Oviduct-Specific Enhanced Green Fluorescent Protein Expression in Transgenic Chickens

<P>In this study, we confirmed the ability of the 2-kb promoter fragment of the chicken ovalbumin gene to drive tissue-specific expression of a foreign EGFP gene in chickens. Recombinant lentiviruses containing the EGFP gene were injected into the subgerminal cavity of 539 freshly laid embryos (stage X). Subsequently the embryos were incubated to hatch using phases II and III of the surrogate shell <I>ex vivo</I> culture system. Twenty-four chicks (G0) were hatched and screened for EGFP with PCR. Two chicks were identified as transgenic birds (G1), and these founders were mated with wild-type chickens to generate transgenic progeny. In the generated transgenic hens (G2), EGFP was expressed specifically in the tubular gland of the oviduct. These results show the potential of the chicken ovalbumin promoter for the production of biologically active proteins in egg white.</P>

최근 변화하는 기후로 인해 외래해충의 유입이 증가하였으며 발달된 교통수단 등으로 인해 새로운 지역으로 확산이 가속화되는 추세(Bale, 2002)이므로 그 중요성이 증대하고 있다. 본 연구는 우리나라의 최근에 침입한 대표적인 흡즙성 해충인 미국선녀벌레를 대상으로 충청지역의 발생분포를 파악하기 위해 수행되었다. 이를 위해 2018년 5월부터 8월까지 충남북 관내 주요 행정구역을 대상으로 미국선녀벌레 발생여부와 피해정도를 중심으로 조사하였다.
HDAC11 Inhibits Myoblast Differentiation through Repression of MyoD-Dependent Transcription

Abnormal differentiation of muscle is closely associated with aging (sarcopenia) and diseases such as cancer and type II diabetes. Thus, understanding the mechanisms that regulate muscle differentiation will be useful in the treatment and prevention of these conditions. Protein lysine acetylation and methylation are major post-translational modification mechanisms that regulate key cellular processes. In this study, to elucidate the relationship between myogenic differentiation and protein lysine acetylation/methylation, we performed a PCR array of enzymes related to protein lysine acetylation/methylation during C2C12 myoblast differentiation. Our results indicated that the expression pattern of HDAC11 was substantially increased during myoblast differentiation. Furthermore, ectopic expression of HDAC11 completely inhibited myoblast differentiation, concomitant with reduced expression of key myogenic transcription factors. However, the catalytically inactive mutant of HDAC11 (H142/143A) did not impede myoblast differentiation. In addition, wild-type HDAC11, but not the inactive HDAC11 mutant, suppressed MyoD-induced promoter activities of MEF2C and MYOG (Myogenin), and reduced histone acetylation near the E-boxes, the MyoD binding site, of the MEF2C and MYOG promoters. Collectively, our results indicate that HDAC11 would suppress myoblast differentiation via regulation of MyoD-dependent transcription. These findings suggest that HDAC11 is a novel critical target for controlling myoblast differentiation.

Background: Postinflammatory hyperpigmentation (PIH) is an often encountered troublesome problem after laser treatment, but the risk factors of PIH are not well known until recently. Objectives: We found predicting risks for PIH after treatment of senile lentigo with 532-nm Nd:YAG laser. Methods: Medical records of 392 outpatients aged over 50 years were analyzed. They have been treated for facial senile lentigo with 532-nm Nd:YAG laser in five university hospitals in 2009~2012. All patients have medical records about PIH development and clinical photographs of lentigo and entire face. Data analyzed by using Pearson``s Chi-square test, Fisher’s exact test, or Linear by linear association. Results: Overall PIH incidence was 27 percent. Mean onset of PIH was 3.6 weeks and mean duration was 10.5 weeks. Characteristics of lentigo are most important to arouse PIH. Lentigo with unclear margin, presence of erythema, thickening, and large facial area involvement are all significantly increase PIH risk. Patient aspects as sex and Fitzpatrick’s skin type do not increase risk of PIH. Skin conditions such as dry and lusterless skin and presence of dyschromia significantly increase PIH. Season of laser treatment is not related to PIH risk. Post-procedure management may reduce PIH, but rather pretreatment may increase PIH. Conclusion: Most important things to reducing PIH risk are to carefully observe characteristics of lentigo including margin, erythema, thickening, and involvement extent.
