Beta-mimetic adrenergic agent is one of the most widely used tocolytic agents and this agent mainly exert it's action on the uterus by the Beta-receptor effect.
The action of this drug on to the uterus is suppression of myometrial contractility, so this agent has been used in the cases of preterm labor.
Author gas performed on experiment to clarify the mode of action of this agent with the adult non-pregnant rabbits equipped with intra-uterine recording ballons and intra-uterine pressure monitored via Stathan pressure transducers and Gilson physiograph. The results of this study were as below.
In estrogen-primed control group, the mean value of uterine contraction was freq, 2.3/min, IUP, 44.8㎜Hg(86.7%), and duration, 216 sec(712%), compared to the control group. This increased uterine contractions was inhibited by isoxsuprine to the level of freq. 2.6/min(13%), IUP, 28.8㎜Hg(20%), and duration, 163sec(513%), compared to the control group.
2. In oxytocin group, the contractions increased by freq. 5.2/min(73%), IUP 44.4㎜Hg(88%), and duration, 239sec(876%), compared to the control group. And this increased cintractility was lowered by isoxsuprine to the level of freq, 2.8/min, IUP, 37.2 ㎜Hg(58%), and duration, 169sec(590%), compared to control group.
3. In spartein sulfate group, the contractions increased by freq. 2.7/min(35%), IUP, 32.4 ㎜Hg(45%), and duration, 228 sec(805%), compared to the control group. And with isoxsuprine, the contractions decreased to freq, 1.0/min, IUP, 18 ㎜Hg, and duration, 174sec(593%), compared to the control group.
4. In ergot alkaloid group (methergine and ergonovine), there was no significant changes of uterine contractions as observed in PG F₂α, oxytocin, and sparteine sulfate, that is, the responsiveness of non-pregnant rabbit uterus to the methergine and ergonovine was not so typical in their actions.
