile; TiO<SUB>2</SUB>), were irradiated by gamma radiation at doses of 1, 5, and 20kGy. After irradiation, changes in Commision Internationale d’Eclairage (CIE) color values (<I>L</I>*, <I>a</I>*, <I>b</I>*) were measured using the color difference meter, and their structural changes were analyzed using X-ray diffraction (XRD) analysis. The slightly change in less than 1 <I>dE</I>* unit by gamma irradiation was observed, and structural changes in the Dancheong were stable after exposure to 20kGy gamma irradiation. In addition, gamma irradiation could be applied to painted wooden cultural properties from the Korean Temple. Based on the color values, gamma irradiation of 20kGy did not affect the Dancheong and stability was maintained for five months. In addition, the fungicidal and insecticidal effect by less than 5kGy gamma irradiation was conformed. Therefore, the optical and structural properties of Dancheong were maintained after gamma irradiation, which suggested that gamma irradiation can be used for the preservation of wooden cultural heritages painted with Dancheong.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Effects of gamma irradiation on the Dancheong were evaluated. </LI> <LI> We confirmed that optical and structural properties of Dancheong were maintained. </LI> <LI> Irradiation can contribute the decontamination for wooden cultural heritages. </LI> <LI> It also can be used for preservation of painted-wooden cultural heritages. </LI> </UL> </P>

        中国跆拳道运动的发展机制研究

        As is known to all, Chinese Taekwondo sport was introduced into foreign countries. At present, Taekwondo sport has been developed in China for many years, and has a great market and development prospects. Although favored by the public, there are still some problems in the process of development. The research background and purpose of this paper is to study Chinese Taekwondo on the basis of continuous development, aiming to analyze and sort out the problems existing in the development of Chinese Taekwondo, and analyze the root causes of the problems, and then comb out the development prospects of Chinese Taekwondo. This paper mainly uses the following methods for research, literature method. After the content and purpose of the study are established, relevant literature about Chinese Taekwondo is consulted on CNKI, China Journal Net and other platforms according to needs, so as to provide a basis for further research. Survey interview method. According to the research needs, the investigation questions were sorted out, and detailed information was obtained through field investigation, visits to relevant experts and face-to-face interviews. Questionnaire survey. Relevant questionnaires are designed according to the research content construction needs, and the validity of the questionnaire is tested to make the questionnaire more scientific and reasonable Logical analysis. By classifying and analyzing the survey contents and data, the objective results and conclusions are derived based on the previous experience. Through the research of this article, finally the conclusion and enlightenment point is that although the Chinese public importance of Taekwondo in gradually improve, but compared with other sports for the development of time is very long, the Chinese Taekwondo need continuous efforts, only to get more people’s recognition, in the public more and more people are involved in the Taekwondo movement, like Chinese table tennis, badminton, Chinese Taekwondo movement can have a more long-term development.

        浅析中国跆拳道市场

        Taekwondo is a combination of “Taoist” thought, inheriting the excellent spiritual quality of “etiquette, justice, integrity, patience, self-denial and unyielding”. Since 2000, Taekwondo has been officially included in the 27th Sydney Olympic Games as a sport event due to its competitive and ornamental characteristics. With profound cultural background, the combination of spiritual will cultivation and fight fighting has attracted sports fans from all over the world to take part in this sport. As China and South Korea face each other across the sea and have a wide range of cultural exchanges, among which there are many sports competitions, Taekwondo is also a well-known sport event in China. China's economy has developed rapidly in recent years, there exists a huge development corresponding Taekwondo market prospects, at the same time, there are also many problems. In this paper, the history of Taekwondo is introduced into the Chinese market, the existing scale, existing problems and countermeasures are discussed, and discussed the development situation of Taekwondo in China market.

        中国高校跆拳道运动开展现状及发展对策研究

        In the mid-1990s, Taekwondo was introduced into China. Because of its features in fitness, self-defense and especially in etiquette, it was favored by Chinese university students. Around 2000, Taekwondo began to be on rise in China. At that time, Taekwondo classes and its training classes were gradually developed. It was so popular that Taekwondo could be found in community sports activities. However, with its continuous development in China, there are also some obstacles arising. For example, there is still a big gap between the relevant scientific research and the formally systematic training of Taekwondo in China and those in other developed countries in the world. At present, most of Chinese university students are lively and active, and their practical ability is relatively strong. Based on these, Taekwondo has its unique charm in Chinese contemporary universities, combined with its characteristics. In addition, Taekwondo is not only conducive to campus harmony, but plays an important role in the construction of campus culture. Through the method of literature, logical induction, questionnaire survey and expert interview and so on, this paper carries out a thorough investigation of the present situation of Taekwondo in Chinese universities in order to sort out the university students’ cognition on the Taekwondo and its development trend, and to study their learning interests and motivations and so on.

