Chip curling occurred during cutting process for difficult-to-cut material detracts product qualities and productivity. Among of method preventing the phenomenon, high pressure injecting cutting oil is an alterative. In this study, the optimal nozzle was designed by CFD method and it was conducted to analyse on the effect of high pressure injection on chip shape generated during cutting process and wear of insert by experimental method. As the result, it could be confirmed that high pressure injection is favorable for preventing chip curling and insert from wearing.
Production of Interferon-$\gamma$ from Human Peripheral Blood Lymphocytes

인체혈액에서 분리한 임파구세포를 PHA-M, Con A, PWM등의 mitogen으로 자극시켜 여러가지 조건하에서 감마-인터페론을 생산하였다. 유도물질로 사용된 T-세포 mitogen인 PHA-M이나 Con A가 B-세포 mitogen 인 PWM보다 감마-인터페론 유도에 훨씬 효율적이었으며 이로부터 임파구세포 중에서 T-세포가 감마-인터페론을 생산하는 주요세포원 임을 확인하였다. 감마-인터페론 생산시 Con A의 적정농도는 $5-10\;{\mu}g/ml$이었다. 유도개시 약 48시간 경과 후 인터페론이 최대로 생산되었고 그 이후에도 인터페론의 활성은 감소되지 않았다. Phorbolester인 TPA를 첨가할 경우 Con A 또는 PHA-M 단독으로 인터페론을 유도시킨 것보다 10배 이상의 감마-인터페론의 생산이 증가되었다. 생산배양액에 첨가되는 혈청의 농도가 인터페론의 생산에 마치는 영향은 매우 극적이어서 태아 송아지 혈청농도 2%에서 최대로 생산되었고 농도가 증가함에 따라 생산역가는 급격히 떨어졌다. 혈청 대신 사용한 알부민 단백질도 비슷한 효과를 나타냈다. 혈액중에 임파구세포에 처리된 Con A가 DNA 합성을 촉진시키는 성질을 이용하여 $^3H$-thymidine 표식으로 cpm을 측정해 본 결과, 이와같은 현상은 10-20%의 높은 혈청농도에서 Con A가 과량의 혈청단백질과 흡착하여 실제적인 Con A 농도를 적정농도 이하로 떨어뜨리기 때문인 것으로 나타났다. Interferon-$\gamma$(IFN-$\gamma$) was produced from human peripheral blood lymphocytes (Hu-PBL) by stimulating them with various kinds of mitogens. T-cell mitogens such as Con A and PHA-M were much more potent than B-cell mitogen such as PWM in producing IFN-$\gamma$ from Hu-PBL. The combined treatment of TP A with these mitogens enhanced IFN-$\gamma$ production ten fold compared with the single treatment of each mitogen. Both DNA synthesis and IFN-$\gamma$ production yielded bell-shaped dose-response curves for a given population of Hu-PBL in response to Con A. The effect of fetal calf serum(FCS) on IFN-$\gamma$ production was found to be critical: titer of IFN-$\gamma$ produced was very low in the absence of FCS and reached peak point at 2% FCS, beginning to decrease precipitously with an increasing FCS concentration up to 20%. Using the relationship between IFN-$\gamma$ production and mitogenic stimulation, it was found that one of factors responsible for the decrease of IFN-$\gamma$ production at the high serum concentration could be the binding of Con A to serum proteins which rendered Con A to be removed from the production system.

This paper investigated the rhythmic features of speakers of English as a lingua franca in Asian contexts. A speech corpus of 150 conversations between speakers of English in Asia with different L1 backgrounds was collected and %V, ΔV, VarcoV, and nPVI-V of each speaker were analyzed. It was found that L1 difference of the speakers and the speakers’ daily use of English influenced %V, while the speakers’ daily use of English influenced ΔV. The gender difference of the speakers also affected the rhythm of the utterances in VarcoV. A weak correlation between the two speakers’ rhythm in each conversation was also found in %V and ΔV. No significant effects were found in nPVI-V. The results revealed that the speakers tended to accommodate the rhythm of their utterance to that of the interlocutors’. Further study on the speaking rate of the speakers is required to overcome some inconsistencies found in the results of the rhythmic metrics used in this study.
생물방제 및 생물비료 활성을 가지는 세균의 분리 및 특성

다기능성 농업용 미생물 제제를 개발하기 위하여 생물방제 및 생물비료 활성을 가지는 미생물을 탐색하였다. 본 연구실에서 분리 및 동정된 균주가운데 Pantoea agglomerans 및 Bacillus megaterium을 실험균주로 선정하였으며, 경남 밀양에 위치하는 양계장 부근 부엽토로부터 새로운 다목적 세균 MF12를 분리하였다. 형태학적, 배양적, 생화학적 특성 및 16S rDNA 염기서열을 분석한 결과, MF12는 Bacillus pumilis로 동정되었다. 이 균주들의 불용성 인산 가용능, IAA 및 siderophore 생성능, ammonification ability, 식물병원성 진균 세포성분 분해효소 생성능 및 항진균능을 조사하였다. P. agglomerans는 고체배지에서 불용성 인산을 가용화할 수 없었으나 액체배지에서는 가용성 인산을 생성하였다. 상기 모든 균주들은 배양시간에 따라 3~639 ㎍/ml의 IAA를 생성하였으며, P. agglomerans만이 siderophore를 생성하였다. 이 균주는 pectinase와 lipase를 생성하였다. B. megaterium은 amylase, protease 및 lipase를 생성한 반면 B. pumilis는 protease와 lipase를 생성하였다. P. agglomerans는 Fusarium oxysporum과 Colletotrichum gloeosporioides의 생육을 억제하였으며, B. pumilis는 Botrytis cinerea, Sclerotinia sclerotiorum 및 Phythium ultimum의 생육을 억제하였다. To develop multifunctional microbial inoculant, microorganisms with antagonistic activity and biofertilizing activity were screened. Pantoea agglomerans and Bacillus megaterium from our laboratory culture collection, and strain MF12 from soil near poultry farm in Miryang were selected. On the basis of morphological, physiological studies and 16S rDNA sequence analysis, isolate MF12 was identified as the Bacillus pumilis. Three strains were studied for insoluble phosphate solubilization, indole-3-acetic acid (IAA) and siderophore production, ammonification ability, hydrolytic enzyme production and antifungal activity against phytopathogenic fungi. P. agglomerans did not produce any visible clear zone on agar plate containing 0.5% Ca₃(PO₄)₂as a sole phosphorus source. However, this strain could solubilize insoluble phosphate in liquid medium. All strains produced IAA ranged from 3~639 ㎍/ml depending on culture time and had ammonification ability. Among three strains, only P. agglomerans produced siderophore. P. agglomerans produced pectinase and lipase, B. megaterium produced amylase, protease and lipase while B. pumilis produced protease and lipase. P. agglomerans showed antifungal activities against phytopathogenic fungi, Fusarium oxysporum and Colletotrichum gloeosporioides. B. pumilis showed antifungal activities against Botrytis cinerea, Sclerotinia sclerotiorum and Phythium ultimum.
