CBR1은 cytosolic short-chain dehydrogenase/reductases 그룹에 속하며, NADPH와 함께 탈수소 반응을 하는 metabolic enzyme으로 인간의 간, 심장, 위, 신장조직 등에 광범위 하게 퍼져 있다. 특히 이 효소는 Anthracycline계 항암제인 doxorubicin 이나 daunorubicin이 생체내로 들어왔을때 각각의 C-13번 카보닐 그룹을 알코올로 환원시켜 doxorubiciol과 daunorubicinol을 생성한다. 이로 인해 항암제인 doxorubicin과 daunorubicin이 감소하게 되며, 항암 효과 역시 감소하게 된다. 또한 Anthracycline계 C13번 카보닐 그룹의 알코올화는 심장 독성을 유발시키는데 이 약을 복용한 많은 환자들에게서 울혈성 심부전 같은 치명적인 부작용이 나타났다. 이러한 문제를 해결하기 위해 Anthracyclin계 C13번 카보닐 그룹의 알코올화에 관여하는 CBR1을 억제시키는 억제제를 개발함으로써 항암효과를 증진시고, 심장과 연관된 부작용을 줄이는 것이 방안으로 대두되었다. 최근에는 CBR1에 선택적으로 억제성을 보이는 hydroxy-PP와 hydroxy-PP-Me가 보고되었고, 이러한 결과를 바탕으로 우리는 가장 좋은 효과를 보인 hydroxy-PP-Me를 기본 물질로 한 여러 가지 유도체들의 합성을 실시하고 cell viability가 수행하였다. 처음 총12개의 유도체들이 합성되어 test되었으며, 대부분의 물질들이 독소루비신과 함께 암세포에 가해질때 독소루비신 한 개만 넣어진 것보다 암세포를 더 많이 죽이는 것이 확인되었다. 그 중 16번 물질은 약 50%정도의 cell viability를 보이며 가장 좋은 효과가 확인 되었다. 따라서 16번의 세개의 작용기 부분 중 중요한 역할을 하는 작용기를 알아보고자 한부분씩 고정시킨 다음 다른 두 부분을 변형시켜 추가적으로 17개의 물질을 합성 하였다.
CBR1 Inhibitor과 같이 bis-amides 역시 최근 항암후보물질로 개발되어 오고 있다. 따라서 항암활성효과를 보이는 bis-amides의 합성이 수행되었다. Bis-amide는 일차 아민, 알데하이드 또는 케톤, 아이소시아나이드, 카복실산을 이용하여 한번에 원하는 물질을 생성하는 Ugi reacton이 이용되었으며, 여러 가지 유도체들을 만들어 cytotoxicity를 수행하였다. Bis-amides 유도체들 중 12번 물질이 세개의 암세포에 가장 강한 항암활성 효과를 보였으며, 이 물질은 항암후보물질 개발을 위한 lead compound로 기대 된다.

Carbonyl reductase 1 (CBR1) is enzyme of our interest since it plays a major role in metabolism of the anticancer anthracyclines. CBR1 belongs to the cytosolic short-chain dehydrogenase/reductase (SDR) family with NADPH-dependent oxidoreductase activity. It is well known to metabolize the anthracyclines such as doxorubicin and daunorubicin which are clinically highly effective in cancer therapy; CBR1 transforms them to the alcohol metabolites, leading to serious adverse effects such as cardiotoxicity in patients undergoing treatment with these drugs. Inhibition of CBR1 activity would be helpful to ameliorate the side effects of the anthracyclines so that development of CBR1 inhibitor is highly desirable.
As CBR1 inhibitors, hydroxy-PP-Me has recently developed to inhibit CBR1 and to increase the sensitivity of cancer cell lines to doxorubicin and daunorubicin treatment. So, hydroxy-PP-Me was chosen as a lead, and pyrrolo-pyrimidine derivatives were developed as CBR1 inhibitor. In the initial investigation, 12 derivatives were synthesized and most compounds induced enhancement of doxorubicin-mediated cell killing. Especially, the analog 16 reduced the cell viability by 50% which is comparable to hydroxy-PP-Me (12), and, interestingly, it showed does-dependent cell viability unlike hydroxy-PP-Me (12). Based on characteristic of the pyrrolo-pyrimidine 16 to well fit into the active site, lead optimization was further explored maintaining the key functionality of compound 16. Additional 17 derivatives were synthesized, replacing two of three functional groups to different groups whereas one kept as unchanged.
Along with CBR1 inhibitors, bis-amides were developed as candidates of anticancer agents. Since the capability of bis-amides with potent cytotoxicity against cancer cell lines is newly reported, a variety of bis-amides with different substituents were exploited. Bis-amides were synthesized simply by the Ugi multicomponent reaction in which, mostly, solid products are obtained. The Ugi reaction is time and cost efficient because it is one-pot reaction, using a carbonyl compound (ketone or aldehyde), an amine, an isocyanide and a carboxylic acid.
Among the bis-amide derivatives, compound 12 showed potent cytotoxic effects on three cancer cell lines with lower micromole concentration, thus allowing the bis-amide 12 as a potential lead for anticancer agents.
