microRNAs regulate a diverse spectrum of cancer biology, including tumorigenesis, metastasis, stemness, and drug resistance. To investigate miRNA-mediated regulation of drug resistance, we characterized the resistant cell lines to 5-fluorouracil by inducing stable expression of miRNAs using lenti-miRNA library. Here, we demonstrate miR-551a as a novel factor regulating cell survival after 5-FU treatment. miR-551a-expressing cells (Hep3B-lenti-miR-551a) were resistant to 5-FU-induced cell death, and after 5-FU treatment, and showed significant increases in cell viability, cell survival, and sphere formation. It was further shown that myocyte-specific factor 2C is the direct target of miR-551a. Our results suggest that miR-551a plays a novel function in regulating 5-FU-induced cell death, and targeting miR-551a might be helpful to sensitize cells to anti-cancer drugs.
Pluripotin(SC-1) 처리를 통한 단위발생 마우스 배아줄기세포 생산 효율 향상

Various small molecules can be used to control major signaling pathways to enhance stemness and inhibit differentiation in murine embryonic stem cell (mESC) culture. Small molecules inhibiting the fibroblast growth factor (FGF)/ERK pathway can preserve pluripotent cells from stimulation of differentiation. In this study, we aimed to evaluate the effect of pluripotin (SC-1), an inhibitor of the FGF/ERK pathway, on the colony formation of outgrowing presumptive mESCs. After plating the zona pellucida-free blastocyst on the feeder layer, attached cell clumps was cultured with SC-1 until the endpoint of the experiment at passage 10. In this experiment, when the number of colonies was counted at passage 3, SC-1-treated group showed 3.4 fold more mESC colonies when compared with control group. However, after passage 4, there was no stimulating effect of SC-1 on the colony formation. In conclusion, SC-1 treatment can be used to promote mESC generation by increasing the number of early mESC colonies.
VR 기반 사회상황 이야기 중재가 자폐스펙트럼장애 학생의 의사소통기술에 미치는 효과

본 연구는 자폐스펙트럼장애 학생의 의사소통기술을 향상시키기 위한 목적으로 VR 기반 사회상황 이야기 중재를 적용하여 그 효과를 알아보았다. 특수학교에 재학 중인 만 13세 자폐스펙트럼장애 남학생 1명을 대상으로 하였으며, 행동 간 중다기초선 설계를 사용하여 기초선, 중재, 유지 단계로 진행하였다. 대상 학생의 도전적 행동을 파악하고, 수업 상황에서 적절하게 사용할 수 있는 의사소통기술을 습득할 수 있도록 VR 기반 사회상황 이야기를 제작하였다. 자료는 사건기록법을 활용하여 행동의 발생 빈도를 측정하였다. 중재 결과, 반응하기, 시작하기, 요구하기 3가지 의사소통기술 모두 증진되고 유지되었다. 자폐스펙트럼장애가 가진 시각 정보처리 강점을 활용한 VR 기반 사회상황 이야기 중재가 긍정적인 결과에 기여한 것으로 보인다. 본 연구 결과는 VR 기반 사회상황 이야기 중재가 자폐스펙트럼장애 학생의 의사소통기술을 증가시킬 수 있는 효과적인 중재로 활용될 수 있음을 시사한다. This comprehensive study sought to investigate the transformative potential of a social story intervention employing virtual reality technology, specifically targeting the enhancement of communication skills among students with autism spectrum disorders. The focal point of this inquiry centered on a 13-year-old male student diagnosed with autism spectrum disorder, who was enrolled in a specialized educational institution. The research design encompassed a meticulous multiple baseline across behaviors design methodology, comprising distinct phases including baseline, intervention, and maintenance. We identified the student‘s challenging behavior and crafted a virtual reality-based social story to help him learn communication skills that can be properly used in the class. The data was collected and analyzed utilizing the event recording method, enabling the precise measurement of behavioral frequency. The findings of this intervention revealed significant improvements across all three domains of communication skills: responsiveness, initiation, and requests. Notably, the innovative utilization of virtual reality within the social story framework appeared to harness the unique visual information processing strengths commonly found in individuals with autism spectrum disorders, contributing to the positive outcomes observed. This investigation’s outcomes have noteworthy implications, suggesting that a social story intervention centered on virtual reality technology can serve as a potent and efficacious strategy for augmenting the communication skills of students grappling with autism spectrum disorders. The results underscore the potential of technology-assisted interventions to usher in positive transformations in the realm of special education and support for these students.

This study describes a theoretical analysis about burner type and throttling type among the regeneration methods of ceramic filter trap.<br/> In this study, we compared with parameters of regeneration characteristics about these two reqeneration types. <br/> So throttling type showed more security than burner type in the thermal stress aspect and this regeneration simulation program can ingorm the data of optimal design of filter trap and actual application test.

The generation of cartilaginous tissues using pluripotent stem cells is one possible solution for cartilage regeneration, as the mass production of cartilaginous cells is required to cure defects and diseases of cartilage. Parthe-nogenetic ESCs (PESCs) are a useful stem cell source. The defects in full-term development of this cell type enable researchers to avoid ethical concerns. Moreover, in female patients, if the PESCs are derived from the patient’s own oocytes, the cells will have that patient's genetic information. Here we present data demonstrating that PESCs can be differentiated into chondrogenic cells with induction medium containing multiple factors such as ascorbic acid, dexam-ethasone, bone morphogenetic protein-2 (BMP-2), and transforming growth factor- beta (TGF-β), and the induction can be promoted by exogenous insulin-like growth factor 2 (IGF-2), which is silenced in PESCs. Before chondrogenic induction of the PESCs, floating aggregates called embryoid bodies (EBs) were formed in suspension culture for 5days. In the first series of experiments, the EBs were transferred to chondrogenic induction medium that contained serum, ascorbic acid, dexamethasone, BMP-2, BMP-4 and TGF-β, and the cells were cultured for an additional 21 days using either a two-dimensional (2D) or three-dimensional (3D) culture system. In the second series of experiments, we investigated the effects of different concentrations of IGF-2 supplementation (0, 1, 10 or 100ng/mL) in a 3D culture system. After induction, the gene expression of the chondrocyte-specific markers, CHORDIN-LIKE 1, COLLAGEN-2, AGGRECAN, DECORIN, MMP13 and PAX-1, was analyzed real-time PCR. In experiment 1, the cells from all experimental groups after chondrogenic induction exhibited morphology characteristic of murine chondrocytes as con-firmed by Alcian blue and Safranin O staining. The 3D culture system showed increased induction of chondrogenic differentiation in comparison to the 2D system. In experiment 2, the group supplemented with 100ng/mL IGF-2showed the highest induction of chondrogenic differentiation in comparison to other groups. The expression levels of all genes were 7-to 35-fold higher in the group treated with 100ng/mL IGF-2 in comparison to the IGF-2-free control. The result showed that supplementation with the IGF-2 improves the efficiency of chondrogenic differentiation of murine PESCs in a 3D culture environment. The addition of IGF-2 may re-activate imprinted genes in PESCs that are only expressed in the paternal genome and are normally silent in PESCs. Our findings suggest that compensative sup-plementation of imprinting factor(s) may improve the efficiency of differentiation to specific lineages in monogenic stem cells.
