        In-line Monitoring of an Oxide Ion in LiCI Molten Salt Using a YSZ Based Oxide Ion Selective Electrode

        The electrode potential characteristics of a YSZ based membrane metal oxide electrode have been studied in molten LiCL at $700^{\circ}C$ by the potentiometric method. The electrode exhibited a good potential response to log[$O^2$] and data reproducibility. The calibration plot (potential vs. log[$O^2$] was found to be linear, obeying the Nernst equation. The electrode potential showed a good reversibility corresponding to increase/decrease of the oxide ion present in the molten LiCl. The physical and chemical durability appeared to be sound after several repeated uses, resulting in reproducible results. However, "the proposed electrode" failed when metallic Li was present in the melt.

        HPLC를 이용한 농식품 중 당류의 신속정량 분석

        HPLC를 이용하여 농식품에 함유된 fructose, glucose, sucrose, maltose, lactose 등 5종의 당류에 대한 신속하고 정확한 동시분석법을 확립하기 위하여 시험을 실시하였다. 수용성 당류의 HPLC 분석조건은 RI 검출기를 이용하여 column은 ZORBAX Carbohydrate (4.6 ㎜ ID×250 ㎜, 5㎛)를 이동상은 75% ACN을 사용하였고 칼럼온도 35℃, 시료 주입량은 10 μL, 유속은 1 mL/min의 조건으로 30분간 분석하여 방해 peak없이 5종의 표준용액을 분리하였다. 표준용액 검량선의 R<sup>2</sup>=0.999 이상으로 5종 모두 직선성이 우수하였고, 표준용액의 크로마토그램을 이용하여 분석의 정량한계와 검출한계를 산출하였다. 농식품의 당류 추출방법을 구명하기 위하여 표준인증물질 BCR644을 이용 추출 방법별 정확도를 비교한 결과, 증류수 100% 추출처리에서 fructose 95.7%, sucrose 98.7%, lactose 102.7%로 가장 우수하여 농산물 당류 추출시 적용하였다. 내부표준물질인 분유를 이용하여 반복성과 재현성을 확인하여 본 분석법에 대한 정밀도를 검증하였고 reference value 값으로 glucose는 1.17 g/100 g, maltose는 0.85 g/100 g, lactose는 45.54 g/100 g으로 설정하였고, 허용한계 95% 수준(2*SD)을 경고한계로, 허용한계 99.7% 수준(3*SD)을 거절한계로 산출하여 품질관리차트를 작성하고 농식품 당류 분석에 활용하였다. To determine sugars content of agricultural products and foods, simultaneous quantitative analysis was carried out on fructose, glucose, sucrose, maltose, and lactose by HPLC-RI. Analysis conditions were set as column ZORBAX car-bohydrate (4.6 ㎜ ID×250 ㎜, 5 ㎛), the mobile phase of 75% ACN, the column temperature of 35℃, sample injection amount of 10 μL and the flow rate of 1 mL/min. Five standard solutions were isolated without interfering peak within 30 minutes and the calibration curves of standard were confirmed excellent linearity from 0.10% to 1.00% with R<sup>2</sup>≥0.999. Based on the chromatogram of the standard solution, the limit of quantification (LOQ) and the limit of detection (LOD) values were calculated. The accuracy of the analytical values were highest at 100% water extraction method to the fructose 95.7%, sucrose 98.7%, lactose 102.7% respectively, compared with reference value of a certified reference material (BCR644), by applying the four solvent extraction methods. Using an in-house quality control material (infant formula), repeatability and reproducibility values of this experiment were verified on the basis of AOAC guideline and reference values were set up at 1.17 g/100g of glucose, 0.85 g/100g of maltose, and 45.54 g/100g of lactose. Quality control charts were drawn up and used for sugars analysis of agricultural products.

        Sessile organism, plants constitutively challenged with pathogens have been developed various strategies for protection, such as preformed and inducible defense mechanisms. Receptor-like Proteins(RLPs) play critical roles in defense response as well as in plant development and growth. The domain structure of RLPs consists of extracellular leucine–rich repeats, a transmembrane domain, and a short cytoplasmic tail. Here, we identified putative 170 RLP genes from pepper genome using in-house bioinformatics pipeline. The distribution of RLPs on pepper pseudomolecule showed uneven spread and a number of RLPs were physically clustered by tandem array in the specific chromosome. Motifs analysis of pepper RLPs showed conserved LRR sequences (LxxLxxLDLxxNxxxGxIP). To understand further functional and evolutionary characteristics, evolutional relationship and gene profiling analysis are on progress.

