본 연구는 학령전기 아동 어머니의 모아상호작용에 대한 인식을 분석하고 파악하기 위하여 시도되었으며, 우리나라 학령전기 아동 어머니들은 아동의 발달을 촉진하고 어머니에게 민감한 아동과 상호작용 하는 것을 중요한 모아상호작용으로 인식하고 있었으며, 어머니가 일방적으로 아동에게 상호작용을 해 주는 것이 아니라 아동도 어머니와 동등한 입장에서 능동적으로 상호작용하는 것을 모아상호작용으로 인식하고 있었다.

Streptomyces Peucetius에서의 ${\varepsilon}$-rhodomycinone 추출 및 이종균주에서의 rhodomycin D 생산 연구

Streptomyces peucetius가 생산하는 anthracycline 계열의 doxorubicin은 치료목적으로 사용되는 중요한 항암제 중 하나이다. Doxorubicin은 rhodomycin D에서부터 몇 단계의 생합성 과정을 더 거쳐 생산되는데, 생물학적 활성을 갖기 위해서는 deoxy-sugar의 전이가 반드시 일어나야 한다. 본 논문에서는 이종균주인 Streptomyces venezuelae에 11개의 유전자를 형질 전환하여 TDP-L-daunosamine를 생산하고 이것을 ${\varepsilon}$-rhodomycinone에 전이하여 rhodomycin D를 생산하는 연구를 수행하였다. S. peucetius 유래의 7개 유전자 dnmU, T, J, V, Z, Q, S.를 당 합성 및 전이를 위해 plasmid 형태로 전이하였으며, S. venezuelae의 desIII, IV와 doxorubicin 내성 유전자인 drrA, B는 chromosomal DNA에 삽입하였다. Aglycone 기질인 ${\varepsilon}$-rhodomycinone을 확보하기 위하여 6L의 고체 배지에 S. peucetius를 배양하여 유기용매로 추출하고 preparative HPLC로 분리 정제하였다. 결과적으로 이종균주인 S. venezuelae에서 ${\varepsilon}$-rhodomycinone에 당 전이가 일어난 생산물을 확인함으로써 deoxy-sugar의 생합성 및 전이에 필요한 최소한의 유전적 정보를 확인할 수 있었다. 또한, 유사서열 단백질 모델링을 통하여, 최초로 당 전이 반응에 필수적인 도움효소 DnrQ의 구조를 예측하였다. Anthracycline antibiotics doxorubicin (DXR) is clinically important cancer therapeutic agent produced by Streptomyces peucetius. DXR result by further metabolism of rhodomycin D (RHOD) and require a deoxy-sugar component for their biological activity. In this study, production of TDP-L-daunosamine and its attachment to ${\varepsilon}$-rhodomycinone (RHO) to generate RHOD has been achieved by bioconversion in Streptomyces venezuelae that bears eleven genes. S. peucetius seven genes (dnmUTJVZQS) were transformed by plasmid and S. venezuelae two genes desIII, IV and two more S. peucetius drrA, B genes were integrated into chromosomal DNA. To generate the feeding substrate RHO, 6L S. peucetius grown on agar plate was harvested, extracted with organic solvent and then purified using preparative HPLC. Recombinant S. venezuelae grown on agar plate containing RHO was harvested and its n-butanol soluble components were extracted. The glycosylated product of aromatic polyketide RHO using heterologous host S. venezuelae presents the minimal information for TDP-L-daunosamine biosynthesis and its attachment onto aglycone. Moreover, the structure of auxiliary protein, DnrQ, was predicted by fold recognition and homology modeling in this study. This is a general approach to further expand of new glycosides of antitumor anthracycline antibiotics.

