Polyphenol oxidase plays major rele in the browning of plant tissues. The enzyme, polyphenol oxidase was partially purified from an extract of Lotus root by ammonium sulfate fractionation, which resulted in a 148 fold increase in specific activities. The enzyme was relatively heat stable, The substrate specificity of the Lotus root polyphenol oxidase showed high affinity toward pyrogallol, gallic acid and catechol, but not towards monophenol. Potassium cyanide, cysteine, potassium metabisulfite, sodium diethyldithiocarbamate were potent inhibitors. It seems that the enzyme was partially purified from Lotus root is polyphenol oxidase containing copper.
