        마로 부터 분리한 Acetylmannan의 Paraquat 독성 억제 효과

        Paraquat is a useful nonselective herbicide widely used throught the world. However accidental or intentional ingestion of the herbicide cause fatal pulmonary injuring. But there is not suitable antidote of paraquat intoxication and therapeutic agents now be used are not effective. So, in this study we intended to evaluate the inhibitory effects of acetylmannan from Dioscorea balolas on paraquat toxicity. 100mg/kg acetylmannan from wild or cultured Dioscorea batatas was administered orally to male SD rats for 3 days and the administration time interval was 24 hours. After one hour of final administration, 50 mg/kg paraquat was administered intraperitonially. After 24 hours, the biochemical parameters of blood and tissues were examined. In paraquat treated groups, sGPT, BUN, creatinine, ALP levels were increased by 2 to 4 times of normal values. However in acetylmannan from wild Dioscorea batatas treated groups, sGPT, BUN, creatinine, ALP levels in blood and lung tissue were significantly decreased to normal levels. In acetylmannan from cultured Dioscorea batatas treated groups, BUN, creatinine were significantly decreased to normal values, but not in sGPT, ALP levels. Therefore, we concluded that acetylmannan from wild Dioscorea batatas can be used as an antidote of paraquat toxicity.

        Sodium Azide와 2-AF에 대한 마(Dioscorea batatas Decene)의 항변이원성

        The objective of this study was to screen the antimutagenicity of yam enzymatic browning reaction product(YEBRP), mucopolysaccharide and dietary fiber from yam to the mutagen of sodium azide and 2-aminoflourene(2-AF). Antimutagenicity of YEBRP on the mutagenicity of sodium azide showed no difference compared to control without YEBRP but that of 2-AF was high in all substrate.(P<0.01) On the mutagenicity of sodium azide and 2-AF, antimutagenicity of mucopolysaccharide and dietary fiber were high (p<0.01) in α-cellulose and hemicellulose. Antimutagenicity of α-cellulose on the mutagenicity of 2-AF was high at 5 hours reaction time but that was decreased as the reaction time increased.

      • Gene profile of fibroblasts identify relation of CCL8 with idiopathic pulmonary fibrosis

        <P><B>Background</B></P><P>Idiopathic pulmonary fibrosis (IPF) is characterized by the complex interaction of cells involved in chronic inflammation and fibrosis. Global gene expression of a homogenous cell population will identify novel candidate genes.</P><P><B>Methods</B></P><P>Gene expression of fibroblasts derived from lung tissues (8 IPF and 4 controls) was profiled, and ontology and functional pathway were analyzed in the genes exhibiting >2 absolute fold changes with <I>p</I>-values < 0.05. <I>CCL8</I> mRNA and protein levels were quantified using real-time PCR and ELISA. <I>CCL8</I> localization was evaluated by immunofluorescence staining.</P><P><B>Results</B></P><P>One hundred seventy eight genes differentially expressed and 15 genes exhibited >10-fold change. Among them, 13 were novel in relation with IPF. <I>CCL8</I> expression was 22.8-fold higher in IPF fibroblasts. The levels of <I>CCL8</I> mRNA and protein were 3 and 9-fold higher in 14 IPF fibroblasts than those in 10 control fibroblasts by real-time PCR and ELISA (<I>p</I> = 0.022 and <I>p</I> = 0.026, respectively). The <I>CCL8</I> concentrations in BAL fluid was significantly higher in 86 patients with IPF than those in 41 controls, and other interstitial lung diseases including non-specific interstitial pneumonia (<I>n</I> = 22), hypersensitivity pneumonitis (<I>n</I> = 20) and sarcoidosis (<I>n</I> = 19) (<I>p</I> < 0.005, respectively). Cut-off values of 2.29 pg/mL and 0.43 pg/mL possessed 80.2 and 70.7% accuracy for the discrimination of IPF from NC and the other lung diseases, respectively. IPF subjects with <I>CCL8</I> levels >28.61 pg/mL showed shorter survival compared to those with lower levels (<I>p</I> = 0.012). <I>CCL8</I> was expressed by α-SMA-positive cells in the interstitium of IPF.</P><P><B>Conclusions</B></P><P>Transcriptome analysis identified several novel IPF-related genes.         <▼1><P>The power factor of mesoporous LSMO films was enhanced with an increase in the surface area.</P></▼1><▼2><P>The thermoelectric power factor of La0.7Sr0.3MnO3 (LSMO) is closely related to its oxygen-deficient nature. In this study, the oxygen content of mesoporous LSMO films was controlled using various annealing atmospheres (oxygen, nitrogen, air, and argon) to investigate the relationship between oxygen deficiency and the power factor. The effect of the mesoporous structure on the power factor of the prepared LSMO films was also studied through analyses of structural and electrical properties. The oxygen-deficient state induces an increase in the lattice parameter of LSMO, which results in an increase in the electrical resistivity and an enhanced Seebeck coefficient. This phenomenon was emphasized upon the introduction of a pore structure in LSMO because of an increase of the unstable surface area. The oxygen-deficient state results in an increase in the amount of manganese in the Mn<SUP>3+</SUP> valence state and in an increase of the lattice parameter; these effects were confirmed through photoemission spectroscopic analysis. As a result, LSMO can be applied as a thermoelectric material because of the successful preparation of LSMO films with a mesoporous structure and because of the enhancement of the power factor as a consequence of increased oxygen deficiency.</P></▼2>

