        Pandanus fascicularis Lam Extract Inhibits Pro-Inflammatory Cytokines Production in LPS-Stimulated RAW 264.7 Cells

        Pandanus fascicularis Lam has been shown to exert to a variety of physiological effects on edema, tumors, leprosy, spasm, inflammation, pain, and rheumatoid arthritis. In this study, the effects of a P. fascicularis extract on the production of lipopolysaccharide (LPS)-induced pro-inflammatory cytokines interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF)-α expression and on the production of the inflammatory mediator, nitric oxide (NO), in RAW 264.7 cells were examined. The P. fascicularis extract decreased LPS-induced NO production. Moreover, enzyme-linked immunosorbent assays revealed that the P. fascicularis extract concentration-dependently suppressed LPS-induced production of the pro-inflammatory cytokines IL-1β, IL-6, and TNF-α. These results suggest that the P. fascicularis extract has potential as an antiinflammatory therapeutic substance for use in the prevention of the inflammatory disorder.

        Inhibition of Nelumbo nucifera Stamens-derived Kaempferol on FcεRI-mediated GATA-1 Expression

        The transcription factor, GATA-1, plays an important role in the FcεRI α chain expression in mast cells and basophils. This study was conducted to investigate the downregulation of the transcription factor GATA-1 by kaempferol isolated from Nelumbo nucifera stamens in FcεRI-mediated allergic reactions. Kaempferol inhibited FcεRI-mediated histamine release. Western blotting analysis and RT-PCR showed that the protein and mRNA expression of GATA-1 was suppressed by kaempferol in a dose-dependent manner. These results suggest that kaempferol may inactivate basophils by downregulating the FcεRI α chain expression via the inhibition of the GATA-1 expression

        Kaempferol Isolated from Nelumbo nucifera Stamens Inhibits Phosphorylation of ERK 1/2, Syk, and Lyn in FcεRI-mediated Allergic Reaction

        Cross-linking of FcεRI with multivalent allergen and specific IgE on mast cells and initiates activation of signal cascades that lead to IgE-mediated allergic responses. Kaempferol is a phenolic compound contained in natural substances including the stamens of flavonoid-rich Nelumbo nucifera stamens. In this study, the effects of kaempferol on allergic reaction induced by an FcεRI α chain antibody were investigated. Western blot analysis revealed that the protein expression of FcεRI and the phosphorylation of Syk, Lyn, and extracellular signal-regulated kinases (ERK) 1/2 were dose-dependently inhibited by kaempferol. β- Hexosaminidase was reduced to 97.9, 89.1, 75.1, and 55.8% when treated with 5, 10, 20, and 40 μΜ kaempferol, respectively. From the results, the negative regulation of basophil activation with kaempferol via the suppression of FcεRI expression and degranulation may be suggested to be through inhibition of Lyn and Syk, and ERK 1/2 activation.

        Isolation and Identification of Flavonoids from Gujeolcho (Chrysanthemum zawadskii var. latilobum) as Inhibitor of Histamine Release

        In an ongoing search of compounds from natural sources that exhibit anti-allergic properties, 3flavonoids were isolated from the ethyl acetatefraction of methanol extracts of gujeolcho (Chrysanthemum zawadskii var. latilobum), with guided histamine release inhibitory effects. The isolated compounds were identified as eriodictyol-7-O-β-D-glucuronide, 5,7-dihydroxy-4-chromene, and 6-methoxyluteolin based on spectroscopic evidences including NMR and MS spectra. These results suggest that gujeolcho extracts and 3 compounds would be candidates as therapeutic or preventive agents for allergic reaction.

        Inhibitory Effects of Blueberry Root Methanolic Extract on Degranulation in KU812F Cells

        The effect of blueberry (Vaccinium angustifolium)root methanolic extract (BRM) on the A23187 plus phorbol myristate acetate (PMA)-induced degranulation in human basophilic KU812F cells was investigated. The total phenolic content (TPC) was 170±1.9 mg gallic acid equivalents (GAE)/g BRM. BRM reduced levels of histamine and β-hexosaminidase released from stimulated KU812F cells. Intracellular calcium concentration ([Ca2+]i)were determined by spectrofluorometric analysis using Fura 2-AM was found to be reduced by BRM with dosedependent manner. Moreover, Western blot analysis revealed that protein kinase C (PKC) translocation was inhibited by BRM with dose-dependent manner. These results indicated that BRM inhibited histamine and β-hexosaminidase through the suppression of Ca2+ influx and PKC translocation. Therefore, we suggest that BRM is potent inhibitor of degranulation in mast cells and basophils, and may be useful in preventing the allergic reactions.

        Fermented Product Extract with Lentinus edodes Attenuate the Inflammatory Mediators Releases and Free Radical Production

        Lentinus edodes contains functional metabolites such as polysaccharopeptides, lectins, and secondary metabolites. Fermented soybean paste is representative fermented materials in Korea, and is gradually increasing due to various biological activities. In the present study, ethanol extracts of fermented products with/without L. edodes were designated as SPL and SP, and prepared to develop safer and therapeutic functional foods with antioxidant and anti-inflammatory activities for treatment of inflammatory disorders. SP and SPL extracts exhibited antioxidant effects via inhibiting radical activities. Inflammatory mediators, nitric oxide (NO), prostaglandin E2 (PGE2), interleukin (IL)-1β, IL-6, IL-8, tumor necrosis factor (TNF)-α, and inducible nitric oxide synthase (iNOS) production and nuclear factor-kappa B (NF-kB) activation were down-regulated by two extracts. SPL extract more strongly enhanced the antioxidant and anti-inflammatory activities than SP extract. Its’ activities shown more longer fermentation period and more strong inhibitory effects. Taken together, our results suggested that fermented product with medicinal plant has synergic effect and SPL can be a potential candidate for treatment of inflammatory bowel diseases.

