        약물-고분자물질 결합체 합성연구 : 5-Fluorouracil과 사람의 혈청 Albumin 및 Poly-L-lysine 결합체 합성

        The durg-macromolecule conjugates i.e. 5-fluorouracil-1-acetyl-human serum albumin(FU-AA-HSA, 8) and 5-fluorouracil-1-acetyl-poly-L-lysine(FU-AA-polylys, 9) have been synthesized and purified by sephadex G-25 gel filtration with 0.05M phosphate buffer(pH 7.5). The analyses of conjugates gave the molar ratio of FU-AA : HSA of 70-100:1 and FU-AA: poly-L-lysine of 109:1. The weight percent of FU-AA(as $FU-CH_2CO-$) in FU-AA-HSA conjugate was 16-22% and the one in FU-AA-polylys was 22.4%.

        9 - β - D - Arabinofuranosyladenine - 5 ' - methoxyacetate 및 1 - β - D - arabinofuranosylcytosine - 5 ' - methoxyacetate 의 합성 및 Sarcom 180 암에 대한 항암작용

        9-β-D-Arabinofuranosyladenine-5`-methoxyacetate (ara-A-MA) and 1-β-D-arabinofuranosylcytosine-5`-methoxyacetate (ara-C-MA) were synthesized. Their antitumor activities were evaluated against sarcoma 180 tumor cells inoculated in mice. When administered intraperitoneally once daily for 6 days, average life-spans for those which received ara-A-MA, ara-C-MA, ara-C and saline (0.9%) were; 20.8±2.9(19.4 ㎎/㎏/mouse) and 19.1±2.5 (1.8 ㎎/㎏/mouse) days; 23.1±3.3(18.0 ㎎/㎏/mouse) and 20.1±2.2(1.8 ㎎/㎏/mouse) days; 22.0±2.5(13.9 ㎎/㎏/mouse) days; and 16.3±4.1 days, respectively. This survival study indicates that ara-A-MA and ara-C-MA exhibit antitumor activities as much potent as their parent compounds of ara-C. DNA biosynthesis in Sarcoma 180 cells in vitro were also inhibited by ara-A-MA. At the concentrations of 10^(-3) and 10^(-4) M, ara-A-MA and ara-C-MA showed 57 and 50%, 94 and 80% inhibitions of DNA synthesis in comparison with that of negative control group, respectively.

        안지오텐신 변환효소(變換酵素) 억제(抑制) 작용(作用) 물질(物質)의 합성(合成)

        Phenylurea, phenylthiourea, benzylcarbamate and toluenesulfonate of L-alanyl-L-proline(comp. $3{\sim}6$) were synthesized and their effects against angiotensin-converting enzyme (ACE) were tested. Comp. 3 showed only mild inhibitory activities against ACE while comp. $4{\sim}6$ were inert indicating that those functional groups were not suitable for interactions with ACE. Ortho-hydroxy- or ortho-carboxy-benzamide of proline (comp. 7) and phenylalanine (comp. 8 and 9) were also tested. Of the benzamides, ortho-hydroxy function was unsuitable to exert inhibitory activities against ACE. Ortho-carboxy group of 9 seemed to have mild interactions with active site of ACE possibly because of the shorter distance between the amide and ortho-carboxyl group of the compound than the corresponding two active sites of the enzyme.

        Foliar application of biostimulants affects physiological responses and improves heat stress tolerance in Kimchi cabbage

        This study investigated the effects of foliar applications, such as brassinosteroid (BR), methyl jasmonate (JA), salicylic acid (SA), and sodium nitroprusside (SNP), on the growth, yield, photosynthetic characteristics, and antioxidant enzyme activity of Kimchi cabbage (Brassica rapa L. ssp. pekinensis) exposed to heat stress. Kimchi cabbages were grown in extreme weather growth chambers under controlled conditions (air temperature, relative humidity, and solar radiation of the Daegwanryeong region, 37° 40′ N, 128° 32′ E). At 18 days after transplanting, Kimchi cabbages were treated with foliar application of biostimulants (vs. non-sprayed control) and exposed to heat stress (air temperature 30/25 °C) and mean daily air temperature under normal conditions (21.2 ± 1.6 °C day/night, respectively) for 5 days. The foliar application concentrations were 50, 100, and 200 nM BR; 50, 100, and 200 μM JA; 1, 2, and 4 mM SA; and 0.25, 0.50, and 1.00 mM SNP. At 38 days after foliar application, the shoot fresh weight of Kimchi cabbages treated with 200 nM BR was 2123 versus 1422 g plant−1 (control). The head weight and yield were the greatest with 2 mM SA treatment, which produced 3806 versus 2184 kg ha−1 (control). Heat stress induced a low photosynthetic rate in Kimchi cabbage, while foliar application of biostimulants enhanced photosynthesis recovery. During the 3 days after heat stress, catalase and peroxidase enzyme activities of Kimchi cabbage increased by 1.76- to 2.08-fold on average compared to the control. Results indicated that foliar application of biostimulants reduced physiological damage and enhanced the activity of the antioxidant enzymes, thereby improving heat stress tolerance in Kimchi cabbage.

