      • Alterations in Surface Proteins and the Action of Fibroblast Growth Factor in Differentiating Chick Myoblasts

        In order to study the cell fusion mechanism in chick embryo muscle cells in culture, the changes of cell surface proteins and the effect of FGF during myogenesis were analyzed by using lactoperoxidase-catalyzed iodination and purified FGF addition to the culture medium, respectively. Both quantitative and qualitative changes were observed in $^{131}I$-labelled surface proteins during myogenesis. The proteins with molecular weight of 245K and 66K decreased as the fusion proceeded, and 130K and 93K proteins appeared in the course of fusion. The 245K protein seems to be the LETS protein, and the appearance of 130K and 93K proteins may be due to proteolytic cleavage of the 245K protein. At the same time, four labelled proteins were detected in the cultured medium, which seem to be released from the membrane surface proteins by proteolysis in the course of myoblast fusion. In order to examine the effect of mitogen on myoblast fusion, FGF was partially purified from bovine brain. When FGF was added to culture medium, myoblast exhibited an increase in DNA synthesis and the delay in fusion. These results suggest the possibility that the onset of myoblast fusion is due to the depletion of replication·promoting components in culture medium. The changes of cell surface protein, the engagement of these proteins in myoblast fusion, and the effect of environmental factors on myoblast fusion were discussed. 세포의 융합기작의 일단을 규명하기 위하여 계배 근원세포의 막표면 단백질을 lactoperoxidase를 써서 iodination하여 융합 전후의 막단백질의 변화를 분석하였고, fibroblast growth factor(FGF)를 소의 뇌로부터 추출하여 근원세포의 DNA 함성과 융합에 미치는 영향을 검토하였다. 융합과정 중에 막표면 단백질의 정성적 및 정량적 변화를 볼 수 있었는데, 융합이 진행됨에 따라 245K 와 66K 단백질은 점차 감소하였고 130K와 93K 단백질은 새로 출현하였다. 245K 단백질은 large, external, transformation-sensitive 단백질로 추정되며, 이 단백질의 가수분해 결과 130K와 93K 단백질이 출현하는 것으로 추정되었다. 또한 배양액 내에서 발견된 4개의 표지단백질도 막단백질의 가수분해 산물로 생각되었다. FGF를 배양액에 첨가하였을 경우 근원세포의 DNA 합성률은 증가를 보인 반면 융합은 지연되는 현상을 관찰할 수 있었다. 이러한 사실은 근원세포의 융합이 배양액 내에 존재하는 DNA 합성을 촉진하는 물질이 고갈됨에 따라 개시되는 것이 아닌가 하는 점을 암시해 주는 것으로 사료되었다. 근원세포의 융합과정에서 나타나는 막표면 단백질의 변화와 외적 요인이 세포융합에 미치는 영향에 대해 논의하였다.

        배양한 계배흉근의 근원세포의 증식과 분화에 대한 Chemically defined Media 의 효과

        Chemically defined media (CDM) are described that allow substantial proliferation and subsequent differentiation in primary cultures of cells derived from embryonic chick breast muscle. MCDB 201 (MCDB) and DME are found to be superior to RPMI 1640 (RPMI) as a basal medium, and MCDB specifically promotes cell proliferation whereas DME stimulates cell fusion. Ovotransferrin is found to be more effective than human transferrin and is effective only in Fe-complexed form but not in Fe-free form. The most suitable CDM appears to be that contains MCDB as a basal medium, as judged by criteria such as fusion index, cell proliferation, rate of DNA and protein synthesis, pattern of muscle-specific protein synthesis, and cell morphology.

      • Comparative Study of Chemically Defined Media on the Proliferation and Differentiation of Chick Myoblasts in Culture

        계배 흉근에서 추출한 근원세포의 증식과 분화를 연구하기 위한 chemically defined media(CDM)를 개발하여 그 효과를 비교검토하였다. 기본배지로서는 MCDB 201과 DME가 RPMI 1640에 비해 양호하였으며 특히 MCDB는 근세포의 증식에, 그리고 DME는 근세 포의 융합에 유효함을 나타냈다. Ovotransferrin은 human transferrin에 비해 효과가 컸으며 ovotransferrin은 Fe-complex형에서만 유효하였다. 세가지 CDM을 비교하기 위해서 융합지수, 세포증식률, DNA와 단백질 합성률, 근특이 단백질의 합성양상, 그리고 세포의 형태등의 관점에서 판단하였을때 MCDB 201을 기본배지로 사용한 CDM이 가장 양호한 것으로 나타났다.       • A Study on the CdR Aminohydrolase in Drosophila melanogaster

