Pelvic actinomycosis is a chronic, progressive, granulomatous and suppurative disease caused by an anaerobic or microaerobic Gram-positive organism, not fungi. Actinomyces species exhibit branching, filamentous growth and appear in the human skin, oral cavity, gastrointestinal tract. Actinomyces infection in human is relatively rare, however, prolonged use of an intrauterine contraceptive device (IUD) is a well known risk factor of pelvic actinomycosis. Pelvic actinomycosis can mimick pelvic malignancy leading to mutilating surgical excision, and diagnostic problems necessitated a laparotomy in many patients. The pathohistological diagnosis is based on the characteristic microscopic image and specific staining of sulfur granule. Adequate surgical excision and administration of antibiotics show good prognosis. We experienced 2 cases of pelvic actinomycosis with the intestinal obstruction, confirmed pathohistologically after laparotomy.

This study selected the theme street as a destination for Hyanggyo-ro, which accelerated the downturn after the relocation of the Gyeonggi-do office was decided. Currently, there is no suitable space for the COVID-19 situation at this destination. Therefore, acceleration of decline is inevitable because users are reluctant to visit. In order to overcome this situation, the government intends to select a notice that allows distancing and seek ways to utilize it to promote the activation of streets.
OVCAR-3와 SKOV-3 난소암 세포주에서 Flow Cytometry를 이용한 Apoptosis와 p53 단백의 측정

OVCAR-3와 SKOV-3 난소암 세포주에서 taxol 투여에 의해 유발된 고사 (apoptosis)의 연구를 통하여 두가지 세포주들 사이의 p53의 발현과 세포사의 생물학적 기전을 알아보고자 본 연구를 수행하였다. RPMI 1640 배양액으로 배양한 OVCAR-3와 SKOV-3 난소암 세포주를 taxol (paclitaxel)을 투여하지 않은 대조군과 taxol을 24시간 및 48시간 동안 투여한 투여군으로 나누어 실험을 진행하였다. 수확한 세포들을 먼저 annexin V-FITC (fluorescein isothiocyanate)와 anti-cytokeratin 항체인 clone CAM5.2와 clone MNF116과 반응시킨 다음 세포들을 세척하고 p53 항체를 가하여 반응시켰다. 세포들을 세척한 후 FITC와 공유 결합된 goat anti-mouse (GAM) immunoglobulin G (IgG)-FITC (이하 GAM IgG-FITC) 및 GAM IgG-phycoerythrin (PE) 이차 항체와 반응시켜 염색하였으며, 이어서 propidium iodide (이하 PI)를 이용하여 DNA를 염색하였다. 세포들의 분석에는 488 ㎚의 레이저가 장착된 표준 FACScan flow cytometer로 측정하였다. Taxol을 투여한 후 SKOV-3와 OVCAR-3 세포주 모두에서 G_2M에 세포들이 정지된 것으로 보아 정지된 세포들이 궁극적으로는 세포사 과정으로 진입할 것으로 생각되었다. Cytokeratin과 annexin V 및 G_0G_1 전분기 분획으로 표현된 고사 분획의 출현이 OVCAR-3 세포주에 비해 SKOV-3 세포주에서 낮은 것은 SKOV-3 세포주의 taxol에 대한 낮은 감수성을 나타내는 결과로 생각되었다. SKOV-3 세포주에서 p53 발현은 관찰되지 않았고, G_0G_1 전분기 분획의 미미한 증가 및 cytokeratin과 annexin V으로 측정된 고사 분획의 변화가 관찰되지 않았다. OVCAR-3 세포주에서 p53이 taxol의 투여 여부와 관계없이 발현되었지만 투여 시간에 따른 증가는 관찰되지 않았던 점과 G_0G_1 전분기 분획과 cytokeratin과 annexin V으로 측정된 고사 분획의 완만한 증가로 미루어 볼 때 p53의 발현이 세포주기 변화와 G_2M 분기의 정지와 무관하였으나 고사의 출혈과 관련되어 있을 것으로 생각되었다. 이러한 관찰은 향후 종양으로부터 채취한 종양 세포 부유액에 항암 화학요법제를 투여한 후 유발되는 고사의 정도를 신속하고 정확한 flow cytometry로 측정함으로써 항암 화학요법제의 투여 전 감수성을 알아보는 검사법의 개발 가능성을 암시하는 것으로 생각되었다. Objective : Taxol (paclitaxel)-induced apoptosis was studied to understand their biological mechanism correlated with the expression of p53 in the SKOV-3 and OVCAR-3 ovarian cancer cell lines. Materials and Methods : The SKOV-3 and OVCAR-3 cell lines were cultured in RPMI 1640 medium without taxol (control group) and with taxol for 24 h and 48 h (experimental group). After harvest, the cells were stained with annexin V-FITC (fluorescein isothiocyanate) and anti-cytokeratin antibodies (clone CAM5.2 and clone MNF116). They were washed and stained with p53 antibody. After then the secondary antibodies, i.e., FITC- or phycoerythrin (PE)-conjugated goal anti-mouse (GAM) immunoglobulin G (GAM IgG-FITC or GAM IgG-PE) were added in the cells and they were incubated in the dark. DNA of these cells were stained sequentially with propidium iodide (PI). Standard FASCcan equipped with a 499 ㎚ single laser was used for the analysis of these cells. Results : Both of SKOV-3 and OVCAR-3 cell lines were arrested in the G_2M phase after treatment of taxol, suggesting that these cells would eventually enter into the stage of cell death. Fractions of negative cytokeratin and positive annexin V and amount of sub-G_0G_1 fraction indicative of apototic fractions were lower in the SKOV-3 cell line compared with that in OVCAR-3 cell line, probably as a result of lower sensitivity of SKOV-3 cell line to the taxol. p53 expression were not detected in SKOV-3 cell line. On the basis of observed findings in SKOV-3 cell line and findings of high expressions of p53 regardless of taxol treatment, no increase in their expressions according to culturing time, and gradual increases in sub-G_0G_1 fractions and in fractions of negative cytokeratin and positive annexin V indicative of apoptosis in OVCAR-3 cell line, we concluded that the expression of p53 would not be associated with cell cycle changes and the arrest in the G_2M pahse but associated with the appearance of apotosis. Conclusion : Our results suggest that flow cytometric detection of the apoptotic fractions would be an effective, fast, and accurate method for the chemosensitivity test in tumor cells before the administration of anti-cancer drugs in gynecologic cancer patients.