        Suppressive Effects of Vaccinium angustifolium Root Extract via Down-Regulation of Activation of Syk, Lyn, and NF-κB in FcεRI-Mediated Allergic Reactions

        Vaccinium angustifolium, reported as the lowbush blueberry, has a rich polyphenolic content with which biological activities have been closely associated. In this study, the effects of V. angustifolium root extract (VAE) on the anti-FcεRI α chain antibody (CRA-1)-induced FcεRI-mediated signaling factors, protein tyrosine kinases (PTK), Lyn, Syk, and nuclear factor kappa-B cells (NF-κB) in KU812F cells were investigated. The total phenolic content of VAE was found to be 170±1.9 mg gallic acid equivalents/g. Western blot analysis revealed that VAE dose-dependently inhibited FcεRI-mediated phosphorylation of PTK involving Lyn and Syk. Evaluation of intracellular reactive oxygen species (ROS) by spectrofluorometric analysis using 2’7’-dichlorofluorescin-diacetate revealed that they were reduced by VAE in a dose-dependent manner. Moreover, VAE reduced the levels of β-hexosaminidase released from CRA-1-stimulated KU812F cells. It was identified that VAE suppressed CRA-1-induced activation of NF-κB by Western blot analysis. Our results show that VAE may contribute to the inhibition of allergic actions via inactivation of basophils through the inhibition of β-hexosaminidase release and ROS production, which occurs as a result of inhibition of PTK, Syk, Lyn, and NF-κB.

        Vaccinium angustifolium Root Extract Suppresses FcεRI Expression in Human Basophilic KU812F Cells

        Vaccinium angustifolium, commonly known as the lowbush blueberry, is a rich source of flavonoids, with which various human physiological activities have been associated. The present study focuses on the investigation of the effect of the methanolic extract of V. angustifolium root extract (VAE) on high affinity immunoglobulin E receptor (FcεRI) α chain antibody (CRA-1)-induced allergic reaction in human basophilic KU812F cells. The total phenolic content of VAE was found to be 170±1.9 mg gallic acid equivalents/g. Flow cytometry analysis revealed that the cell surface expression of FcεRI was suppressed in a concentration-dependent manner upon culture with VAE. Reverse-transcriptase polymerase chain reaction analysis showed that the mRNA level of the FcεRI α chain was reduced in a concentration-dependent manner as a result of VAE treatment. Western blot analysis revealed that the protein expression of FcεRI and the phosphorylation of extracellular signal-regulated kinases (ERK) 1/2 were concentration-dependently inhibited by VAE. We determined that VAE inhibited anti-CRA-1-induced histamine release, in addition to the elevation of intracellular calcium concentration ([Ca2+]i), in a concentration-dependent manner. These results indicate that VAE may exert an anti-allergic effect via the inhibition of calcium influx and histamine release, which occurs as a result of the down-regulation of FcεRI expression through inhibition of ERK 1/2 activation.

        구절초 에탄올 추출물의 FcεRI α chain 발현 저해효과

        KU812F 세포는 세포표면에 FcεRI을 발현하고 있어, IgE를 매개로 한 알러지 반응에 있어 중요한 역할을 담당하고 있는 것으로 알려져 있다. KU812F 세포를 이용하여, FcεRI α chain 발현에 있어 구절초 에탄올 추출물의 저해효과를 조사하였다. 본 연구 결과, FcεRI α chain의 세포표면 발현량 및 mRNA수준을 구절초 추출물이 감소시키는 것을 flow cytometry 및 RT-PCR로 각각 확인하였다. 또한, 구절초 추출물은 CRA-1 자극에 의한 히스타민 유리를 농도 의존적으로 억제시켰다. 이러한 결과는 구절초 추출물이 FcεRI α chain 발현을 감소시킴으로써, 히스타민 유리가 억제되고, 이를 통해 항알러지 활성에 기여하는 것으로 생각된다. Human basophilic KU812F cells express a high affinity for IgE receptor, FcεRI, which plays an important role in immunoglobulin E (IgE)-mediated allergic reactions. Using the human basophilic KU812F cells, we assessed the inhibitory effects of the ethanolic extract from Chrysanthemum zawadsaki(CZ) on FcεRI α chain expression. Flow cytometric analysis showed that cell surface FcεRI α chain expression was suppressed when the cells were cultured with CZ extract. RT-PCR analysis showed that the mRNAs for FcεRI α chain decreased by the treatment of CZ extract. Moreover, CZ extract was also observed to result in a reduction in the release of histamine from anti-FcεRI antibody (CRA-1)-stimulated cells. These results suggest that CZ extract may exert anti-allergic activity through down-regulation of FcεRI α chain expression and a subsequent decrease in histamine release.
