        로타바이러스 위장관염이 있는 환자에서 로타바이러스 G-P 유전형에 관련된 임상적특성 비교

        배경 : Group A 로타바이러스 감염은 전세계적으로 소아에서일어나는 심한 설사의 주된 원인이다. 로타바이러스 감염증의 적절한 처치를 위해 로타바이러스에 감염된 환자에서 G와 P 유전형에 따른 임상적 특성을 알면 도움이 될 것이다. 방법 : 2002년 3월부터 1년간 조선대학교병원에 입원한 환자의설사변 검체를 대상으로 Dipstick ‘Eiken’Rota kit로 로타바이러스 항원검사를 시행하였다. 로타바이러스 항원검사에 양성인 대변 검체로 group A 로타바이러스에 대한 RT-PCR 검사를 하였고 그에 양성인 PCR 산물로 G 및 P형에 대한 PCR 검사를 시행하여 유전형을 검사하였다. 결과 : 유전형 검사를 시행한 19검체 중 가장 우세한 균주는G4P[6] (51.3%)이었고 그 다음은 G2P[4] (19.3%), G1P[8](7.6%), G3P[8] (5.0%), G9P[8] (4.2%)의 순이었다. 의무기록 분석이 가능하였던 10명의 환자를 대상으로 각 로타바이러스유전형의 특징을 알아보기 위해 임상적 및 역학적 연구를 시행하였다. 로타바이러스는 각 유전형별로 설사, 구토, 탈수, 발열, 병원감염, 다른 병원에서 전원된 환자빈도 및 평균 중증도 점수의 측면에서 유의한 차이를 보였다. 특히 G4P[6] type에 감염된 환자는 다음과 같은 독특한 특징을 나타냈다. 대부분의 환자들이 비교적 미약한 증상을 보이고 다른 산부인과에서 전원된 신생아였으며그들 증례 중 68.4%가 병원감염이었다. G4P[6]형은 유행시기가거의 일년 내내 지속되었다. G4P[6], G2P[4], G1P[8], G3P[8]및 G9P[8] 균주에 감염된 환자가 나타내는 증상의 평균 중증도점수는 각 6.8, 9.5, 8.0, 9.0 및 10.8이었다.

        Identification and Characterization of a Novel Enzyme Related to the Synthesis of PUFAs Derived from Thraustochytrium aureum ATCC 34304

        In Thraustochytrids, Thraustochytrium aureum ATCC 34304 was able to produce high levels of several polyunsaturated fatty acids. In the present study, a novel gene encoding protein was cloned from the DHA rich microbe, T. aureum ATCC 34304. The functional analysis of a novel gene was demonstrated by its heterologous expression in Pichia pastoris. The gene was able to synthesize C20 and C22 PUFAs, as well as, to mediate different elongations (${\Delta}$9, ${\Delta}$6, and ${\Delta}$5) and one ${\Delta}$5 desaturation activities. The conversion rates of the ${\Delta}$9 elongation (n-3) and ${\Delta}$5 desaturation products were found to be higher in response to the novel enzyme than the controls (TaElo and Tad5, respectively). The other ${\Delta}$9 elongation (n-6) and ${\Delta}$5 elongation products were slightly lower than those of the control (TaElo). The full length of the 1,374 bp gene contained 458 amino acids that showed very limited homology with desaturases and elongases from various organisms. In addition, the rate of synthesis of PUFAs was evaluated at temperatures ranging from 10 to $30^{\circ}C$. The elongation products were found to decrease dramatically and the desaturation products were found to increase dramatically at $10^{\circ}C$. TaNE was confirmed to be a multifunctional enzyme with higher activity towards ${\Delta}$6 elongations than ${\Delta}$9, ${\Delta}$5 elongations, and ${\Delta}$5 desaturation.

        The effects of culture condition on the growth property and docosahexaenoic acid production from Thraustochytrium aureum ATCC 34304

        The effect of the composition of artificial sea water (ASW) medium on the growth properties and docosahexaenoic acid (DHA) production from Thraustochytrium aureum ATCC 34304 was investigated. A maximum dry cell weight (DCW) of 3.71 g/l was obtained when the NaCl and MgSO4 concentration in the ASW media were 15 and 0 g/l, respectively. The maximum initial specific growth (ISG) rate of 0.16 was attained at 7.5 g/l NaCl and 18 g/l MgSO4,while the minimum ISG rate (0.02) was obtained at 5.0 g/l NaCl and 4.5 g/l MgSO4. The least doubling time required for biomass production was 4.3 h at 7.5 g/l NaCl and different MgSO4 concentrations. A maximum of 7.9 g/l DCW was obtained on the fourth day of cultivation at 30 g/l glucose and 2.5 g/l (each) yeast extract (YE) and peptone. The DHA content in the lipids was significantly affected by the concentration of glucose and nitrogen sources (YE and peptone) in the ASW medium. At the lowest glucose (10 g/l) and YE/peptone (0.5 g/l) concentration and highest glucose (30 g/l) and YE/peptone (2.5 g/l) concentration, the DHA content was 34.725 and 40.33%, respectively, relative to total lipid content. However, the DHA content in the lipid was not affected by the NaCl and MgSO4 concentration. At the lowest NaCl (2.5 g/l) and MgSO4 (4.5 g/l) concentration and highest NaCl (60 g/l) and MgSO4 (18 g/l) concentration the DHA content was 39.62 and 38.48%, respectively. The maximum DHA content in the lipid was 49.01% after four days of cultivation when 7.5 g/l NaCl and 4.5 g/l MgSO4 were in the ASW medium. The growth properties of T. aureum ATCC 34304 for biomass production and DHA yield in the lipid content were found to be affected by NaCl and glucose concentration.
