        합성 올리고데옥시뉴클레오타이드의 피부투과

        Antisense phosphorothioate oligodeoxynucleotide(PS-ODN) against $TGF-{\beta}$ was developed as scar formation inhibitor. The scar was caused collagen deposition due to overexpression of $TGF-{\beta}$ in wounded skin. The percutaneous absorption of partially modified PS-ODN(25 mer) was investigated for the purpose of its effective delivery. Though PS-ODN has high molecular weight (MW=8,000) and polyanionic charge, it was permeated through skin. The skin permeation of PS-ODN was markedly increased by the removal of stratum corneum and dermis. Moreover, the skin permeation of PS-ODN was decreased in the following order; hairless mouse skin>rat skin>human cadaver skin. Thus, PS-ODN represents a logical candidate for further evalution due to the potential for delivery into the wounded skin.

        High-Resolution X-Ray Photoelectron Spectroscopy Study of a Sb<sub>2</sub>Te<sub>3</sub> Thin Film with the Polycrystalline Phase

        스퍼터를 이용하여 실리콘 기판위에 제작된 안티몬-테레니움 다결정 박막을 방사광을 이용한 고해상도 엑스선 광전자 분광법 실험을 수행하여 화학적 상태를 분석하였다. 엑스선 회절 실험을 통해 제작된 안티몬-테레니움 박막은 롬보헤드럴 구조를 가지는 다결정임을 확인하였다. 엑스선 광전자분광법을 수행하기 위하여 표면의 산화막 제거를 위해 저에너지 네온 이온 스퍼터링을 빔에너지 1 kV로 1 시간동안 수행하였고, 이를 통해 표면 산화막이 완벽히 제거됨을 확인하였다. 또한, 스퍼처링에 의하여 표면 비정질화된 상태를 결정화 상태로 만들기 위해 상변화온도인 $100^{\circ}C$에서 5 분간 초고진공상태에서 열처리를 수행하였다. 이후 획득되어진 테레니움 4d와 안티몬 4d 속전자레벨 분석에서 각각의 묶음에너지가 40.4 그리고 33.0 전자볼트임을 확인할 수 있었으며, 각각은 단일한 화학적 상태를 나타내고 얻어진 피크의 밀도분석을 통해 화학적조성비가 2 : 3임을 확인하였다. We investigated chemical states of a $Sb_2Te_3$ thin film with the polycrystalline phase by using high-resolution x-ray photoelectron spectroscopy with synchrotron radiation. The $Sb_2Te_3$ thin film was formed by sputtering. The rhombohedral phase was confirmed by x-ray diffraction. To remove the surface oxide, we performed $Ne^+$ ion sputtering for 1 hour with the beam energy of 1 kV and post-annealing at $100^{\circ}C$ for 5 min in ultra-high vacuum. We obtained the Te and Sb 4d core-levels spectra with the peaks at the binding energies of 40.4 and 33.0 eV, respectively. The full-width of half maximum of both the Te and Sb $4d_{5/2}$ core-levels is 0.9 eV. The Te and Sb core-levels only show a single chemical state, and we also confirmed the stoichiometry of approximately 2 : 3.

      • Escherichia coli M1 RNA 유전자의 전사 종결

        Escherichia coli RNase P의 RNA 성분인 M1 RNA를 코드하는 유전자에 연속적으로 존재하는 종결제 T1, T2, T3의 효율을 측정하였다. 이를 위하여 M1 RNA 유전자의 3'-부분에 있는 염기배열을 차례로 결실시키고, 결실된 염기배열을 갖는 M1 RNA 유전자를 Gal K 발현 벡타인 pKO100에 클로닝하여 갈락토키나제 활성도를 측정하였다. 주된 종결반응은 첫번째 종결제인 T1에서 94%의 효율로 일어났으며, 세 개의 종결제가 모두 존재할 때는 거의 100% 효율로 종결되었다. 또한 이러한 종결제를 통과하여 전사된 RNA가 가지는 ORF (open reading frame)들에서 만들어 질 수 있는 폴리펩티드에 대해서도 검토하였다. The deletion experiment at the 3´flanking sequences of the gene for M1 RNA, the RNA component of Echerichia coli ribonuclease P, was carried out to estimate the efficiency of tandem terminators, T1, T2, and T3. The serially deleted sequences of the M1 RNA gene were cloned into GalK expression vector pK0100 and relative galactokinase activities were measured. The major termination occurred at the first terminator, T1, and three terminators all together functioned with almost 100% efficiency. Biosynthesis of the polypeptides encoded by readthrough transcripts from the terminators was also discussed.

