To develop a HPV-18 oral vaccine, HPV-18 E6 gene was cloned into an expression vector, under the control of CaMV 35S dual promoter and 5`UT region of TEV. The constructed vector (pUS-E6) was used to transform tobacco plants by the Agrobacterium-mediated leaf disc method. The transformants were then regenerated to whole transgenic plants. The HPV-18 E6 gene was found to be expressed in the transgenic tobacco plants as determined by PCR, RT-PCR, Northern analysis and Western analysis. These results show a possibility that HPV oral vaccine may be developed by expressing the HPV proteins in tomato or other vegetables.

담배에서 human papil1omavirus-18형 E6(HPV-18 E6) oncogene을 발현시키기 위하여 E6 유전자를 Cauliflower mosaic virus(CaMV)의 35S dual promoter와 tobacco etch virus(TEV) 5'-untranslated region의 조절을 받도록 벡터를 제조한 후, Agrobacterium-mediated leaf disc 방법으로 담배에 도입하였다. Kanamycin 저항성을 갖는 형질전환체를 선별하고, 완전한 식물체로 재생한 형질전환체의 잎조직에 대하여 PCR 방법과 Northem blot, Westem blot 분석을 한 결과, HPV-18 E6 유전자가 형질전환체 담배에서 성공적으로 발현됨을 알 수 있었다. 서울에는 지상 60층이 넘는 초고층 건물과 지하 8층 이상의 초심층 건축물, 지하철과 상가, 백화점, 호텔, 전 시장등이 연결된 대규모 복합유통시설 지역 등이 있다. 이런 지역이나 시설에서 화재와 같은 재난사고가 발생시 진압 작전을 위해 투입된 소방관의 위치를 제대로 파악할 수 없으면 투입된 소방관의 안전은 물론 효율적인 지휘통제가 제한되어 결국 더 많은 자체 피해와 더불어 화재 진압 및 시민구조에 제한을 받을 수밖에 없다. 이러한 대형재난에 유비쿼터스 기술을 활용하여 투입한 소방력의 효율적인 운영과 과학적인 진압작전을 구사하여 시민의 생명과 재산을 최대한 보호할 수 있는 소방대응지원 시스템 구축방안을 제시하였다. In Seoul, there are lots of skyscrapers that have above 60 stories and buildings that have more than 8 basement levels, as well as massive distribution complex region which is connected to subways, departments, malls, hotels, and exhibition halls. When an accident, such as fire and explosion, happens in these areas or structures, if we can't find where fire-fighters are, who go into the building to suppress the fire, we couldn’t be sure of their safety as well as effective command. Actually, it may cause much more damage itself and restrict either fire suppression or lifesaving. To protect people’s life and properties as much as possible, this study will show the method to construct system of disaster-management supports with effective operation of fire force and scientific fire strategy in the scene by using Ubiquitous technique to enormous disasters.

Novel Promoter Polymorphism in RUNX2 Is Associated with Serum Triglyceride Level

Much research evidence supports the hypothesis that chronic, low-grade inflammation related to innate immunity may play an important role in the pathophysiology of type 2 diabetes mellitus (T2DM). Runt-related transcription factor 2 (RUNX2; MIM# 600211) acts as a scaffold that controls the integration, organization, and assembly of nucleic acids. To examine whether the novel promoter variant in orkuO is associated with the risk of T2DM and related phenotypes, RUNX2-742G >T was genotyped in 378 T2DM patients and 382 normal controls recruited in the Korean T2DM Study. Statistical analysis revealed that RUNX2-742G >T was associated with serum triglyceride level (TG) in nondiabetic controls, although it was not associated with the risk of T2DM. Individuals who carry T/T, T/G, and G/G genotypes had the highest (2.061 ± 0.20), intermediate (2.01 ± 0.19), and the lowest (1.97 ± 0.18) levels of log [TG (mmol/l)] (P = 0.007), respectively. Our data on this important variant of orkuO suggest that lipid metabolism might be affected by genetic polymorphisms in the promoter region.
Chromosomal information of 1,144 Korean BAC clones

We sequenced 1,841 BAC clones by terminal sequencing, and 1,830 of these clones were characterized with regard to their human chromosomal location and gene content using Korean BAC library constructed at the Korean Science (KCGS). Sequence analyses of the 1,830 BAC 1,144 clones were assigned to a single chromosome, 190 clones apparently assigned to more than one chromosome, and 496 clones to no chromosome. Evaluating gene content of the 1,144 BAC clones, we found that 706 clones represented 1,069 genes of which 415 genes existed in the BAC clones covering the ful sequence of the gene, 180 genes covering a 50%~99%, and 474 genes covering less than 50% of the gene coverage. The estimated covering size of the KBAC clones was 73,379 kilobases (kb), in total coresponding to 2.3% of haploid human genome sequence. The identified BAC clones will be a public genomic resource for mapped clones for diagnostic and functional studies by Korean scientists and investigators worldwide.
