만 5세 산들반 교사의 자연친화교육 경험에 관한 자문화기술지 연구

본 연구의 목적은 자문화기술지 연구를 통해 유아교육기관에서 만 5세 유아를 대상으로 실시하는 자연친화교육의 과정에서 연구자인 교사가 어떤 경험을 하고, 교사의 경험이 내포하고 있는 의미를 탐색하는 것이다. 수집된 자료는 반성적 자료, 참여관찰, 심층면담, 문화적 인공물 등이며, 자료수집 후 연구문제를 중심으로 자료를 분석하였다. 첫째, 만 5세 산들반의 자연친화교육에서 나타난 교사의 경험은 ‘눈으로 보고 가슴으로 느낀 자연친화교육의 가치’와 ‘교사의 발목을 잡는 자연친화교육 상의 딜레마’로 나타났다. 둘째, 만 5세 산들반의 자연친화교육에서 나타난 교사 경험의 의미는 ‘자연친화교육을 연구하는 교사로 거듭나기’와 ‘생활 속에서 자연애호 실천하기’로 나타났다.

The ancient Jeongchiwi local wine-drinking rite(正齒位 鄕飮酒禮), as described in the Three Ritual Classics, had a strong national characteristic. It was explained as one of the rituals that form the foundation for properly governing the country as a place to maintain the order between the older and the younger. When the Han and Tang dynasty commentators explained the Jeongchiwi local wine-drinking rite, they also recognized that the ritual was performed in the context of an ancient state’s political and social operation. Throughout the history of ceremonial institutions, the ancient characteristics of the Jeongchiwi local wine-drinking rite have either disappeared or been preserved. Among them, the aspect of the state attempting to operate the rite through the local officials within the bureaucratic system was maintained, which was also the case in the Joseon Dynasty. At the beginning of the dynasty, Joseon made a manual for the local wine-drinking rite as part of organizing national rites and named the local officials as both the organizers and key participants. These officials were also considered beings who would embody the will of the state. This trend continued in the late Joseon Dynasty, as evidenced by discussions in the central court and private letters among intellectuals. It is crucial to understand the national context of the Jeongchiwi local wine-drinking rite, which previous scholarship has overlooked in favor of its local aspect.
우엉중 Polyphenol Oxidase의 정제 및 특성에 관한 연구

Polyhenol oxidase was extracted from fresh burdock(Aclium Lapa.L) roots and purified by a combination of ammonium sulfate fractionation, sephadex G-100 and DEAE-cellulose column chromatography. The physico-chemical and enzymological propertics of the purified enzyeme were investigated and the result obtained were as follows. 1. Burdock polyphenol oxidase was purified about 63.4 folds and the recovery of enzyme activity was 39.7%. 2. The purified enzyem was active with o-diphenols such as (+)-catechin, epictechin and chlorogenec acid ; it showed affinity toward triphenols but inactive toward m-diphenols, p-diphenols and monophenols. 3. The Km value of the purified enzyme was 5mM with (+)-catechin as substrate. 4. Inhibitor studies indicated that sodium diethyldithiocarbamate(DETC)and sodium bisulfate were the most potent. DETC was a linear simple non-competitve inhibitor with respect to (+)-catechin at pH 7.3with Ki value of 11.25mM. 5. Optimum temperature for activity was 30℃ and optimun pH for activity was 7.0. 6. The purified enzyme was almost stable under the temperature of 30℃. 7. Molecular weight of the burdock polyphenol oxidase was estimated to be about 58.000.
복숭아중의 Polyphenol Oxidase에 관한 연구

Polyphenol oxidase was extracted from three species of peach which were sampled three times the developing stages. The experimental results were summarized as follows. 1. A PPO activity of Gidobakdo and Chang-bangchosang were increased according to growing. 2. A PPO from three species were active with o-diphenols and trihydroxyphenols. But it were inactive toward m-diphenols, p-diphenols and monophenols. 3. Optimum pH from three species was 7.0 and was not change a optimum pH according to growing. 4. The Km values of a PPO was increased during the developing stages. 5. Inhibitor studies indicated that L-ascorbic acid and L-cysteine in all samples were the most potent in their inhibition reaction.