        CdR aminohydrolase의 활성을 老化와의 관련하에서 보기 위해서 초라피(Drosophila melanogaster)의 이효소의 활성을 발생단계별로 측정하여 다음의 결과를 얻었다. 1. 초파리에서 CdR이 분해되는 과정은 쥐나 생쥐와 마찬가지로 $CdR \\to UdR \\to U$임을 알수 있었다. 2. CdR-aminohydrolase외 활성은 유충과 번데기에서 검출이 불가능하였고 성체에서만 검출되었다. 3. 성체의 효소활성은 젊은 개체에 비해서 늙은 개체에서 증가를 보였다. 4. 실험의 결과는 CdR-aminohydrolase의 활성이 초파리의 老化또는 발생단계와 관련성이 있음을 시사하였다. CdR aminohydrolase activity in varying developmental stages of Drosophila melanogaster was studied in an attempt to correlate with ageing. The results obtained are as follows: 1. The catabolic pathway of CdR in Drosophila melanogaster seemed to be $CdR \\to UdR \\to U$. 2. The enzyme activity was demonstrated in the adults and no activity was observable in both larva and pupa. 3. The enzyme activity of the adult was found to be higher in older flies than in younger ones. 4. The results were of suggestive of a possibility that enzyme activity might be correlated with ageing and/or developmental stages of Drosophila melanogaster.

        중학생의 일상스트레스와 스트레스 대처방식이 학교적응에 미치는 영향

        The purpose of this study was to examine the effects of middle school students’ daily life stress and problem-focused coping, emotion-focused coping, wishful thinking, and social support seeking on their school adjustment. Data were collected from 356 students selected purposefully from 6 middle schools in Daejeon Metropolitan City through self-administered survey using structured questionnaires. SPSS Statistics 20.0 software was used for descriptive statistics and hierarchical multiple regression analysis. The findings indicated that (1) the lower the degree of middle school students’ daily life stress was, the higher the degree of their school adjustment was; (2) out of stress coping strategies, the more problem-focused coping and social support seeking were used, the higher the degree of their school adjustment was; (3) there were no moderating effects of coping strategies on the relationship between middle school students’ daily life stress and their school adjustment. Based on the findings, possible implications for improving adolescent’s school adjustment were discussed.

        Train control by radio based communication is one of the interesting new fields in train signal. The radio based control has more benefits, low headway and low construction and maintenance cost, than conventional control. In this paper, a safe and efficient train operation is introduced. Triple radio communications, train to train, train to station, and train to central traffic control, are used to increase the reliability. One of these communications channels has a fault; the others can take the functions of it. Absolute position of a train is transmitted to station via radio communication. In the station, the interlocking mechanism should be activated as the legacy.

        남녀 고등학생 청소년이 지각한 진로장벽이 정신건강에 미치는 영향과 긍정심리자본의 조절 효과

        This study aimed to examine the effect of career barriers as risk factors for the mental health of male and female high school students. This study also aimed to examine the moderating effect of positive psychological capital on the relationship between career barriers and mental health of male and female high school students. One hundred and ninety-five male and 218 female students in the second year of 8 high schools agreed to respond to a survey conducted with the high schools’ cooperation in Daejeon Metropolitan City. SPSS 26.0 software and PROCESS macro version 3.5 were used for the data analysis. The t-test, cross-tabulation analysis, correlation analysis, and hierarchical regression analysis were conducted as part of the study. In particular, the moderating effect’s significance was analyzed by the specific value selection method using the PROCESS macro program and the Johnson-Neyman method. First, the results of the study confirmed that higher perceived career barriers for both male and female students led to significantly worse mental health status. Second, as for the moderating effect of positive psychological capital on the relationship between career barriers and mental health, it was found that the positive psychological capital moderated the relationship between perceived career barriers and mental health only in male students. Finally, practical suggestions for improving the mental health of male and female high school adolescents were discussed based on the findings.
