        Effects of Aralia cordata Thunb. on Proteoglycan Release, Type II Collagen Degradation and Matrix Metalloproteinase Activity in Rabbit Articular Cartilage Explants

        Background & Objective: Articular cartilage is a potential target for drugs designed to inhibit the activity of matrix metalloproteinases (MMPs) to stop or slow the destruction of the proteoglycan and collagen in the cartilage extracellular matrix. The purpose of this study was to investigate the effects of Aralia cordata Thunb. in inhibiting the release of glycosaminoglycan (GAG), the degradation of collagen, and MMP activity in rabbit articular cartilage explants. Methods : The cartilage-protective effects of Aralia cordata Thunb. were evaluated by using glycosaminoglycan degradation assay, collagen degradation assay, colorimetric analysis of MMP activity, measurement of lactate dehydrogenase activity and histological analysis in rabbit cartilage explants culture. Results : Interleukin-la (IL-1a) rapidly induced GAG, but collagen was much less readily released from cartilage explants. Aralia cordata Thunb. significantly inhibited GAG and collagen release in a concentration-dependent manner. Aralia cordata Thunb. dose-dependently inhibited MMP-3 and MMP-13 expression and activities from IL-1a-treated cartilage explants cultures when tested at concentrations ranging from 0.02 to 0.2 mg/ml. Aralia cordata Thunb. had no harmful effect on chondrocytes viability or cartilage morphology in cartilage explants. Histological analysis indicated that Aralia cordata Thunb. reduced the degradation of the cartilage matrix compared with that of IL -1a-treated cartilage explants.

      • 일반화된 지수생존모형의 추정

        일반화된 지수생존모형(peneralized exponetial survival model)을 고려하여 이 모형의 모수를 추정하는 수정된 FS(modified Fisher scoring)방법을 제안한다. 이를 위해 우도방정식(likelihood equations)을 유도하고 초기추정치(initial estimate)를 포함한 추정알고리즘(estimating algorithm)을 개발한다. We consider generalized exponetial survival model and propose a modified Fisher scring method which estimates the model parameters. Also, the fitting algorithm including an initial estimate is developed.

        본 논문은 지수수명분포에서 제2종 중도절단된 가속수명자료를 이용하여 고장률의 사전분포의 평균을 알 때, 정상조건하에서 하나의 미래 관찰치의 예측문제를 사전분포의 모수에 대하여 적률추정량에 기초한 불편추정량(unbaised estimator)을 이용하는 경험적 베이즈 접근방법을 적용시켜 경험적 베이즈 예측분포와 예측구간에 대하여 연구하였다. In accelerated life tests(ALTs), the failure time of a test item is observed under high stress conditions, and based on these data, one intrests statistical inference on the parameters of lifetime distribution at the normal stress. Under the type-II censoring, we assume that the life distribution is exponential distribution. When the information on the prior is partially known, we device the empirical Bayesian analysis of ALTs data in order to obtain the empirical Bayesian predictive density and predictive intervals for a future observation under the normal stress level.

        This paper set up the nonparametric test statistic U₂, which expanded U₁-statistic with degree(3,3) into the form of a two-sample U-statistic with degree(5,5), say U₂for the purpose of developing the statistic that may be more efficient than the U₁-statistic suggested by Shetty & Govindarajulu(1988), in a two-sample location test. We compared the U₂test with the previous test, in the sense of ARE, under some special population distributions. To obtain the ARE of teh U₂test w.r.t. the previous test, the asymtotic null distribution and the efficacy of U₂were derived. Conclusively, we discovered that the U₂test expanded to degree(5,5) was more efficient than the previous test in heavy-tailed distributions like the double exponential and Cauchy.

        열공형과 비열공형 피질하 혈관성 치매에서 위험인자의 차이에 관한 비교 연구

        Backgrounds and Objectives: Vascular dementia is a group of dementing disoders arising from various stroke syndrome. Among these. subcortical ischemic vascular dementia (SIVD) is regarded as a relatively distinct clinical entity. However, MRI patterns of SIVD are not homogenous. In some patients, lacunes are dominant, and in others, subcortical white matter changes are. This study was designed to compare risk factor profiles between SIVD with and without multiple lacunes. Methods: We divided 47 subjects (22 males, mean age. 68 years) recruited from VADAPET (Multicenter Trial For Evaluation Of The Changes In the PET Images Of Subcortical Vascular Dementia Patient) study into two groups one with more than 5 lacunes in deep gray matter (lacune group) and the other with 5 or less(non-lacune group) Clinical characteristics and laboratory findings of two groups were compared. Results: Nineteen of 47 patients (40%) belonged to the lacune group. The lacune and non-lacune groups d d not differ in the following variables: age, hypertension, diabetes mellitus, hyperlipidemia heart disease, history of stroke or TIA, history of trauma or major surgery, family history of hypertension stroke, or dementia, age at diagnosis of dementia, body mass index, white blood cell count, ESR, CRP, fibrinogen, hemoglobin A1C, total cholesterol. LDL cholesterol creatinine, proteinuria, glucosuria, and microhematuria. However, male sex, smoking alcohol. hemoglobin, and HDL cholesterol were possibly associated more with lacune group SIVD than with non-lacune group (p<0 1) Multivariate analyses revealed that smoking, hemoglobin, and HDL cholesterol were independent predictors of SIVD with multiple lacunes Conclusion: Our study suggests that SIVD with multiple lacunes may be significantly different in smoking habits hemoglobin, and HDL cholesterol from SIVD without multiple lacunes.

      • On Estimation of Poisson-Gamma Model with Multivariate Count Data

        다변량 개수자료에서 변량효과를 통해 의존성을 모사하는 포아송 변량효과 모형은 최근 상당한 관심을 불러일으키고 있다. Lee와 Nelder (1996)는 변량효과 모형의 추론에 대해 다단계우도의 사용을 제안하였다. 변량효과의 분포가 감마인 포아송-감마모형하에서, 그들의 다단계우도 추정법 보다 더 우수한 수정된 다단계우도 추정법을 제안한다. 우리의 방법의 타당성을 보이기 위해, Cox와 Reid (1987)의 주변우도 추정법과 비교하고 하나의 예를 제시한다. Poisson random effects models which describe dependence via random effects n multivariate or repeated count data have been recently considerable attention in the literature. Lee and Nelder (1996) proposed to the use of hierarchical-likelihood (or h-likelihood) for inference from models with random effects. For the estimation of Poisson-gamma model where the random effects follow a gamma distribution, we propose a corrected h-likelihood method which provides better performance than the h-likelihood method. Comparison is made with marginal likelihood method of Cox and Reid (1987). An example is presented to illustrate our method.

        A correction of SE from penalized partial likelihood in frailty models

        The penalized partial likelihood based on restricted maximum likelihood method has been widely used for the inference of frailty models. However, the standard-error estimate for frailty parameter estimator can be downwardly biased. In this paper we show that such underestimation can be corrected by using hierarchical likelihood. In particular, the hierarchical likelihood gives a statistically efficient procedure for various random-effect models including frailty models. The proposed method is illustrated via a numerical example and simulation study. The simulation results demonstrate that the corrected standard-error estimate largely improves such bias.
