      • IL-17A induces osteoblast differentiation by activating JAK2/STAT3 in ankylosing spondylitis

        <P><B>Background</B></P><P>IL-17A has recently emerged as a potential target that regulates the extensive inflammation and abnormal bone formation observed in ankylosing spondylitis (AS). Blocking IL-17A is expected to inhibit bony ankylosis. Here, we investigated the effects of anti IL-17A agents in AS.</P><P><B>Methods</B></P><P>TNFα, IL-17A, and IL-12/23 p40 levels in serum and synovial fluid from patients with ankylosing spondylitis (AS), rheumatoid arthritis (RA), osteoarthritis (OA), or healthy controls (HC) were measured by ELISA. Bone tissue samples were obtained at surgery from the facet joints of ten patients with AS and ten control (Ct) patients with noninflammatory spinal disease. The functional relevance of IL-17A, biological blockades, Janus kinase 2 (JAK2), and non-receptor tyrosine kinase was assessed in vitro with primary bone-derived cells (BdCs) and serum from patients with AS.</P><P><B>Results</B></P><P>Basal levels of IL-17A and IL-12/23 p40 in body fluids were elevated in patients with AS. JAK2 was also highly expressed in bone tissue and primary BdCs from patients with AS. Furthermore, addition of exogenous IL-17A to primary Ct-BdCs promoted the osteogenic stimulus-induced increase in ALP activity and mineralization. Intriguingly, blocking IL-17A with serum from patients with AS attenuated ALP activity and mineralization in both Ct and AS-BdCs by inhibiting JAK2 phosphorylation and downregulating osteoblast-involved genes. Moreover, JAK2 inhibitors effectively reduced JAK2-driven ALP activity and JAK2-mediated events.</P><P><B>Conclusions</B></P><P>Our findings indicate that IL-17A regulates osteoblast activity and differentiation via JAK2/STAT3 signaling. They shed light on AS pathogenesis and suggest new rational therapies for clinical AS ankylosis.</P><P><B>Electronic supplementary material</B></P><P>The online version of this article (10.1186/s13075-018-1582-3) contains supplementary material, which is available to authorized users.</P>

        Effect of Cytokines and bFGF on the Osteoclast Differentiation Induced by 1α,25-(OH)<SUB>2</SUB>D<SUB>3</SUB> in Primary Murine Bone Marrow Cultures

        <P> Bone is a complex tissue in which resorption and formation continue throughout life. The bone tissue contains various types of cells, of which the bone forming osteoblasts and bone resorbing osteoclasts are mainly responsible for bone remodeling. Periodontal disease represents example of abnormal bone remodeling. Osteoclasts are multinucleated cells present only in bone. It is believed that osteoclast progenitors are hematopoietic origin, and they are recruited from hematopoietic tissues such as bone marrow and circulating blood to bone. Cells present in the osteoclast microenvironment include marrow stromal cells, osteoblasts, macrophages, T-lymphocytes, and marrow cells. These cells produce cytokines that can affect osteoclast formation. In vitro model systems using bone marrow cultures have demonstrated that IL-1β, IL-3, TNF-α, bFGF can stimulate the formation of osteoclasts. In contrast, IL-4 inhibits osteoclast formation. Knowledge of cytokines and bFGF that affect osteoclast formation and their capacity to modulate the bone-resorbing process should provide critical insights into normal calcium homeostasis and disorders of bone turnover such as periodontal disease, osteoporosis and Paget s disease.

        The Effect of Extract from Sea Buckthorn on DNCB-induced Atopic Dermatitis in NC/Nga Mice

        Sea Buckthorn (Hippophae rhamnoides L.) has been used in traditional medicine for the treatment of cough, indigestion, circulatory problems and pain. The associated anti-inflammatory effect of this agent is achieved via the inhibition of Nf-${\kappa}B$ signaling, a property that has been demonstrated to effectively control the symptoms of various skin disorders, including atopic dermatitis. Accordingly, the purpose of this study was to assess the efficacy of Sea Buckthorn in reducing the production of lipopolysaccharide (LPS) activated nitric oxide (NO) by inhibiting the Nf-${\kappa}B$ pathway, as measured by the symptoms of atopic dermatitis (AD) occurring secondarily to inflammation and immune dysregulation. Our data demonstrate that Sea Buckthorn significantly decreased the LPS-induced production of NO (p<0.001). Atopic dermatitis was induced by repeated application of 2,4-dinitrochlorobenzene to the dorsal skin of mice. Topical application of 5% Sea Buckthorn extract improved the symptoms of AD, specifically reducing disease severity scores, scratching behaviors and epidermal thickness. When compared to the control group, animals treated with Sea Buckthorn exhibited increased serum IL-12 levels and decreased serum TNF-${\alpha}$, IL-4 and IL-5 levels. Such a modulation of biphasic T-helper (Th)1/Th2 cytokines may result in a reduction in serum IgE levels. Our findings suggest that mechanism of action of Sea Buckthorn in the treatment of AD is associated with a marked anti-inflammatory effect as well as an inhibition of Th2-mediated IgE overproduction via the modulation of biphasic Th1/Th2 cytokines. Such results suggest that topical Sea Buckthorn extract may prove to be a novel therapy for AD symptoms with few side effects.