        노화 생쥐의 복부지방에서 수영운동에 의한 AMPK와 GLUT4 단백질 변화

        본 연구의 목적은 노화된 생쥐에 있어서 수영운동에 따른 AMPK와 GLUT4의 발현을 알아보는 것이다. 이에 본 실험에서는 노화된 생쥐에게 수영운동을 시행하였으며, 몸무게 및 복부지방의 무게를 측정하였고, 지방세포의 크기변화를 Oil-Red 염색법으로, 지방조직에서 AMPK와 Glut-4의 단백질을 분석하였다. 연구대상은 2개월 SWV계 숫컷 생쥐 10마리를 청년대조군(n=10), 12개월 노화된 SWV계 숫컷 생쥐 20마리를 노화대조군과 노화운동군(n=10)의 3군으로 분류하였고, 유산소운동은 주당 5회 40분씩 시행하였으며, 주 1회 체중을 측정하였다. 체중의 변화는 노화대조군이 다른 군에 비해 실험 종결 때까지 높은 체중을 유지하였으나, 노화운동군에서는 수영운동 2주후부터 7주까지 지속적으로 체중이 줄어(p<.05, 001)들었다. 혈당변화는 노화운동군이 다른 두 군에 비해 2주부터 뚜렷한 감소(p<.05)를 보였으며 실험종결 때까지 낮은 혈당을 유지하였다. 8주간 유산소운동 후 복부지방량의 변화는 노화운동군이 다른 두 군에 비해 확실하게 줄어들었고(p<0.001), 노화대조군의 지방세포크기는 비대해져 있었으나, 노화운동군에서는 작은 지방세포의 크기를 보여 주었다. 특히 AMPK와 GLUT-4의 발현은 노화운동군에서 다른 두 군에 비해 뚜렷하게 증가되었다. 이상의 결과를 종합하면 8주간의 수영운동은 노화된 생쥐의 체중 및 복부지방을 감소시켜 지방비대의 억제효과를 나타내었고, AMPK의 단백질발현증가는 지방생성 증가를 억제함을 알 수 있었으며, GLUT-4의 단백질발현증가는 혈당의 수치를 조절하는 효과가 있었다. 따라서 장기간의 수영운동은 노화로 인한 대사의 기능을 향상시켜 대사성질환으로 부터 보호할 수 있을 것으로 생각된다. The present study was designed to examine the protein changes of AMPK and GLUT-4 by swim exercise in fat tissue of aging mice. We performed aerobics exercise by swimming, measured body weight(BW)and measured blood glucose(BG), and performed Oil-Red stain for adipocyte and western blot for AMPK and GLUT-4 protein in fat tissue. SWV(n=30) mice at age of 2 months and 12months old were divided into 3 groups as follows: (1) Old control group (n=10, O-Con), (2) Young control group(n=10, Y-Con) (3) Old+exercise group (n=10, O-Ex). The swimming exercise was performed 5 days /week with duration of 40minutes for 8weeks. BW and BG were measured once per week. Results showed that the BW of O-Con group was is higher than other groups, interestingly, BW of Old-Ex was significantly decreased (p<.05, p<.001) from 2nd. BG of O-Ex groups showed markedly decreased (p<.05, p<.001) from 2 weeks after exercise and continuously lower than other groups. Retroperitoneal fat fed weight of O-Ex group was significantly decreased (p<.001) and adipocyte size of O-Con group was bigger than other groups, but O-Ex group smaller sized adipocyte than other groups. Particulary, AMPK and GLUT-4 which are related with lipid metabolism was markedly inhibited compared to Y-Con group, whereas O-Ex group showed increased of AMPK and GLUT-4 protein expression than other groups. These results suggest that swim exercise for 8 week should inhibit adipocyte hypertrophy by controling the BW and decreasing retroperitoneal fat fed through AMPK protein expression and controling protective of BG the by decreasing the BG through GLUT-4 protein expression in fat tissue of aging mice. Thus, exercise may be helpful to improvement of lipid metabolism function and may be protective effect from lipid metabolism disease.