목적: 본 연구의 목적은 난소 질환에서 혈청내 VEGF농도를 측정하여 다른 임상 요소와 종양 표지 인자들과의 상관 관계를 평가하는 것이다. 연구방법: 사전 동의를 한 76명의 양성 난소 질환자, 9명의 정상인, 4명의 경계성 암환자, 53명의 난소암 환자들의 혈청 VEGF를 ELISA 법으로 측정하였다. 결과: 혈청 VEGF값이 난소 양성 질환자군 (157.7±197.8 pg/ml)과 난소 악성 질환자군 (344.7±372.2 pg/ml)에서 통계적으로 유의한 차이를 보였다 (p<0.005). 결론: 혈청 VEGF값은 난소 양성 질환과 악성 질환에서 감별 진단에 도움을 줄 수 있을 것으로 기대된다. 그래서 우리는 혈청 VEGF 값이 난소질환에서 하나의 진단요소로서 유용하리라 기대하는 바이다. Methods: We measured the serum VEGF concentrations by ELISA in 76 patients with benign ovarian disease, as well as 9 normal volunteers, 4 patients with ovarian borderline malignancy and 53 patients with ovarian cancer. Results: There is a significant statistical difference between the serum VEGF levels in patients with benign ovarian disease (157.7±197.8 pg/ml) and ovarian cancer (344.7±372.2 pg/ml) (p<0.005). Conclusion: Serum VEGF may help in the differential diagnosis between benign and malignant ovarian disease. We suggest that serum VEGF may be useful as a diagnostic tool in ovarian disease.

N/A Objective : The aim of this study is to improve the pregnancy outcome and evaluate the prognostic factor after McDonald operation of IIOC patients. Methods : From January 1, 1995 to December 31, 1999, 230 cases of IIOC were admitted and treated with McDonald operation at department of Obstetrics and Gynecology, Medical college of Catholic university. Among these patients, 187 cases were analyzed. We classified the above patients into 4 groups by gestational weeks of the operation (1st group before 14weeks, 2nd group 14+1-16weeks, 3rd group 16+1-18weeks, 4th group after 18weeks), and we analyzed the mean age of the patients, parity, indications, operation time, delivery methods and fetal survival rate on each groups. Results : The fetal survival rate was 76.08%, and duration of pregnancy was 21.65 weeks±6.39 in the 1st group, 78.5%, 19.24 weeks±6.367 in the 2nd group, 88.2%, 18.25 weeks±5.15 in the 3rd group, 75.67%, 12.12 weeks±7.19 in the 4th group. The 3rd group shows higher fetal survival rate with significantly. Conclusion : We can have higher fetal survival rate and prolongation of duration of pregnancy in the IIOC patients when we do the McDonald operation in the 16+1-18th gestation weeks.