        The purpose of this study was to investigate the contents and standards of Information Literacy or Information Technology Literacy current trend in America, and to give some suggestions for the preparation of prospective teachers in Korea. Although the concept Information Literacy seemed to be coined in the late 1970's, the term became more popular in the 1990's. Both Technology Literacy and Information Literacy are in popular to use. The Technology in Technology Literacy connotes Educational Technology, meaning familiarity and proficiency in use of technological systems, software, and information. For the contents and standards of Information or Technology Literacy, ISTE(International Society for Technology in Education) issued two national standards in this area: one for students, and the other for teachers. However, these standards, despite their extensive influence on leaders of schools and libraries, did not hint any level of mastery in specific system, application, and behaviors. So there remains need for the investigation of both students and teachers required mastery levels for graduation or certification.

      • 土壤에서 PCNB의 殘留消長과 微生物 活性에 미치는 影響

        PCNB를 土壞에 處理하여 PCNB의 分解樣相, 殘留消長, 微生物, 生菌數 및 酵素生性에 미치는 影響을 經時的으로 調査하여 얻는 結果를 要葯하면 다음과 같다. 湛水土壞에서의 半減期는 7.9日이었고 非湛水土壞에서는 22.5日로서 湛水土壞에서는 분해가 非湛水土壞에서보다 빨랐다. 減菌湛水土壞에서의 半減期는 82.4日이었고 減菌非湛水土壞에서는 89.9日로서 減菌土壞에서는 非減菌土壞에서보다 分解速度가 緩慢하였다. 非湛水土壞에 PCNB를 5㎍/g濃度로 處理한 45日 후에 PCNB의 약 46%가 그 分解産物인 pentachloroaniline(PCA)으로 變換되었으며 減菌土壞에서는 PCA가 確認되지 않았다. PCNB가 微生物生菌數 에 미치는 影響을 조사한 結果 bateria와 fusarium,屬菌은 藥劑의 影響을 받지 않았고 actinomycetes와 fungi는 약간의 沮害를 받았다. 또 PCNB가 效素活性에 미치는 影響은 phoshoatase, protease, dehydrogenase 및 ß- glucosidase의 順으로 沮害率이 增加하였다. This study was carried out to investigate the persistence and the biodegradation pattern of pentachloronitrobenzene(PCNB) in soil. The effect of PCNB on soil microorganisms and enzyme activities were also determined. The results obtained are summarizexi as follows . Half-life of PCNB was 7.9 days in unsterilized submerged soil and 22.5 days in unsterilized moist soil. However, half-life of PCNB was 82.4 days in sterilzed submerged soil and 89.9 days in sterilized moist soil. PCNB was more rapidly degraded in unsterilized submerged soil than in sterilized soil. Loss of PCNB in unsterilized submerged, moist soil was accompanied by an increase in pentachloraniline (PCA), the principal degradation product, residue. But PCA was not detected in the sterilized soil. About 46% of the original PCNB was degraded in 45 days incubation in unsterilized submerged soil, which was evidenced by the production of PCA. Of the effect of PCNB on soil microorganisms and enzyme activities, number of bacteria and fusarium in soil was nearly unchanged, that of actinomycetes and fungi was decreased, and that of pythium was decreased strongly. The effect of PCNB on me activities in soil increased in the order: phosphatase<pro-tease<dehydragenase<ß-gluccsidase.

      • 미국 교사교육의 개혁동향 및 시사점

        This paper was intended to investigate the main currents and some recent reform movements in American teacher education for the purpose of getting some insights for the new reframing of Korean teacher education in coming days. For this purpose, traditional and current practices in pre-service education, in-service training, and accreditation system of teacher education programs were scrutinized in detail. The result was presented as following : i) the features if pre-service education in the form of candidates selection, training institutions, education programs, and certification. ii) the training programs of in-service education in terms of types, institutions, and ‘teacher center’. And iii) the evaluation procedures of NCATE and A-plus model of CTEE as typical cases of accreditation system. The paper concludes that would be reformers in Korean teacher eeducation programs can have some useful ideas from American teacher education, such as internship model, extended years of education, regular renewals of certificates and related further education, and accreditation system of teacher education programs in various institutions.

        This paper suggests the table for a mobile agent path which performs on transfer a mobile agent in security and based on which introduced the transfer protocol of a mobile agent. As the last prodceure of this, shows that how to write the closing time of a mobile transferring by using a Time stamp and keep not only preparing for the stop and attack for a receiving host but also the record of the transfer for a mobile agent. Later this paper will analyzes the security for an attack of a receiving host, a transmitting host and the third part host based on proposed system.

      • WO₃계 NO_(x)센서의 검지 특성과 바인더 첨가 효과

        The effects of various binderssuch as PVA, silica sol and alumina on the sensing characteristics of TiO_(2) (4 wt. %)-doped WO_(3)-based NO_(x). sensors were investigated. The sensor with PVA (polyvinyl alcohol) binder showed an improved sensitivity (R_(gas)/R_(air)- in NO_(x) 15 ppm = 375), response time and cross-sensitivity (selectivity) for interference gases (CO, SO_(2), CH_(4), C_(3)H_(8)). It could also be operated at a lower consumption power (about 1.6W at 180℃). The sensors with silica sol and alumina binders showed a decreased sensitivity (R_(gas)/R_(air) in NO_(x) 15 ppm = 63 and 31, respectively) for NO_(x) gas sensing, but they showed an improved cross-sensitivity, response and recovery time (1 -2 min. and 1 min., respectively) than the sensor fabricated without binder.

        The purpose of this study was to analyse the expectations of classroom teachers in middle school students, concerning teachers' personality traits, class management, instruction and student guidance. Generally students seem to favor more good than able teachers of kind and generous mind in twenties, who can deal their students with fair treatments, and guide with concrete instructional and disciplinary principles.
