RAPD분석을 통한 여름양송이(Agaricus bitorquis)의 교잡주 선발

Agaricus bitorquis is an edible white mushroom of the genus that is cultivated at high temperature (25±1℃) unlike A. bisporus is cultivate at 16±2℃. Unlike Agaricus bisporus, an edible white A. bitorquis mushroom is cultivated at high temperature (25±1℃). Most farmers cultivate this mushroom for a long cultivation period in Korea. For this reason, we made heterokayons to develop a new cultivar that generate fruitbodies for short cultivation period. Over one hundred SSIs(single spores isolates) were collected from selected A. bitorquis ASI1151 and ASI1349 strains. Seventy-three SSIs were germinated on CDA(compost dextrose agar) media after 20 days (minimum) or 83 days (maximum) incubation under different media condition. The mycelial growth rate of germinated SSIs was different. 9 homokaryons in ASI 1151 and 11 homokaryons in ASI 1349 from SSIs were selected by OPN-02 primer in RAPD analaysis. Also this primer was used to select heterokaryon that cross among each homokaryon with compatible locus. Therefore 44 compatible matings were confirmed of 99 crossed lines.

This study was carried out to investigate storage stability of harvested mushroom in developed varieties of button mushroom through identification of quality change during 35 days. The mushroom harvested up to 2nd flush mushroom was stored on different storage temperature(4, 7, 10oC) and weight, length, thickness, color of pileus and stipe in fruit body was tested every 7 days. The morphological trait and color of stipe in fruit body were influenced by harvesting period and storage temperature. On the morphological traits of fruit body, mushroom harvested at 2nd flush showed smaller difference than those at 1st flush and mushroom stored at 4oC indicated more difference than ones at the other temperature. Mostly color of pileus in brown button mushroom had lower E (color difference) than white mushroom during storage period. The color of pileus of fruit body in white mushroom had low E at 4oC whereas that of brown mushroom was low at 10oC. The safe period of storage based on the L value is 7 days under a specified environmental condition. ‘Seolgang’ was stored for 14 days as good quality of mushroom, ‘Saedo’ had best quality after harvesting and ‘Seojeong’ was most stable during storage change in white mushroom varieties.

Currently, the spawn of the mushroom Agarcus bisporus is produced by a method developed in the 1980s, and anew spawning method needs to be developed to improve the quality of the spawn. In this study, the condition for a maximum mycelium weight(5.92±0.52 g/L) was shaking culture (24 hours/day) at 24℃ and 120 rpm in CDB (compost dextrose broth). Based on this, the ventilated liquid culture method (2.5 L/min) was cultured for 10 days. This method was appropriate, andwhen the inoculum was cultured at 50 g/mL for about 10 days, it was cultured well without agglomeration and shaking of seed.

A new brown button mushroom cultivar, ‘Hogam’, C34 line, was made by crossing homokaryons, ASI1164 -37 and ASI1175-66, selected by RAPD analysis and by cultivating three times. Mycelium of ‘Hogam’ on CDA (compost dextrose agar) media grew well at 25oC. The optimum pin-heading temperature of new variety and optimum growing temperature was 14- 18oC. The thickness of the mature cap and stipe were thicker than a control, ‘Dahyang’ that developed in 2010. The color of pileus was light brown and lighter than ‘Dahyang’. Days required from casing to first harvesting were three days longer than control strain, but the weight of harvested fruiting body increased by 1.35 times. ‘Hogam‘ cultivar are expected to contribute to the diversification of domestic mushroom cultivars.

