        맥엽 및 항산화 조성물의 생리 기능적 특성에 관한 연구 : 아질산염 소거능을 중심으로 Study on the Nitrite Scavenging Effect

        맥엽과 이를 이용한 항산화 조성물을 제조하여 이에 존재하는 nitrite scavenging 효과와 이에 따른 발암원으로 부터의 보호 효과를 조사한 결과는 다음과 같다. 맥엽과 항산화 조성물 모두 수층에 비하여 메탄올층에서 아질산염 소거능이 2배 이상 높게 나타나 M-BL50의 0.01㎎/㎖ 농도에서 약 64.3%, M-AM 0.01㎎/㎖의 처리군이 이보다 약간 높은 68.3% 정도로 나타내었다. 마우스 단핵구 기원인 RAW 264.7(macrophage, mouse)에 맥엽과 항산화 조성물의 수층과 메탄올층을 처리하고 세포 생존율을 측정한 결과 수층 처리군에 비해 메탄올층 처리군에서 모두 세포 생존율이 더 낮게 측정되었으며 모든 시료에서 농도의존적으로 세포 증식이 억제되었다. 특히, M-AM을 처리한 군의 경우 모든 농도에서 가장 낮은 세포 생존율을 나타내어 항산화물 첨가에 의해 세포 증식 억제 효과가 상승됨을 알 수 있었으며, IC_(50) 수치 또한 메탄올층과 A-AM 시료 처리군에서 현저히 낮은 수치를 나타내었다. 또한 세포 내 아질산염 소거능을 측정한 결과 역시 마찬가지로 맥엽 입자 크기에 따른 차이는 나타나지 않았으며 수층 처리군에 비해 메탄올층 처리군과 항산화물을 첨가한 군에서 모두 높은 소거능을 나타내어 맥엽 및 기타 항산화물을 섭취함으로써 체내에서의 니트로사민 생성을 억제시킬 수 있을 것으로 예상된다. This study was conducted to estimate the biofunctional characteristics, namely nitrite scavenging effect of barley leaves (BL) and their mixture (AM) with other antioxidants. Aqueous or methanol extracts were obtained from BL and AM. Aqueous and methanol extracts from barley leaves had relatively higher nitrite scavenging effects and its activities and contents increased in a concentration-dependent manner. The activities and contents of methanol extracts obtained from BL and AM were higher than those of aqueous extracts. Especially, AM containing BL and other antioxidant mixture had the highest activities and contents increased in a concentration-dependent manner. BL or AM were added to macrophage cells of RAW 264.7 Survival rates of the cells treated with BL and AM were measured to be different. IC_(50) value decreased in a concentration-dependent manner by the addition of aqueous or methanol extracts from BL and AM. Especially, methanol extracts of AM had the highest nitrite scavenging effects. Thus, BL and AM may have protective effect against carcinogen and immune ability against reactive molecules through NO (nitric oxide) signal pathway. From the above results, barley leaves appear to contain natural anticancer and immune-related agents and may have potentiality to be used as functional food ingredients.

      • 표준 영상의 압축/ 복원을 위한 양자화/ 역양자화 기능 모듈의 설계 및 구현

        본 논문에서는 표준 영상 데이타의 압축/복원을 고성능으로 처리하기 위한 양자화/역양자화 기능 모듈에 대한 설계와 구현에 대하여 논한다. 기능 모듈의 성능은 SOG(Sea Of Gate) 구현을 통하여 20 MHz와 50 MHz의 클럭 속도에서 동작을 확인하였다. In this paper, a design and implementation for the quantization/inverse quantization functional module is presented, which aims at the high performance of data compression/decompression in the standard image information processing. The design, implemented in SOG (Sea Of Gate), has been verified at the clock speeds of 20MHz and 50MHz(in process).

        주의력결핍 과잉행동장애 아동의 지적수준에 따른 정신병리와 실행기능의 차이

        To investigate psychopathoIogy and executive functions of attention deficit hyperactivity disorder (ADHD) children according to intelligence level, this study included 197 ADHD children who visited the outpatient department of neuropsychiatry of YeungNam University Medical Center, from July 2000 to June 2002. The children were divided into groups based on their intelligence levels. They were compared by the Personality Inventory for Children (PIC), Conncers' Continuous Performence Test (CPT), and Wisconsin Card Sorting Test (WCST). There were significant differences in PIC, on the subscales of verbal development, socialization and autism. In the CPT, there was no significant difference. In the WCST, there were significant differences in the total number of errors, the number of perseverative errors, the number of completed categories and the number of trials needed to complete the first category. Considering these results, the intelligence level of ADHD children is related to their disabilities and behavioral symptoms.Executive functions such as abstract thinking, categorization, working memory and flexibility had significant relationship to the intelligence levels of ADHD children. Therefore, the intelligence level of children with ADHD influences the higher executive functions of regulating attention and information processing rather than attentional functions and capacity alone.

        Two Flexible Loops in Subtilisin - like Thermophilic Protease , Thermicin , from Thermoanaerobacter yonseiensis

        A gene that encodes a thermostable protease, coined thermicin, has been isolated from Thermoanaerobacter yonseiensis that is expressed and characterized in E. coli.. In order to elucidate the molecular characteristics on thermostability of the enzyme, molecular modeling and mutagenesis technology were applied. In the modeling structure, the structural core, including the active site, was well conserved; whereas, the two loop regions were unique when compared to thermitase. The mutant enzyme with the small loop deleted (D190-I196), based on modeling structural information, showed identical enzyme activity. However, when the large loop was deleted (P233-P244), a little lower K_m and even a lower kcat was found. This indicates that the large loop could influence catalytic activity. However, the unfolding temperature (T_m), which was determined by a differential-scanning calorimetry for the mutant enzyme deleted the small loop, was 96℃. This is 14℃ lower than that for the parent thermicin. These results suggest that the small loop may play a role in maintaining the proper folding of the enzyme at high temperatures, whereas the large loop might be related to catalysis.