돼지 血淸 alpha-1-protease inhibitor 의 分離

For the preliminary step to characterize the structural and immunological relationship of alpha-1-protease inhibitor molecule among mammalian species, alpha-1-protease inhibitor was purified from porcine serum through the procedures of salt fractionation, Cibacron blue F3GA-agarose, DEAE-cellulose, Concanavalin A-Sepharose and Sephadex G-100 gel chromatography. The specific inhibitory activity of purified porcine alpha-1-protease inhibitor against porcine trypsin was 9.59 unit/㎎ of protein, and the recovery was 26% with purification factor of 105-fold. The isolated alpha-1-protease inhibitor showed a single band on alkaline polyacrylamide gel electrophoresis and a single precipitin arc against rabbit anti-porcine serum on immunoelctrophoresis, which indicates that the alpha-1-protease inhibitor preparation is pure. This alpha-1-protease inhibitor also displayed a single band on SDS-polyacrylamide gel electrophoresis. The molecular weight of porcine alpha-1-protease inhibitor was estimated to be about 60,000 by SDS-polyacrylamide gel electrophoresis, and no immunological identity with human alpha-1-protease inhibitor was found.

이 연구는 인간중심상담의 정수라고 할 수 있는 인간관계의 필요충분조건들의 관계에 대해 탐색하였다. 그런데 이 세 가지 조건들의 관계는 진정성의 내용을 무엇으로 하느냐에 따라 크게 달라짐을 알 수 있었다. Rogers가 인간중심상담의 초기에 강조했던 일치성을 진정성의 내용으로 보는 경우, 세 가지 필요충분조건들은 상담자가 활용할 수 있는 도구들로서 균등한 중요성을 가진 것으로 판단된다. 반면, Rogers가 후기에 강조했던 현전재성을 진성성의 내용으로 삼는 경우, 진정성은 세 가지 조건의 중심 위치에서 공감과 무조건적 긍정적 존중을 활용하며 지속적으로 성장하는 자아실현의 과정이라고 말할 수 있다. 그리고 이 경우, 필요충분조건 세 가지는 상담자와 청담자 모두를 자아실현의 과정에 참여시키는 생산적 상담관계를 구성한다는 주장이 가능하다.

In this study, the influence of pressure assisted mild thermal inactivation (PAMTI) on E. coli ATCC 10536 was examined at 200 MPa and temperature range of 20-50℃ . Inactivation rate significantly increased (p<0.05) as temperature and time increased at 200 MPa. The maximum inactivation (7.91 log reduction) was obtained at 50℃ for 30 min under 200 MPa, which meant the complete inactivation of E. coli ATCC 10536. Inactivation kinetics were evaluated with the first order inactivation rate (k), activation energy (Ea), thermal death time (TDT), and z value. Kinetic parameters were significantly (p<0.05) influenced by variation temperature of PAMTI. In this study, the synergistic effect of pressure and temperature were found in the inactivation of E. coli ATCC 10536 through PAMTI.

아동 · 청소년의 행복 보장을 위한 독일의 교육복지 개념 고찰

사회적 양극화 현상으로 사회 계층별, 도시와 농산어촌 간의 지역 격차의 심화뿐만 아니라, 한부모가정, 다문화 가정, 조손 가정 등 신소외 계층이 증가해 가정의 해체와 기능의 상실이 현저해 지고 있다. 따라서, 정부는 아동 · 청소년의 불평등한 교육기회를 개선하고 소외계층의 교육적 성장 지원을 위해 2006년부터 전국적으로 교육복지사업을 실시하고 있다. 그러나, 교육복지에 대한 개념 연구는 정책 입안 연구에 치중되어 있어, 교육복지에 대한 개념이나 방향 그리고 법적 근거는 불명확한 상태이다. 따라서 본 연구에서는 모든 아동 · 청소년이 자신이 태어난 사회적 계층에 좌우되지 않고 자신의 잠재력을 계발해 자율적인 삶의 주체로 성장하도록 교육 기회의 향상을 지원하고, 소외된 아동 지원 뿐만 아니라 부모의 양육능력을 강화시켜 공적으로 행복을 보장하는 교육복지의 개념을 독일의 사회교육학(Sozialpaedagogik)과 아동 · 청소년지원법(KJHG)을 중심으로 살펴보고, 한국에서의 시사점을 제언하였다.
