- In order to investigate whether or not the alcohol -treated rat liver cells canbe protected by Korean Citrus junos and medicinal herbs, We compared the serumbiochemistry of rats administered both alcohol and the complex of Korean Citrus junosand medicinal herbs to control rats treated with alcohol alone. The activities of Aspar-tate aminotransferase(AST) and Alanine aminotransferase(ALT) in the citron 3(Citron3, less mellowed citron which was ripened for three months) ± Phellinus linteus, Alnusjaponica, Dendropanax morbifera and citron 4 (Citron 4, completely mellowed citronwhich was ripened for four months) ± Phaseolus radiatus, Cordyceps militaris groupwere significantly low when compared with the negative control group (p 0.05). Thelevels of triglyceride (TG) in all experimental groups were significantly lower than thenegative control group (p 0.05). The concentrations of total cholesterol in the citron3± Phellinus linteus, Alnus japonica, Dendropanax morbifera and citron 4 ± Phaseolusradiatus, Cordyceps militaris, Phellinus linteus were lower than the negative controlgroup (p 0.05). The activities of alcohol dehydrogenase (ADH) in all experimentalgroups were significantly high when compared with the normal control group (p 0.05).These results suggest that the complex of Korean Citrus junos and medecinal herbscould be an excellent candidate for protecting rat liver cell damage induced by alcohol.

This study sought to investigate the ameliorating effects of a Korean medicinal herb (KMH) complex on the impacts of alcohol consumption in rat hepatocytes and in reducing the total cholesterol levels and the total lipid levels in the serum. We compared the body weight gain and ratio of the liver, the kidney to body weight, and also the serum biochemistry of the rats administered with both the alcohol and the KMH complex to the control rats treated with alcohol alone. The clinically important enzyme markers (Aspartate Aminotransferase, AST, and Alanine Aminotransferase, ALT) of rats, administered with both the alcohol and the KMH complex treatments, were compared with those in the control group. The treatment regimen (KMH complex) significantly reduced the serum AST and ALT levels, indicating the hepato-protective effects of the KMH complex. Furthermore, total cholesterol and total lipid levels were significantly reduced. These results indicate that the KMH complex may positively mediate the effects of alcohol on hepatocytes and the general liver functions.

- In order to investigate whether or not CCD -986sk cell line can be affectedby Korean Citrus junos and medicinal herbs, we examined the MTT assay when wetreated Korean Citrus junos and medicinal herbs in CCD -986sk human fibroblast cellline. The samples that added complex of grinded extracts of Korean Citrus junos andboiling -water extracts from Korean medicinal herbs were tested for cell proliferationactivity by means of a modification of the MTT assay. Among mixture of Citron 3(Citron 3, less mellowed citron which was ripened for three months) and boiling-waterextracts, the group Citron 3 ± Phellinus linteus showed significantly strong cell pro-liferation activity. And among mixture of Citron 4 (Citron 4, completely mellowedcitron which was ripened for four months) and boiling-water extracts, the groupCitron 4 ± Cordyceps militaris and Citron 4 ± Phellinus linteus showed significantlystrong cell proliferation activity, respectively. These results suggest that complex ofKorean Citrus junos and medicinal herbs could be an excellent candidate for protec-tion of human skin aging.
한약 탕제를 이용한 항 Herpes virus 제제의 개발 연구

In order to search for anti-HSV agents from natural complex products, we extended the number of specimens. Both methanol extract and boiling water extract of the natural complex products were screened to detect anti-HSV activity by MTT assay. Anti-HSV activities of thirteen natural complex products extracted by methanol and boiling water were screened. Three of 13 natural complex products extracted by methanol showed efficacy against HSV. Natural complex products showing anti-HSV activities as methanol extracts were No.3, 6, 11 and their Sl were 323.809, 2811.041 and 708.20. As water boiling extracts, No.8 and No.11 have displayed Sl of 16.45 and 60.39 respectively. Especially anti-HSV activities of natural complex products extracted by methanol No.6 was stronger than other ones.
Korean Medicinal Herb Extracts Inhibit Melanin Formation in Clone M-3 Mouse Melanocyte Cell Lines

- In order to search for anti -melanin formation agents from Korean medic-inal herbs, we selected 21 Korean medicinal herbs, based on a review of Korean tradi-tional medicine books and the recommendations of Korean traditional medicaldoctors. We tested for inhibition effect of melanin pigmentation of Clone M-3 mousemelanocyte cell lines when we treated the extracts of 21 medicinal herbs in the mousemelanocyte cell lines, respectively. Among 21 medicinal herb extracts, 5 extractsshowed a inhibition effect of melanin formation. The sample Phaseolus radiatus L,Cordyceps militaris, Pinellia ternata, Phellinus linteus and Citrus junos Tanaka showeda significantly little formation of melanin pigments compared with control groups.Especially extract of Citrus junos Tanaka was more potent than the others. Theseresults suggest that extract of Korean Citrus junos may represents an excellentcandidate for inhibition of melanin pigmentation at in vitro level.
Study on the Subacute Toxicity of Complex of Pine Needle Oil and Korean Medicinal Herbs against Rats

Pine needle oil and Korean medicinal herbs (KMH) are known as effective therapeutic agents on various blood vessel disease. We have already reported the ameliorative effect of complex of pine needle oil and Korean medicinal herbs against hyperlipidemia. But safety and non-toxicity of pine needle oil and Korean medicinal herbs to normal animal cells have not been studied clearly. In this study, we investigated whether pine needle oil and Korean medicinal herbs show side effects on rat or not. These materials were administered to rats, and subacute toxicity was examined by measuring the hematological values, CBC differentiation, biochemical levels of blood (TP, total protein; albumin; ALP, alkaline phosphotase; AST, aspatate aminotransferase; ALT, alanine aminotransferase; T- Chol., total cholesterol; T- Bil., total bilirubin) and urine analysis, suggesting that the sample have no side effects and cytotoxicity. These results indicate that the complex of pine needle oil and Korean medicinal herbs may effective non-toxic, safety therapeutic agents on hepatocytes and hyperlipidemia.

Cytotoxicity of Saeng-Maek-San (SMS), a Korean medicinal herb complex, was studied on vero cell lines, ATCC-CCL81. The boiling-water extract of SMS (100㎕), at the concentration of 200 ㎎/㎖, was diluted sequentially in two-fold steps with PBS. The cells were incubated with different concentrations of SMS, in order to determine the cytotoxicity of the SMS. After 2 days of incubation, the formazan produced by live vero cells was determined by the MTT (tetrazolium-based colorimetric) assay. IC_50 of SMS was determined for 50% cell viability. The assessment indicates that cell's viability was lower than 50% at concentrations of SMS over 12.5㎎/㎖. Therefore, SMS exhibits no cytotoxicity on vero cell line at concentration lower than 12.5㎎/㎖.