        9-$\beta$-D-Arabinofuranosyladenine-5'-methoxyacetate (ara-A-MA) 및 1-$\beta$-D-Arabinofuranosylcytosine-5'-methoxyacetate (ara-C-MA)를 합성하였다. 이들 물질의 항암작용을 검토한 바, sarcoma 180 암세포를 이식한 mice중 생리식염수만 공급 받은 negative 대조군은 생존일수가 $16.3{\pm}4.1$일이었고, positive 대조군인 ara-C(13.9 mg/kg/mouse)를 투여 받은 실험군은 생존일수가 $22.0{\pm}2.5$ 일이었다. ara-A-MA를 투여 받은 시험군은 $20.8{\pm}2.9$일(19.4 mg/kg/mouse) 이었고, ara-C-MA를 투여 받은 실험군은 $23.1{\pm}3.3$일(18.0 mg/kg/mouse) 및 $20.1{\pm}2.2$일(1.8 mg/kg/mouse)의 생존일수를 각각 나타내었다. 한편 이들 합성 화합물은 DNA 합성을 억제하는 작용을 나타내었는데, ara-A-MA는 Fischers media에 배양한 sarcoma 180 암세포의 DNA 생합성을 $10^{-3}\;M$ 농도에는 57% 그리고 $10^{-4}\;M$에서 50%를 억제하고, ara-C-MA는 94% 및 80%를 각각의 농도에서 억제시켰다. 이들 결과는 ara-A-MA 및 ara-C-MA가 ara-A 및 ara-C의 prodrug으로서 역할을 할 수 있는 가능성을 시사한다. 9-$\beta$-D-Arabinofuranosyladenine-5'-methoxyacetate (ara-A-MA) and 1-$\beta$-D-Arabinofuranosylcytosine-5'-methoxyacetate (ara-C-MA) were synthesized.         여주의 유전자원별 과실특성과 Charantin 성분 함량

        국내에 보유하고 있는 여주 유전자원을 대상으로 과실특성과 charantin 함량을 분석하고, 추후 유전자원을 다양한 용도로 활용하고자 수행하였다. 2010년에 선발된 수량이 높은 계통 2점의 charantin 함량을 HPLC 분석한 결과, 착과 후 일수에 따른 함량의 변이를 보였는데, BG1은 착과 후 24일 15일순으로 charantin 함량이 높게 나타나 착과 후반부에 charantin 함량이 높게 나타난 반면 BG7은 착과 후 일수가 13일에 가장 높았고, 16일 19일 순으로 charantin 함량이 높게 나타나 착과 전반부에 charantin 함량이 높아지는 것으로 나타났다. Charantin 성분 함량이 높은 여주품종 선발 및 수확시기 규명을 위하여 한국, 중국, 인도, 일본 등에서 도입한 유전자원 33점과 시판종 1점중 수량이 높은 13점의 과실특성 및 charantin 함량을 HPLC 분석한 결과 charantin 함량이 높은 104615, K169995, 엔에스454을 선발하였다. 따라서 수량이 높은 13점의 여주자원중 charantin 함량이 높았던 K169995나 NS454은 새로운 품종 육성이나 가공용으로 활용할 수 있으며, 수량이 많았던 K161952나 K051434 등은 과실 생산 목적으로 활용할 수 있을 것으로 생각되고, 착과후 시기별로 용도에 따라 수확을 하여 이용하면 좋을 것으로 생각된다. Bitter gourd (Momordica charantia L.) has long been used for food and medicinal plant in Korea, China and Japan. This study aimed at evaluating productivity, and vitamin-C and charantin contents in bitter gourd (Momordica charantia L.) accessions. The contents of charantin of these two accessions were analyzed using HPLC with the UV-diode array detection. The highest fruit yield was observed in accessions, ‘BG1’ and ‘BG7.’ The vitamin-C contents of fruits in these two high-yield bitter gourd accessions, ‘BG1’ and ‘BG7,’ depended on days after fruit set and were highest in 24 days and 17 days after fruit set, respectively. The charantin contents of the two accessions were different according to the number of days after fruit set. The charantin content of ‘BG1’ was highest in fruits harvested at 24 days and followed by 15 days after fruit set. The charantin content of ‘BG7’ was highest in fruits harvested at 13 days and followed by 16 and 19 days after fruit set. The charantin contents of 13 M. charantia accessions with relatively high yield potential were analyzed and three accessions, 104615, K169995 and NS454, were selected based on their relatively high yield and charantin content. These accessions will be used for breeding program and processed foods.

