Evodia rutaecarpa (Juss.) Benth.(ER) and Chaenomeles sinensis Koehne (CS) have multiple applications and were known to have anti- inflammatory effects. In the current study, we investigated to clearly understand the mechanism of therapeutic role for CS, ER and their combination in CIA model mice. Methods : DBA/1OlaHsd mice were immunized with bovine type II collagen. After a second collagen immunization, mice were treated with CS, ER and their combination once a day for 7 weeks. Cytokine production and gene expression were assessed during CIA (collagen-induced arthritis) model mice in knee joint, lymph node (LN) using ELISA and FACS analysis. The severity of arthritis within the knee joints was evaluated by histological assessment of cartilage destruction and pannus formation. Result : Oral administration of CS, ER and their combination (150 ㎎/㎏) significantly suppressed the progression of CIA, and significantly suppressed the progression of CIA and inhibited the production of TNF-α and IL-6 in serum. The erosion of cartilage was dramatically reduced in mouse knees after treatment with CS plus ER. Conclusion : These result suggest that CS plus ER significantly suppressed the progression of CIA and that this action was characterized by the decreased production of TNF-α, IL-6 and collagen II specific antibody in serum.

연구목적: 폐경 여성에 있어서 혈중 에스트라디올 (estradiol, E2) 농도와 안압 수치 사이에 어떤 연관성이 존재하는지를 알아보고자 하였다. 연구재료 및 방법: 1999년 1월부터 2000년 12월까지 2,462명의 폐경 여성들에게서 혈중 E2 농도의 측정과 함께 안압을 측정하였다. 혈중 E2 농도 측정은 방사면역측정법 (radioimmunoassay, RIA)을 이용하였으며 안압은 골드만 압평 안압계 (Goldmann applanation tonometer)를 사용하여 측정하였다. 모든 자료는 양 표본 t-검정, 선형 회귀 분석, 일원배치 분산 분석에 의거하여 분석하였다. 결과: 평균 연령은 56.2±5.6세였다. 평균 E2의 농도는 13.4±12.9 pg/ml였고, 좌안압, 우안압, 그리고 양측 평균 안압의 평균치는 각각 13.1±2.6 mmHg, 13.0±2.4 mmHg, 그리고 13.0±2.6 mmHg로 확인되었다. 혈중 E2의 농도에 따라 환자를 2군 또는 4군으로 분류하였을 때 각 군에 따른 평균 안압의 수치에는 통계적으로 유의한 차이가 없었다. 선형 회귀 분석 결과에서도 평균 안압과 나이, 그리고 평균 안압과 혈중 E2 농도 사이에 유의한 상관관계가 없었다. 결론: 폐경 여성에 있어 혈중 E2 농도와 안압 사이에 직접적인 연관성은 존재하지 않는 것으로 생각된다. Objectives: To evaluate the association between serum estradiol (E2) level and intraocular pressure (IOP) in postmenopausal women Methods: From January 1999 to December 2000, serum concentration of estradiol and IOP were measured in 2,462 postmenopausal women. Serum concentration of estradiol was measured by radioimmunoassay and IOP was measured using Goldmann applanation tonometer. All data were analyzed using Student``s t-test, analysis of variance (ANOVA), and linear regression analysis. Results: The mean age of the study subjects was 56.2±5.6 years. The mean E2 concentration was 13.4±12.9 pg/ml. The mean IOPs were 13.1±2.6 mmHg, 13.0±2.4 mmHg, and 13.0±2.6 mmHg for the left eyes, right eyes, and the average of both eyes, respectively. When the patients were divided into 2 groups or 4 groups according to the E2 concentration, the IOP of each group did not differ significantly. On linear regression analysis, no significant association was found between IOP and age or IOP and serum E2 concentration. Conclusion: This study suggests that there is no direct association between IOP and serum E2 concentration in postmenopausal women.

Objective: Sinapis alba L. (SA) is a korean traditional herbal medicine that is usually used to prevent or treat inflammatory diseases, such as respiratory infection and rheumatoid arthritis. However, the effects of SA supplementation in vitro on serum antibody levels, splenocyte and peritoneal macrophage immune responses have not yet been determined. In this study, we examined the effect of SA on the production of Th1/Th2 cytokines. Methods: Splenocytes were isolated from naive C57BL/6 mice. Cells were enriched for CD4+ cell populations by first staining the cells with anti-CD4 (BD PharMingen, Calif, USA). CD4+ T cells were selected on a (CS) column, and the flow-through was collected as CD4+ T cells. Isolated cells were activated by overnight incubation on 24-well plates coated with 1ug/mL anti-CD3, 1ug/mL anti-CD28 and with SA (100ug/mL). Primary macrophages were collected from the peritoneal cavities of mice (8-week-old female C57BL/6). The peritoneal macrophages were washed and plated with RPMI-1640 overnight for the experiments. After 48-hours cultures, samples were centrifuged at 2000 rpm for 10 minutes, and the supernatants were stored at -80℃. Mouse IL-4, IFN-y and TNF-a were quantified using ELISA kits (BioSource International, Camarillo, Calif, USA) according to the manufacturer`s protocols. Results: SA at 100ug/ml decreased the generation of Th1 cytokine (IFN-y) by 0.5-fold. However, SA has no effect on Th2 (IL-4) production. Conclusions: These results suggest that SA may play an important role in the control of T-cell-mediated autoimmunity by down-regulation of Th1 cytokine (especially IFN-y, TNF-a). These data may contribute to the design of new immunomodulating treatments for a group of autoimmune diseases.