Cultivation of brown mushrooms, rather than that of white variants is preferred by Korean mushroom farmers, as the former are resistant to diseases. However, brown mushrooms were cultivated only in selective eco-friendly agricultural farms due to lack of consumer awareness. After providing information about brown mushrooms to respondents through a 1-minute video clip, a survey was conducted on social network service (SNS) to assess recognition and preference for brown mushrooms. A food evaluation was then conducted among 200 people randomly selected from the survey respondents. Most respondents (83%) had not encountered brown button mushrooms previously, and 98% of the respondents were willing to buy these mushrooms because they were “curious about its taste” (44%). In the food evaluation, 32% of the respondents found the brown button mushrooms to be delicious, 28% reported a good flavor, and 31% described a good texture. In addition, we confirmed that 95% of respondents were interested in purchasing brown mushrooms after sampling. Therefore, in the present study, we evaluated public perception, preference, and taste of brown button mushrooms, and confirmed that availability of information on nutrition and benefits s of mushroom consumption could induce consumers to buy brown button mushrooms.
ITS 계통분석을 이용한 주름버섯류 수집균주의 특성평가

Phylogenic relationships and morphological characteristics were classified and investigated among the 233 collected strains of Agaricus spp. The 38 strains were differently identified as different characteristic group using analysis of ITS regions in rDNA. A. bitorquis was showed close relationship in groupA whereas A. campestris was in groupC as different characteristic group among with A. bisporus. There was no phylogentic difference with strains collected by country and different pileus colored Agaricus bisporus. Also the strains were cultivated twice to investigate morphological characteristics of fruiting bodies. The characteristics and yield of collected strains were compared with molecular varieties and seasons by the cultivation. In this result, A. campestirs showed good yield and quality in terms of hardness off-white mushroom was more harder than other number of A. bisporus. Also earliness and color of pileus was influenced by external environment all conditions.
Inter Simple Sequence Repeat(ISSR) 마커를 활용한 느티만가닥버섯(Hypsizigus marmoreus) 종내 다형성 분석

느티만가닥버섯은 맛과 기능성이 풍부한 버섯으로 많이 활용되고 있다. 하지만 긴 재배기간과 낮은 자실체 수확량, 균이 오랜시간 배양되어 오염이 쉽게 발생되는 문제점을 극복할 새로운 품종육성이 필요하다. 이에 따라 육종모본으로 활용되는 느티만가닥 55균주의 종내 유전자원의 정확한 정보를 얻고자 분자유전학적 ISSR 마커분석을 활용하였다. 사용된 마커 중에서 ISSR 13과 15 마커를 사용했을 때 다형성이 분석되었으며 특히 ISSR 15마커 분석으로 다형성이 쉽게 구분되었다. UPGMA분석법으로 계통도를 분석하였을 때, 일부 백색을 가지고 있는 KMC03106, KMC03107, KMC031083 균주가 두 마커 모두에서 가까운 유연관계를 가졌으며, ISSR 15는 수집년도에 따라 3개의 그룹으로 구분되는 것을 확인할 수 있었다. 이 결과로 ISSR마커의 다형석 분석은 느티만가닥버섯의 몇몇 균주에서는 갓색의 유연관계 확인과 수집 시기에 따른 유전변이 구분이 가능하며 자원의 유전적 다양성 확인할 수 있어, 느티만가닥버섯의 품종육성을 위한 효율적인 모본 선발이 가능할 것으로 사료된다. Hypsizygus marmoreus is a mushroom with abundant flavor and medicinal properties. However, its application is limited by problems such as long cultivation period, low biological efficiency, and microbiological contamination; therefore, there is a substantial need for development of new cultivars of this species. In this study, 55 strains of H. marmoreus were subjected to inter simple sequence repeat (ISSR) analysis to identify markers for the selection of mother strains for breeding from the collected germplasm. ISSR 13 and 15 were confirmed as polymorphic markers. The three strains (KMCC03106, KMCC03107, and KMCC03108) with white cap color were found to be genetically closely related upon UPGMA analysis of both ISSR 13 and 15. Based on the PCR analysis results for ISSR 15, the collected germplasm were differentiated into three groups according to the strain collection year. Thus, ISSR 15 could be a marker for determining the phylogeny of cap color and genetic variations according to the strain collection year. These results suggest that ISSR markers can be effective tools for the selection of mother strains for breeding of H. marmoreus.