        수세미 오이의 항산화 효소 및 항균 활성 검증

        수세미오이(Luffa cynlindrica)는 박과 채소작물로 우리나라에서는 주로 관상용으로 재배되어 왔다. 그러나 최근에는 미용소재, 한방약재, 건강보조용 등으로 수요가 증가하면서 재배면적이 증가하고 있다. 따라서 본 실험은 수세미오이의 기능성을 구명하기 위하여 재래종 수세미오이를 사용하여 수액의 무기성분을 분석하고 수액과 식물체 부위별 항산화효소 활성과 항균활성을 측정하였다. 수세미오이 수액은 K, Ca, P, Mg이 주요 성분이었으며, K의 함량이 470㎎·L<SUP>-1</SUP>으로 가장 높았다. Ca과 Mg의 함량은 오이의 약 2배와 1.7배로 높게 나타났다. 수세미오이의 조직별 항산화효소 활성을 분석한 결과 전체적으로 APX 활성이 CAT와 SOD 활성보다 높았다. 식물체 부위별로 비교해 볼 때 항산화효소 활성은 잎, 뿌리, 꽃, 과실, 종자, 수액보다 줄기에서 가장 낮게 나타났다. 수세미오이 수액의 식품유해균 10종에 대한 항균활성은 Vibrio parahaemdyticus와 Propionibacterium acne에 대하여 미미한 활성을 보였을 뿐 그 외 8종의 균에 대해서는 효과가 없었다. 피부염 등 인체 감염을 일으키는 Candida albicans와 Malassezia furfur에 대한 수세미오이의 부위별 항균활성 분석 결과, 수액의 항균활성이 식물체의 다른 부위보다 높게 나타났다. 전체적으로 Malassezia furfur에 대한 항균활성이 Candida albicans에 대한 항균활성보다 높은 것으로 나타났다. Luffa cylindrica Roem (sponge gourd) belongs to the Cucurbitaceae family and has been cultivated as an ornamental plant in Korea. Recently, its cultivation area has been increased with an increase in demand for it as an ingredient in cosmetics, herbal medicines, and health supplements. We analyzed inorganic components of sap collected from land race sponge gourds. We also measured antioxidant enzyme activities and antimicrobial activities of the plant, seed, and sap to examine functional properties of sponge gourd. The sap of the sponge gourd contained high levels of K, Ca, P, and Mg, with the most abundant mineral in the sap being K (470 ㎎·L<SUP>-1</SUP>). The amounts of Ca and Mg were 2 and 1.7 times more than those found in cucumber (Cucumis sativus), respectively. Ascorbate peroxidase was more active than catalase and superoxide dismutase in various plant parts of sponge gourd. Antioxidant activities were much lower in stems than in other plant parts such as leaves, roots, flowers, fruits, seeds, and sap. In addition, sap showed a very low level of antimicrobial activity against two food-borne pathogens, Vibrio parahaemdyticus and Propionibacterium acne, and none against the other eight tested food-borne pathogens. Antimicrobial activities against Candida albicans and Malassezia furfur, which causes dermatitis, appeared to be higher in sap than in other parts of sponge gourd plants. Overall, the antimicrobial activity against Malassezia furfur appeared to be higher than against Candida albicans.