        A Study on the Hydrogen Economic Law for the Realization of Hydrogen Society in Korea

        한국 정부는 수소 자동차와 연료 전지를 중심으로 한 수소 경제사회를 지향한다. 이 연구의 목적은 2019년 1월 발표된 수소 경제 로드맵을 지원하기 위한 더 나은 입법의 방향을 제공하는 것이다. 수소차와 연료전지 기술에 강점을 가진 한국은 수소 산업이 세계의 이목을 끌고 있는 와중에 한국의 수소 경제관련 입법 동향은 수출, 투자, 관련 산업집단의 공동연구 등 국제협력을 위해서도 중요하다. 한국의 경우 수소경제법, 수소산업진흥법, 수소안전법 등이 모두 국회에 계류돼 있다. 또한 법적 근거 없이 수소차 공급과 충전 인프라 사업이 여러 기관 사이에 분산돼 있어 체계적인 대응이 어려웠다. 그 외에도 입법 미비로 인한 애로점을 극복하기 위하여 수소통합경제법을 제정해 수소경제사회에 대한 체계적인 대응을 할 필요성이 느껴진다. 연구결과, 비교법적으로 일본의 경우 장기적 관점의 수소경제 발전을 위한 법안이 있고, 미국은 민간부문까지 수소정책에 협력할 여지를 두고 있다. 영국의 경우에도 수소사회의 실현을 가속화하고 다양한 이해관계자의 의견을 수렴하는 제도적 장치가 있었다. 현재 국회 계류중이거나 논의 중인 한국 사회의 수소관련 법제 중 수소산업발전법의 경우, ‘기후변화대응기본계획’과도 더욱 밀접히 연계되어야 한다는 결론을 얻었다. 즉, 현재 국가 핵심정책인 수소산업 육성을 위해 에너지 기본계획과 기후변화계획을 함께 검토해야 한다. 또한 수소안전관리사업법은 수소경제에서 가장 중요한 법률이다. 수소에 관한 안전기준이 있어야 한다. 현재의 법안에는 모든 수소 관련 시설이 포함되어 있지 않다는 문제점이 있다. 수소를 고압으로 제조, 충전, 보관하는 시설은 “고압가스안전관리법”에 따라 기준을 준수하도록 되어 있다. 수소를 저압으로 제조, 충전, 저장, 판매 또는 사용하는 시설에는 안전기준이 불충분하다는 문제점도 시정되어야 한다. 또한 수소를 직접 사용하는 연료전지에 대해서는 안전기준이 제공되지 않았다. 수소연료의 안전관리 및 사업법은 공공의 안전을 확보하여 수소의 공급과 이용을 촉진하기 위해 제정되었다고 이러한 현실적인 안전 문제에 대한 대처 기준 등이 강화되어야 할 것이다. 또 수소경제 지속을 위한 체계적인 연구개발 지원도 요원하다. 수소 R&D 사업 중 한국이 투자를 통해 효과적일 수 있는 기술 분야를 뽑아 집중 육성하는 것도 필요하다. 미국과의 안전관리 노하우에도 기술 교류와 협력이 필요하다. 이와 함께 미국의 수소 및 연료전지 관련 행위에 대한 연구와 벤치마킹도 필요할 것이다. 이러한 다양한 정책적 노력과 법적 근거확립은 수소사회의 실현에 일익이 될 것이다. The South Korean government aims for a hydrogen economy society focusing on hydrogen-powered cars and fuel cells. The purpose of this study is to provide directions for better legislation to support the hydrogen economy roadmap. South Korea"s hydrogen industry, which has strengths in hydrogen car and fuel cell technology, draws the attention of the world. Korean legislative trends are also important to other countries for international cooperation, including export, investment, and joint research of related industrial groups. A roadmap to revitalize the hydrogen economy was announced in January 2019. The Act on the Hydrogen Economy, the Act on the Promotion of Hydrogen Industries, and the Act on the Safety of Hydrogen are all pending in the National Assembly. It was difficult to take a systematic response because projects for hydrogen car supply and charging infrastructure were distributed among several agencies. The purpose of legislation is desirable for a systematic response to the hydrogen economy society by enacting the Integrated Hydrogen Economic Act. The Korean government includes hydrogen in the category of climate change technologies. Therefore, the Act on Development of Hydrogen Industries should also be linked to the "Basic Plan for Climate Change Response". In other words, the basic energy plan and climate change plan should be considered together in order to foster the hydrogen industry, which is currently one of the key national policies. The Hydrogen Safety Management and Business Act is the most important law in the hydrogen economy. There should also be safety standards regarding hydrogen. The current bills do not contain all hydrogen-related facilities. Facilities that manufacture, charge, and store hydrogen at high pressure are required to comply with standards in accordance with the “High Pressure Gas Safety Management Act.” Safety standards are insufficient for facilities that manufacture, charge, store, sell, or use hydrogen at low pressure. In addition, safety standards were not provided for fuel cells which use hydrogen directly. The Safety Management and Business Act of Hydrogen Fuel was designed to promote the supply and utilization of hydrogen by securing public safety. In addition, systematic support for R&D is also needed for the continuation of the hydrogen economy. Of the hydrogen R&D projects, it is also necessary for South Korea to draw out technical areas that can be effective through investment and to foster them intensively. It will also need technological exchanges and cooperation in safety management know-how with the United States. In particular, research and benchmarking on the hydrogen and fuel cell-related acts of the US will also be needed.
