Gemcitabine 이 봉입되고 CR2945가 결합된 표적지향적 리포좀 제제의 마우스 장기독성

Gemcitabine 이 봉입되고 CR2945가 결합된 리포좀이 성공적으로 제조 되었으며 이를 PANC-1 췌장암이 이식된 실험용 쥐의 꼬리 정맥에 주사한 후 생길 수 있는 주요 장기에 대한 독성을 평가하였다. 제조된 리포좀 제형의 크기(지름), 표면 하전, 약물봉입효율은 각각 500mm, -16.5mV, 그리고 84%였다. 주요 장기(신장, 간, 폐, 비장)의 헤마톡실린/에오신 염색 결과 본 리포좀 제형은 이러한 장기로의 암 전이를 막아줌을 알 수 있었는데, 이러한 효과는 다른 비교 제형 들로부터는 나타나지 않았다. 이러한 실험결과는 향후 안전하고 효과적인 리포좀 제형의 개발에 도움이 될 것으로 기대된다.

We investigated the antioxidant and angiotensin I converting enzyme (ACE) inhibitory activities of red snow crab Chionoecetes japonicas shell (RSCS) hydrolysate by enzymatic hydrolysis and its molecular weight cut-off fractions. The RSCS hydrolysatewas fractionated through two ultrafiltration membranes of 3 and 10 kDa cut-offs. Three fractions (<3 kDa, 3-10 kDa, and >10 kDa) were evaluated for total amino acid composition, antioxidant activities using 2′-azino-bis[3-ethylbenzthiazoline-6-sulfonic acid](ABTS+) radical scavenging and superoxide dismutase (SOD)-like activities and reducing power assays, and ACE inhibitory activity using Hou’s method. Although all fractions showed activity, the <3 kDa fraction of RSCS hydrolysate exhibited the greatestABTS+ radical scavenging, SOD-like and ACE inhibitory activities. However, these fractions exhibited low reducing power. These results suggest that the low-molecular-weight enzymatic hydrolysate of RSCS could be used as a functional ingredient to control oxidative stress and ACE activity.
국내산 무지개송어(Oncorhynchus mykiss)의 생산지에 따른 영양성분 비교

This study compared the nutritional composition of rainbow trout Oncorhynchus mykiss from different inland-based trout farms. The proximate composition ranges of the fish were 67.10–72.77 g/100 g moisture, 19.13–21.35 g/100 g crude protein, 5.12–11.72 g/100 g crude lipid, and 1.24–1.97 g/100 g ash. In addition, rainbow trout contained significantly different mono- and poly-unsaturated fatty acid concentrations, ranging from 1.31 to 4.33 g/100 g and 1.21 to 3.2 g/100 g, respectively. The content of n-3 and n-6 fatty acids ranged from 0.68 to 1.68 and from 0.49 to 1.61 g/100 g, respectively. The n-6/n-3 ratio was in the range of 0.79–1.59. The vitamin content of the rainbow trout from the different farms was 14.83–49.30 μg/100 g vitamin A, 922.05–3,179.38 μg/100 g alpha-tocopherol, ND–73.59 μg/100 g, gamma-tocopherol, 2.97–14.55 μg/100 g vitamin D, and 76.63–126.03 μg/100 g vitamin B2. Slight differences were observed in the crude lipid, fatty acid, and vitamins A and E contents among the of rainbow trouts from the different farms. The results suggest that the fish nutrient composition, especially that of lipids, was significantly different among rainbow trouts from different farms, but may not be related to the production area.
생굴(Crassostrea gigas)의 선도 변화에 포장용기가 미치는 영향

The shelf life of oysters Crassostrea gigas, in two different types of packaging containers, polyethylene (PE) and polyethylene terephthalate (PET), was determined by evaluating the pH, glycogen and soluble protein content, turbidity, and viable cell count. After 7 days of storage, the pH of the packing water in the PE container decreased to 5.88, while the pH in the PET container decreased to 6.03. In the PE container, the glycogen content of the oysters decreased by 0.85 g/100 g and the soluble protein content and turbidity of the packing seawater increased by 1,927.21 mg/100 g and 3.24 McF, respectively. In the PET container, the glycogen content of the oysters decreased by 0.96 g/100 g and the soluble protein content and turbidity of the packing seawater increased by 1,674.75 mg/100 g and 0.98 McF, respectively. The reaction rate constants (K) were as follows: glycogen content, -0.18 (PE) and -0.10 (PET); soluble protein content, 0.29 (PE) and 0.26 (PET); and turbidity, 0.41 (PE) and 0.06 (PET). These results suggested that PET can be used as a new packaging container material for raw oysters because the quality is maintained and it offers more convenient handling during distribution.

Gemcitabine 이 봉입되고 CR2945가 결합된 리포좀 제제의 in vivo 항암 활성

항암제 gemcitabine 이 봉입되고 CR 2945 를 표적 지향적 리간드로 결합시킨 리포좀을 이용하여 인간 췌장암세포(PANC-I)가 이식된 실험용쥐 (Balb/cnumice) 에 주사하여 그 항암활성을 조사하였다. Gemcitabine 이 봉입되고 CR 2945 가 결합된 리포좀과 CR 2945 가 비결합된 리포좀 군에서는 투여 중간에 약물자체의 독성에 의해 사망한 쥐는 없었으나 gemcitabine을 PBS에 녹여 투여한 군에서는 3회 투여부터 사망한 쥐가 발생하였다. 그러나 항암효과 면에서는 Gemcitabine 이 봉입되고 CR 2945 가 결합된 리포좀군, free gemcitabine 군, 그리고 PBS 군 사이에 통계학적인 차이가 나타나지 않았다. 이는 리포좀에 봉입함으로써 gemcitabine 의 독성은 줄어 들었지만 항암효과는 크게 향상시키지 못했음을 나타내고 있기에 제형 개발에 대한 연구가 더 진행되어야 할 것으로 생각된다.

We conducted this study to isolate novel anti-hyperlipidemic agents derived from natural marine products. To accomplish this, we investigated the effects of ethanolic (EtOH) extracts of Ecklonia stolonifera and its phlorotannin constituents, eckol and dieckol, on serum lipid levels in rats with hyperlipidemia that was induced by a high-cholesterol diet or poloxamer 407. Treatment with the EtOH extracts of E. stolonifera and its phlorotannin-rich ethyl acetate (EtOAc) and n-butanol (n-BuOH) fractions induced a significant reduction in triglycerides (TG), total cholesterol (TC), and low-density lipoprotein-cholesterol (LDL-C) levels, as well as a significant increase in the high-density lipoprotein-cholesterol (HDLC) level in hyperlipidemic rats. However, treatment with the water (H2O) fraction did not exert any significant effects on the serum levels of hyperlipidemic rats. In addition, eckol and dieckol isolated from the active EtOAc fraction induced a significant reduction in serum TG, TC, and LDL-C levels, as well as in the atherogenic index (A.I.). Furthermore, treatment with dieckol induced a greater decrease in the serum TG, TC, and LDL-C levels of hyperlipidemic rats than eckol or lovastatin, as well as an increase in the serum HDL-C levels. Taken together, these results suggest that phlorotannins such as eckol and dieckol have the potential for use for the prevention of hyperlipidemic atherosclerosis.