      • Anti-inflammatory activities of <i>Reynoutria elliptica</i> through suppression of mitogen-activated protein kinases and nuclear factor-κB activation pathways

        <P><I>Reynoutria elliptica</I> has been used in traditional Korean medicine to promote blood circulation, relieve pain, increase dieresis, and alleviate respiratory problems, through as yet undefined mechanisms. We set out to determine whether the anti-inflammatory effects of this plant are linked with its ability to suppress mitogen-activated protein kinases (MAPKs) and nuclear factor-κB (NF-κB) activation in lipopolysaccharide (LPS)-activated RAW 264.7 cells. We found for the first time that the hexane fraction of <I>Reynoutria elliptica</I> (HRE) significantly inhibited LPS-stimulated NO and PGE2 synthesis. This is due to the diminishing of the mRNA and protein expression of iNOS and COX-2, respectively. HRE also suppressed LPS-stimulated TNF-α secretion in a dose-dependent manner, which might be due to the suppression of LPS-induced MAPKs and NF-κB activation. Moreover, our HPLC data demonstrated that the major components of the HRE were bioactive compounds such as emodin-6-Glc, emodin, and physcion. Overall, our results indicate that <I>Reynoutria elliptica</I> could be provided as a potential candidate for anti-inflammation treatment.</P>

        Effects of the Combination of Glucose, Chromium Picolinate, and Vitamin C on Lipid Metabolism in Steers

        This study was conducted to examine the effects of glucose, chromium picolinate (CrP), and vitamin C (Vit C) on lipid metabolism in Korean native steers fitted with indwelling catheters. A total of 12 Korean native steers were randomly allocated to the following treatments: 1) normal control diet, 2) same as 1) +250 g of glucose by intravenous (IV) infusion, 3) same as 2)+13.5 g CrP administered orally, and 4) same as 3)+2.52 g Vit C by IV infusion. Glucose, Vit C, and CrP treatments were administered for five days. At days 1 and 3, serum insulin was higher in treated animals than in those fed the control diet (p<0.05). Serum non-esterified fatty acid (NEFA) concentration in the steers on treatment 2), control+13.5 g CrP, was lower than those on other treatments at 90 min post-infusion on days 1 and 3 (p<0.05). The expression of peroxisome proliferator-activated receptor-${\gamma}$ (PPAR${\gamma}$)2, stearoyl-CoA desaturase-1 (SCD), fatty acid synthase (FAS), and glucose transporter type 4 (Glut 4) in the longissimus muscle of steers on treatment 2 was higher than those on other treatments. In conclusion, the results suggest that CrP is associated with the regulation of gene expression involved in adipogenesis.

        Anti-diabetic and anti-obesitic effects of aqueous extracts of Yangkyuksanhwa-tang and its two major compositions on <i>db</i>/<i>db</i> mice

        <P><B>Abstract</B></P> <P>Type 2 diabetes mellitus (T2DM) is a metabolic syndrome that results from target-tissue resistance to insulin. Obesity is the condition of excess body fat accumulation. T2DM and obesity are both associated with hypertension, hyperlipidemia, and abdominal obesity. In Korean medicine, Yangkyuksanhwa-tang (YKSHT) has been prescribed for patients with T2DM. Oral glucose tolerance tests (OGTT), multiplex assays and hemoglobin A<SUB>1C</SUB> (HbA1C) assessments were performed to determine the anti-diabetic effects of YKSHT and two major compositions of YKSHT, <I>Lonicera japonica</I> Thunb. (LJT) and <I>Rehmannia glutinosa</I> (RG) on <I>db/db</I> mice, a rodent model for T2DM. To study the anti-obesitic effects of LJT, RG or YKSHT, blood profiling including the triglycerides (TGs) and the total, LDL and HDL cholesterol levels were measured. In addition, body index measures such as the liver, retroperitoneal and epididymal fat tissues were collected and weighed. Mice treated with RG or YKSHT showed reduced blood glucose levels after stimulating the plasma GLP-1 levels. The multiplex assay results support the weight-controlling effects of the LJT, RG and YKSHT treatments, showing reducing levels of ghrelin and the induction of peptide YY (PYY) secretion. The YKSHT treatment reduced plasma TG levels and increased HDL cholesterol levels. The weights of the liver, retroperitoneal and epididymal fat tissues were reduced after the YKSHT treatment. Hence, we suggest that YKSHT can be utilized for the prevention and treatment of T2DM and obesity simultaneously.</P>

        Double-deck train has been attracted growing attention as next generation transportation around metropolis because of high passenger carrying capacity. In order to develop high-speed double-deck train with low operational costs, the carbody must be designed as light as possible. In addition, the carbody must be strong enough to ensure the safety of passengers. For these purposes, weight minimization that determines thickness of aluminum extruded panels(AEP) of the carbody can be performed. First, we perform the screening process that select sensitive or/and important design variables through design exploration because the design variables of carbody are too many to treat all of them in the design optimization. Then, weight minimization is accomplished under multi-loading condition such as vertical, compressive and torsional load, while satisfying user specified strength constraints. Finally, the result of design optimization is discussed by comparison with it of initial design.