        콩 근권의 핵심 세균 군집

        콩은 우리나라와 극동아시아가 원산지로 알려져 있으나 국산 콩의 근권 세균 군집에 대한 연구는 미흡하다. 따라서 본 연구에서는 국산 재배콩을 대상으로 차세대 염기서열 분석 방법인 파이로시퀀싱 방법을 사용하여 콩 근권 세균 군집 구조를 해석하고 생육단계별 군집의 변화 및 콩 근권의 핵심 세균 군집을 구명하고자 하였다. 세균 군집 분석 결과, 근권 세균의 군집은 근권과 비근권간에 뚜렷한 차이를 보였으며, 총 21개의 문으로 구성되었다. Proteobacteria가 가장 우점(36.6-42.5%)하였고, Acidobacteria (8.6-9.4%), Bacteroidetes (6.1-10.9%), Actinobacteria (6.4-9.8%), Firmicutes (5.7-6.3%) 등의 순으로 상대풍부도가 감소하였다. 모든 생육단계에 걸쳐 콩 근권의 핵심 세균 군집에는 Proteobacteria에 속한 OTU들이 가장 많이 분포하였으며, 이들 중 Bradyrhizobium에 속한 OTU의 상대 풍부도가 가장 높았다. 본 연구결과는 콩 근권의 핵심 세균 군집은 주로 생육 촉진 기능과 유기물 순환에 관련된 OTU로 구성되어 있다는 것을 보여주었다. Soybean is well known to be originated from Korea and far-east Asian countries, and studies of many root nodule bacteria associated with soybean have mainly-focused on nitrogen fixation, but much less study was carried out on bacterial community in the rhizosphere of soybean. In this study, we analyzed the bacterial community in rhizosphere of Korean soybean, Daepungkong using the pyrosequencing method based on the 16S rRNA gene to characterize the change of the rhizosphere community structure according to the growth stages of soybeans and to elucidate bacterial core community in rhizosphere of soybean. Our results revealed that bacterial community of rhizosphere soil differed from that of bulk soil and was composed of a total of 21 bacterial phyla. The predominant phylum in the rhizosphere of soybean was Proteobacteria (36.6-42.5%) and followed by Acidobacteria (8.6-9.4%), Bacteroidetes (6.1-10.9%), Actinobacteria (6.4-9.8%), and Firmicutes (5.7-6.3%). The bacterial core community in soybean rhizosphere was mainly composed of the operational taxonomic units (OTUs) belonging to the phylum Proteobacteria throughout all growth stages. The OTU00006 belonged to the genus Bradyrhizobium had the highest abundance and Steroidobacter, Streptomyces, Devosia were followed. These results show that bacterial core community in soybean rhizosphere was mainly composed of OTUs associated with plant growth promotion and nutrient cycles.

        들뢰즈의 "주름 : Le Pli"을 통해 본 바로크 건축의 특성

        The primary object of this study lies in reilluminating the meaning of the Baroque, through studying arts and architecture by means of the features of Baroque aesthetics suggested by Deleuze in his writing 'Le Pli' on the basis of Leibniz's 'Pli' concept. Through this study we can see the following. In the first place, the six features - pli, separation between the inner and the exterior, highs and lows, unfolding, texture and paradigm presented as the features of Baroque aesthetics - have to be understood not as being independent features but as those derived from a series of continuous process of folding and unfolding. In the second place, Deleuze's thinking ranging from the 'Pli' of the Baroque to the paradigm derived on the ground of Leibniz thinking is a dynamic world of folding and unfolding, ultimately seeking unity through the topological changes and succession repeating deterritorization and reterritorization. In the third place, most architectural trends claiming 'Fold Architect' or the 'Pli' of Deleuze as their architectural philosophy are committing errors of mass-producing standardized designs by confining the meaning of Deleuze's 'Pli' concept into a simple reproduction of plied superficial things. With this finding in mind, the concept of folding and unfolding examined through reilluminating the Baroque Architecture where the 'Pli' concept is embodied, suggests to us that the problems of contemporary architecture should be rightened, and shows us that it can become an architectural philosophy which can be concretely realized in architecture.
