This study analyzed the chemical composition of essential oil which is extracted from Chrysanthemum boreale as natural substance, and also measured growth, acid production, adhesion capacity, and inhibitory effect of dental biofilm formation of S. mutans. Thus, the next results were drawn.
1. In S. mutans’s growth inhibitory effects of the essential oil extracted from Chrysanthemum boreale, degrees of the growth inhibition were 12%, 45%, 100%, 100%, 100%, and 100% at the concentration (density) of 0.05, 0.1, 0.25, 0.5, 0.75, and 1 mg/ml, respectively. This means that there were similar differences statistically compared with comparison group at the concentration over 0.1 mg/ml (p<0.05).
2. Inhibitory effect of S. mutans’s acid production of the essential oil extracted from Chrysanthemum boreale appeared after being compared with pH of comparison group, after 24 hours cultivation. And, the effect’s concentration of essential oil extracted from Chrysanthemum boreale was shown similarly at 7.33±0.05 after cultivation from adding more than 0.25 ml/mg.
3. The adhesion rates for S-HA of S. mutans were 18%, 40% ,46%, 66%, 72%, and 75% similarly compared with comparison group at the concentration of 0.05, 0.1, 0.25, 0.5, 0.75, and 1 mg/ml.
4. The biofilm formation of S. mutans was observed to be suppressed compared with comparison group as the concentrations of essential oil of Chrysanthemum boreale were higher. Especially, there were clear effects from over 0.1 mg/ml.
5. The formation of S. mutans was suppressed when it is observed using a scanning electronic microscope, after the addition of the essential oil extracted from Chrysanthemum boreale. Also, S. mutans biofilm formed at the surface of artificial tooth was observed to be suppressed, at the concentration of essential oil extracted from Chrysanthemum boreale over 0.1 mg/ml.
6. After sterilization effect was observed using a confocal laser scanning microscopy, that effect of S. mutans was represented over 0.1 mg/ml of the concentration of the essential oil extracted from Chrysanthemum boreale.
7. After conducting the componential analysis of the essential oil extracted from Chrysanthemum boreale using GC/GC-MS analysis, it was composed of 87 compounds. Those components were camphor (20.89%), piperitone (5.27%), epi-sesquiphellandrene (5.16%), a-pinene (4.97%), β-pinene (4.45%), and camphene (4.19%).
Through these kinds of results, the essential oil extracted from Chrysanthemum boreale was observed to have inhibition effect of growth, acid production, adhesion on S-HA, and S. mutans formation and sterilization effect. Therefore, the main components of the essential oil extracted from Chrysanthemum boreale such as camphor, piperitone, epi-sesquiphellandrene, a-pinene, β-pinene, and camphene are presumed to have the effect against vitalizing dental caries.