      • The Mission Strategic Significance of Korean Church Revival Movement

        Since the Great Awakening of Pyung Yang in 1907, the Korean church has gone through remarkable growth. Examining the special conditions which supported such growth would be helpful to the Korean church in formulating its mission strategies abroad. In looking at the past growth of the Korean church, it is obvious that its growth was centered on the layman. Robert Alexander Hardie, a lay missionary from Canada, held a bible conference in Wonsan and this became the starting point of the Pyung Yang’s Great Awakening. As this example shows, the role of laymen is very important in mission work. Second, the Korean church’s growth had at its core the devotion, prayers, and Bible-centered life of its members. Carrying on the momentum of the Great Awakening, churchgoers held Bible conferences throughout the country and continued their life of faith. The main reason that the Korean church experienced such growth was the Bible-loving faith of Korean Christians. In the same way, mission sites need to focus on learning the Bible and praying in devotion. Third, the Korean church’s growth was supported in part by the hospitals and medical services provided by the missionaries. The early missionaries devoted much of their mission work to medical care so that by 1922 there were 22 Christian hospitals in the country. Such examples of medical care should be applied to today’s mission efforts. Fourth, education played a big role in the growth of the Korean church. From the very beginning, missionaries founded mission schools. These schools helped spread the gospel to many of its students. Today’s mission sites can also benefit greatly from the establishment of Christian mission schools. Fifth, the Korean church’s growth was influenced by the cooperation of missionaries from different denominations. These missionaries work together toward the growth of the Korean church. They developed material for Sunday schools, discussed mission policies, and collaborated on effective evangelization. Mission sites can also carry on its work of evangelization through greater cooperation with other denominations. In today’s global society, the Korean church must respond to social demands and cooperate to establish a more comprehensive mission strategy.

        Comparison of Model-simulated Atmospheric Carbon Dioxide with GOSAT Retrievals

        Global atmospheric $CO_2$ distributions were simulated with a chemical transport model (GEOS-Chem) and compared with space-borne observations of $CO_2$ column density by GOSAT from April 2009 to January 2010. The GEOS-Chem model simulated 3-D global atmospheric $CO_2$ at $2^{\circ}{\times}2.5^{\circ}$ horizontal resolution using global $CO_2$ surface sources/sinks as well as 3-D emissions from aviation and the atmospheric oxidation of other carbon species. The seasonal cycle and spatial distribution of GEOS-Chem $CO_2$ columns were generally comparable with GOSAT columns over each continent with a systematic positive bias of ~1.0%. Data from the World Data Center for Greenhouse Gases (WDCGG) from twelve ground stations spanning $90^{\circ}S-82^{\circ}N$ were also compared with the modeled data for the period of 2004-2009 inclusive. The ground-based data show high correlations with the GEOS-Chem simulation ($0.66{\leq}R^2{\leq}0.99$) but the model data have a negative bias of ~1.0%, which is primarily due to the model initial conditions. Together these two comparisons can be used to infer that GOSAT $CO_2$ retrievals underestimate $CO_2$ column concentration by ~2.0%, as demonstrated in recent validation work using other methods. We further estimated individual source/sink contributions to the global atmospheric $CO_2$ budget and trends through 7 tagged $CO_2$ tracers (fossil fuels, ocean exchanges, biomass burning, biofuel burning, net terrestrial exchange, shipping, aviation, and CO oxidation) over 2004-2009. The global $CO_2$ trend over this period (2.1 ppmv/year) has been mainly driven by fossil fuel combustion and cement production (3.2 ppmv/year), reinforcing the fact that rigorous $CO_2$ reductions from human activities are necessary in order to stabilize atmospheric $CO_2$ levels.
