배스(큰입배스, Micropterus Salmoide)는 국내 생태계 교란종으로 이용가치가 없어 구입 후 전량 폐기되고 있다. 버려지는 배스를 활용하는 것은 생태계 와 자원 재활용 측면에서 모두 중요하기 때문에 배스의 이용가치를 높일 수 있는 방법 개발이 필요하다. 본 연구는 배스에서 콜라겐을 추출하고 이 물 질의 세포독성 및 조직병리학적 안정성을 체외 및 생체 내에서 평가하는 것 을 목표로 한다. 배스 콜라겐을 추출하기 위해 먼저 배스를 작은 조각으로 자른 다음 80℃의 증류수에 1시간 동안 열수 추출한다. 추출액을 200 mesh (≤ 127 μm)에 여과한 후에 상온에서 3000 rpm (1736 g) 조건으로 15분간 원심분리한다. 상층액을 획득 후 0.1%(v/v)의 알칼레이즈를 60℃에서 30분간 처리한 후 80℃에서 20분간 비활성화한다. 이후 60℃의 열풍 건조기에서 하 룻밤 건조하고 분말화한다. SDS-PAGE 을 실시하여 배스 추출물에 α-chain 은 100~150 kDa 사이에 존재했고, β-chain 은 250 kDa, γ-chain 은 280 kDa 부근에 위치한 것을 확인하였고 알칼레이즈를 처리했을 때 콜라겐이 10 kDa 이하로 저분자화되는 것을 밴드를 통해 확인하였다. Collagen assay kit 을 사용하여 배스 추출물 내 단백질 총량 대비 콜라겐은 약 60% 존재하 는 것을 확인하였다. DPPH-assay 을 진행한 결과 배스 콜라겐(Bass collagen, BC)와 알칼레이즈 처리 배스 콜라겐(Alcalase treated Bass collagen, ABC)에서 각각 66%, 78%로 항산화 효과가 나타났다. 배스 추출물 을 처리한 RAW264.7 세포에서 세포독성은 발견되지 않고, BC 와 ABC에서 각각 151.65%, 165.04%의 높은 cell viability 을 나타냈다. 배스 추출물을 4주 동안 2일 간격으로 급이한 ICR 에서 적출한 장기는 형태학적 이상을 보이지 않았다. 최종적으로 배스 추출물은 세포 독성 및 장기 독성을 나타내지 않 고 높은 콜라겐 함량과 항산화능을 포함하였다. 따라서 본 연구에서는 폐기 되는 배스로부터 콜라겐을 추출해 배스를 산업적으로 활용할 수 있는 기술 을 개발하였다.

The Largemouth Bass (Micropterus Salmoide) is an ecosystem-disrupting species in South Korea and has no use value, so it is completely discarded after purchase. Using the discarded bass is important in terms of resource recycling, so it is necessary to develop a method for increasing the use value of the bass. This study aims to extract valuable fish collagen from bass and assess the cytotoxic and histopathological stability of this substance both in vitro and in vivo. In order to extract bass collagen, the bass was first cut into small pieces and then immersed in distilled water at 80 ℃ for 1 hour, then the aqueous solution was filtered through 200 mesh (≤ 127 μm). The Solution was centrifuged at 3000 rpm (1736 g) for 15 minutes at room temperature. After taking the supernatant, 0.1% (v/v) of alcalase was treated at 60℃ for 30 minutes and then inactivated at 80℃ for 20 minutes. The solution was dried overnight SDS-PAGE analysis confirmed the presence of α-chains between 100-150 kDa, β-chains at 250 kDa, and γ-chains around 280 kDa in the bass extract. In addition, bass collagen became a low molecular weight of below 10 kDa after alcalase treatment. Using a Collagen Assay Kit, The purity of the bass collagen was approximately 60% of the total protein content in the bass extract. DPPH-assay showed that the antioxidant activity of bass collagen(BC) and alcalase-treated bass collagen(ABC) was 66% and 78%, respectively. RAW264.7 cells treated with bass extract showed no cytotoxic effects. Both BC and ABC demonstrated enhanced cell viability, with values of 151.65% and 165.04%, respectively. The organs extracted from the ICR fed with the bass extract for 4weeks did not show any major morphological abnormalities. As a result, the bass extract demonstrated high collagen content and antioxidant capabilities without indicating cellular or organ toxicity. Thus, this study developed a technique for extracting collagen from discarded bass for potential industrial applications.