      • Dehydration of Glycerin to Acrolein Over H<sub>3</sub>PW<sub>12</sub>O<sub>40</sub> Catalyst Supported on Mesoporous Titania

        <P>H3PW12O40 catalysts supported on mesoporous titania (XH3PW12O40/MT (X = 5, 10, 15, 20, 25, and 30)) were prepared with a variation of H3PW12O40 content (X, wt%), and they were applied to the dehydration of glycerin to acrolein. The prepared catalysts were characterized by FT-IR spectroscopy, X-ray diffraction, N-2 adsorption-desorption, and pyridine-adsorbed in-situ FT-IR spectroscopy analyses. The effect of H3PW12O40 content on the physicochemical properties and catalytic activities of XH3PW12O40/MT catalysts was investigated. It was found that surface area and pore volume of XH3PW12O40/MT catalysts decreased with increasing H3PW12O40 content. Bronsted acidity of Xv/MT catalysts showed a volcano-shaped trend with respect to H3PW12O40 content. The catalytic performance of XH3PW12O40/MT catalysts was closely related to the Bronsted acidity of the catalysts. It was revealed that yield for acrolein over XH3PW12O40/MT catalysts increased with increasing Bronsted acidity of the catalysts. Among the catalysts tested, 20H(3)PW(12)O(40)/MT catalyst with the largest Bronsted acidity showed the highest yield for acrolein.</P>

        Synergistic Improvement in Insulin Resistance with a Combination of Fenofibrate and Rosiglitazone in Obese Type 2 Diabetic Mice

        Peroxisome proliferator-activated receptor (PPAR) ${\alpha}$, which is abundant in the liver, increases lipoprotein lipase activity, resulting in a decrease of triglyceride (TG) levels. $PPAR{\gamma}$, which is abundant in adipose tissue, stimulates adipocyte differentiation and adipogenesis, and results in an increase in insulin sensitivity. Fenofibrate, a $PPAR{\alpha}$ agonist, is commonly used to treat dyslipidemia, and rosiglitazone, a $PPAR{\gamma}$ agonist, is effective in improving glycemic control. To examine the synergistic effects of rosiglitazone in combination with fenofibrate, an obese type 2 diabetes mellitus (DM) mouse model was established by the combined administration of streptozotocin and nicotinamide and fed on a high-fat diet (35% of energy as fat) for 3 weeks. The mice had significantly higher plasma glucose concentrations and insulin resistance, as examined by an oral glucose tolerance test and insulin challenge test compared with normal mice. After establishing a dose-response curve for each drug, the drugs were orally administered for 3 weeks either alone or in combination. After individual administration of fenofibrate, HDL cholesterol levels significantly increased, and plasma glucose and TG levels decreased in obese type 2 DM mice. The individual administration of rosiglitazone showed increased insulin resistance (QUICKI). However, HDL cholesterol and TG levels were not significantly changed. In a combination of fenofibrate at 25 mg/kg and rosiglitazone at 1.25 mg/kg there was a decrease in plasma glucose and TG levels, and a combination of fenofibrate at 50 mg/kg and rosiglitazone at 2.5 mg/kg showed an increase in plasma HDL cholesterol levels. Moreover, parameters related to insulin resistance (HOMA-IR) and insulin sensitivity (QUICKI) were improved significantly. Thus, our results show that combination therapy with lower doses of fenofibrate and rosiglitazone ameliorates the type 2 DM condition to a greater extent than high doses of either individual monotherapy.

        Background/Aims: Endoscopic biliary drainage is an established mode of treatment for acute suppurative cholangitis. Endoscopic retrograde biliary drainage (ERBD) using a plastic stent is suggested to be as effective as endoscopic nasobiliary drainage (ENBD) for temporary biliary drainage in acute suppurative cholangitis. However, there were a few studies comparing between ERBD and ENBD in acute suppurative cholangitis. We compared the safety and efficacy of ERBD and ENBD in patients with acute suppurative cholangitis. Methods: From April to August 2007, forty patients with acute suppurative cholangitis underwent endoscopic biliary drainage with ENBD (22) and ERBD (18). We retrospectively analyzed their safety and efficacy. Results: Mean duration of indwelling catheter were 4.3 days in the EMBD group and 4.6 days in the ERBD group (p=0.67). Leukocyte counts became normal after a median time of 2.7 days in the ENBD group and 1.7 days in the ERBD group (p=0.18). Total bilirubin became normal after a median time of 2.8 days in the ENBD group and 3.4 days in the ERBD group (p=0.44). There were 6 ERCP-related complications in the ENBD group and 10 in the ERBD group (p=0.11). The complications were symptomatic pancreatitis (2 in ERBD), bleeding (3 in ERBD and 4 in ENBD), and increased amylase (2 in ENBD and 5 in ERBD). There were no cases of occlusion or migration of ENBD or ERBD. But, one patient pulled out the ENBD catheter. There was no difference of frequency of procedure (p=0.06) and hospital stay (p=0.79) between the 2 groups. Conclusions: ERBD is safe and effective alternative method of decompressing the bile duct in acute suppurative cholangitis.

