The present study was carried out to evaluate the effect of 5 - amino salicylic acid(5 - ASA) on the hepatotoxicity and nephrotoxicity in carbon tetrachloride(CCl_4) administered male rats. It was investigated by measuring the activity of monoamine oxidase(MAO), the levels of lipid peroxidation, and lipid accumulation in the liver and the kidney.
The results obtained were as follows.
Activity of MAO in the liver mitochondrial fraction was reduced by CCl_4 administration compared to normal group. This reduction caused by CCl_4 administration wree significantly improved by pretreatment with 5 - ASA.
Hepatic lipid peroxidation, and the amounts of total lipid, total chloesterol, and triglyceride of the liver were significantly increased by CCl_4 administration. These increases caused by CCl_4 administration were significantly reduced by pretreatment with 5 - ASA.
Renal lipid peroxidation, and and the amounts of total lipid, and triglyceride of the kidney were significantly increased by CCl_4 administration. These increases caused by CCl_4 administration were slightly reduced by pretreatment with 5 - ASA.
These results suggest that 5 - ASA exerts a chemoprotective effect against carbon tetrachloride toxicity in male rats by mechanism that appears to involve its antioxidants property, just like scavenger of free radicals, and detoxification of electrophilic compounds by the metabolism of carbon tetrachloride.
