Neuroprotective compounds from Reynoutria sachalinensis

Glutamate is a neurotransmitter in central nervoussystem. Overexpression of glutamate leads to oxidativestress, resulting in several neurodegenerative disordersthat include Alzheimer’s disease. The n-hexane fraction ofstems and ethyl acetate (EtOAc) fraction of flowers ofReynoutria sachalinensis provide neuroprotection againstglutamate-induced oxidative toxicity in HT22 cells. In thisstudy, 1-decanol (1), b-amyrin (2), dammaran-3b-ol (3),campesterol (4), daucosterol (5), ergosterol peroxide (6),emodin 8-O-b-D-glucopyranoside (7), quercetin (8) andisoquercitrin (9) were isolated from n-hexane fractions ofstems and EtOAc fractions of flowers of R. sachalinensis. Their neuroprotective activity was evaluated by MTTassay. 1-Decanol, campesterol, ergosterol peroxide, quercetinand isoquercitrin exhibited neuroprotective activity. These compounds decreased reactive oxygen species level,showed anti-oxidant activity with DPPH radical and in aH2O2 scavenging assay. Therefore, the neuroprotectiveactivity of 1-decanol, campesterol, ergosterol peroxide,quercetin and isoquercitrin are associated with antioxidantactivity.
Costunolide와 Dehydrocostuslactone을 이용한 목향의 함량분석

Aucklandia lappa Decne (Compositae) has been used for treatment of abdominal pain, vomiting, diarrhea, chronic inflammation, and antibacterial effect. The quality of these herbs has been affected by many factors such as collection time,place, temperature, cultivation environment and manufacturing process. We used costunolide and dehydrocostuslactone as marker compounds for quality evaluation of rhizome of Aucklandia lappa. 66 samples of Aucklandia lappa were collected from those habitats in Korea and China. The developed HPLC-DAD method was applied to investigate for quality control of Aucklandia lappa samples. The average contents of the costunolide and dehydrocostuslactone were 2.3895% and 0.9258%, respectively. The principal component analysis (PCA) exhibited that classification of Aucklandia lappa according to origin not separated. Results of this study may be satisfactory applied to determination of content criteria of Aucklandia lappa.

Alzheimer’s disease (AD), most common form of dementia is characterized that memory deficit and loss of cog- nitive function. This study was evaluated cognitive enhancing effect of water and ethanol extracts of Codonopsis lanceolata and compared using Morris water maze and passive avoidance test. The water and 70% ethanol extracts (100, 300 and 500 mg/kg) were administered to mice. The neuroprotective effect on glutamate-induced cell death in HT22 cells was addi-tionally investigated using MTT assay. Results showed 70% ethanol extract of Codonopsis lanceolata enhanced cognitive function than water extract, as shown by decrease in escape latency time in Morris water maze test. In passive avoidance test, 70% ethanol extract also increased the latency time compared to the water extract. Furthermore, 70% ethanol extract significantly protected neuronal cell against glutamate cytotoxicity and showed higher than neuroprotective effect of water extract. These results indicate that 70% ethanol extract more improve spatial cognitive ability and protected neuronal cells than water extract.

Alzheimer's disease is progressive neurodegenerative disorder by the loss of memory and learning abilities. Codonopsis lanceolata (C. lanceolata) is traditional medicinal plant used for the treatment of inflammatory diseases. The aim of study was to evaluate the effect of steamed C. lanceolata on scopolamine-induced memory impairment in the Morris water maze test and passive avoidance test. In addition, this study investigated the neuroprotective effects of steamed C. lanceolata on glutamate-induced cell death in HT22 cells using MTT assay. The results showed that steamed C. lanceolata (500 mg/kg body weight, p.o.) reversed spatial memory impairment by scopolamine in Morris water maze test and passive avoidance test. Steamed C. lanceolata attenuated memory impairment by scopolamine compared with common C. lanceolata. In addition, administration of steamed C. lanceolata significantly also reduced cell death. We suggest that steaming process more improve cognitive enhancing and neuroprotective effect of C. lanceolata than common C. lanceolata.
방기의 품질 평가를 위한 Sinomenine, Magnoflorine, Syringaresinol의 함량 분석

Sinomenium acutum has been used for the treatment of rheumatoid arthritis, arrhythmia, and pain. We eval-uated the quality of S. acutum extract by quantitatively analyzing its components such as sinomenine, magnoflorine and syringaresinol with the simultaneous determination method using HPLC-DAD. A total of 53 samples collected from dif- ferent localities were evaluated with this quality evaluation method. Sinomenine, magnoflorine and syringaresinol from tested samples ranged from 0.0649~9.1050%, 0.7460~10.7590% and 0.0010~0.2441%, respectively. In the current study, we were able to exhibit the diverse quality of S. acutum samples collected from various locations using the simultaneous determination method.

This study was designed to determine and compare the cognitive enhancing effect of various Codonopsis lan- ceolata extracts by steaming, fermentation and high hydrostatic pressure process. We prepared water extract of C. lanceolata, steamed C. lanceolata, steamed and fermented C. lanceolata and C.lanceolata by high hydrostatic pressure process and fer-mentation. Cognitive enhancing effect of extracts was evaluated in scopolamine-induced memory impairment mice using by passive avoidance test and Morris water maze tests. MTT assay was conducted to investigate neuroprotective effect on glutamate induced cell death in HT22 cells. Steamed and fermented C. lanceolata water extract decreased escape latency in Morris water maze test and increased the latency time of the passive avoidance test compared to other extracts. Fur-thermore, the steamed and fermented C. lanceolata water extract showed neuroprotective effect. These results suggest that steaming and fermentation process more improve cognitive enhancing effect of C. lanceolata than other process.
백출의 Atractylenolide I과 Atractylenolide III의 함량분석

This study was conducted to determine the content of atractylenolides I and III in Atractylodes japonica. An established HPLC-DAD method was used to monitor contents of atractylenolides I and III in A. japonica obtained from Korea and China and compared with contents of A. macrocephala, A. chinensis, and A. lancea. Quantitative analysis of atractylenolides I and III was carried out on a Shiseido C18 column (S-5 µm, 4.6 mm I.D. × 250 mm) with gradient elution composed of acetonitile-water. The results show that the average content of atractylenolides I and III in A. japonica were 0.0954 and 0.1963%,respectively, and contents of A. lancea were higher than A. macrocephala, A. chinensis. In this study, we identified the differentiation of the quality of A. japonica from different species and collected locations and established content standard of atractylenolides I and III in A. japonica and this content standard was helpful to quality control of A. japonica.