        Insight Into Genes Involved in the Production of Extracellular Chitinase in a Biocontrol Bacterium Lysobacter enzymogenes C-3

        The chitinase producing Lysobacter enzymogenes C-3 has previously been shown to suppress plant pathogens in vitro and in the field, but little is known of the regulation of chitinase production, or its role in antimicrobial activity and biocontrol. In this study, we isolated and characterized chitinase-defective mutants by screening the transposon mutants of L. enzymogenes C-3. These mutations disrupted genes involved in diverse functions: glucose-galactose transpoter (gluP), disulfide bond formation protein B (dsbB), Clp protease (clp), and polyamine synthase (speD). The chitinase production of the SpeD mutant was restored by the addition of exogenous spermidine or spermine to the bacterial cultures. The speD and clp mutants lost in vitro antifungal activities against plant fungal pathogens. However, the gluP and dsbB mutants showed similar antifungal activities to that of the wild-type. The growth of the mutants in nutrient rich conditions containing chitin was similar with that of the wild-type. However, growth of the speD and gluP mutants was defective in chitin minimal medium, but was observed no growth retardation in the clp and dsbB mutant on chitin minimal medium. In this study, we identified the four genes might be involved and play different role in the production of extracellular chitinase and antifungal activity in L. enzymogenes C-3.

        Original Article : Routine Mucosal Closure with a Detachable Snare and Clips after Endoscopic Submucosal Dissection for Gastric Epithelial Neoplasms: A Randomized Controlled Trial

        Background/Aims: The aim of this study was to determine whether the routine closure of mucosal defects after endoscopic submucosal dissection (ESD) can enhance mucosal healing and reduce ESD-associated bleeding. Methods: Patients with gastric epithelial neoplasias and no obvious submucosal invasion were prospectively enrolled. Mucosal defects were left untreated in the control group. In the study group, mucosal closure was attempted with a 2-channel endoscope, a detachable snare, and clips. All participants received a second-look endoscopy the day after ESD, and coagulation therapy was administered to patients with visible vessels and active bleeding points. Results: Fifty-two patients were enrolled in the study, and 26 patients were assigned to each group. Complete mucosal defect closure occurred in 16 patients (61%) in the study group; incomplete closure occurred in 8 patients (31%) in the study group, and failed closure occurred in 2 patients (8%). Coagulation therapy at the second-look endoscopy was performed more often in the control group than in the study group (31% vs 4%, p=0.024). There were no significant differences in the incidence of immediate or delayed bleeding or in the twoweek decrease in hemoglobin between the groups. The prevalence of open ulcers after 8 weeks was significantly lower in the study group than in the control group (18% vs 43%, p=0.012). Conclusions: Routine mucosal closure after ESD supports earlier healing of artifi cial ulcers. A larger-scale trial is necessary to determine whether mucosal closure can reduce ESD-associated bleeding. (Gut Liver 2011;5:454-459)

        Comparative analysis of the microbial communities in raw milk produced in different regions of Korea

        Objective: The control of psychrotrophic bacteria causing milk spoilage and illness due to toxic compounds is an important issue in the dairy industry. In South Korea, Gangwon-do province is one of the coldest terrains in which eighty percent of the area is mountainous regions, and mainly plays an important role in the agriculture and dairy industries. The purposes of this study were to analyze the indigenous microbiota of raw milk in Gangwon-do and accurately investigate a putative microbial group causing deterioration in milk quality. Methods: We collected raw milk from the bulk tank of 18 dairy farms in the Hoengseong and Pyeongchang regions of Gangwon-do. Milk components were analyzed and the number of viable bacteria was confirmed. The V3 and V4 regions of 16S rRNA gene were amplified and sequenced on an Illumina Miseq platform. Sequences were then assigned to operational taxonomic units, followed by the selection of representative sequences using the QIIME software package. Results: The milk samples from Pyeongchang were higher in fat, protein, lactose, total solid, and solid non-fat, and bacterial cell counts were observed only for the Hoengseong samples. The phylum Proteobacteria was detected most frequently in both the Hoengseong and Pyeongchang samples, followed by the phyla Firmicutes and Actinobacteria. Notably, Corynebacterium, Pediococcus, Macrococcus, and Acinetobacter were significantly different from two regions. Conclusion: Although the predominant phylum in raw milk is same, the abundances of major genera in milk samples were different between Hoengseong and Pyeongchang. We assumed that these differences are caused by regional dissimilar farming environments such as soil, forage, and dairy farming equipment so that the quality of milk raw milk from Pyeongchang is higher than that of Hoengseong. These results could provide the crucial information for identifying the microbiota in raw milk of South Korea.
