The purpose of this study was to evaluated the effects of 0-carotene and Ganoderma lucidum on the carcinogenesis induced by implantation of 9, 10-dimethyl-1, 2-benzanthracene (DMBA) on the submandibular gland of the rats. Eighty spraque-Dawley rats, weighting 1 00-200gm were used as experimental animals; divided into 3 experimental groups. The left submandibular gland was exposed and a 4-6mg pellet of powdered DMBA was implanted into the glandular tissue. (group 1, 2, 3) 21 animals (group 2) were supplied with ganoderma lucidum extract (0.4cc/100gmjday), administrated orally 3 times a week. In group 3, 21 animals were supplied with ganoderma lucidum extract and a-carotene (0.4cc/100gm/day) at the same time. Three animals in the each experimental group and 2 animals in control group were sacrificed at 4, 6, 8, 10, 12, 14, 16 weeeks after experiments serially.
While the animals were sacrified, the left submandibular glands were removed and fixed in the 10% formalin, sectioned in paraffin and stained with the Hematoxylin-eosin.
The results were as follows;
1. DMBA implantation (group I) revealed epidermoid cyst with dysplasia 4 weeks later, carcinoma in situ at 6-8 weeks, invasive growth at 10 weeks, well differentiated squamous cell carcinoma at 12 weeks and poorly differentiated squamous cell carcinoma at 16 weeks.
2. Group 2 revealed carcinoma in situ at 10 weeks, invasive growth at 12 weeks, highly differentated squamous cell carcinoma at 14 weeks, well differentiated squamous cell carcinoma at 16 weeks, it revealed highly inhibiting effect on carcinogenesis than group 1.
3. Group 3 revealed dysplasia of epithelium from 4 weeks to 8 weeks, carcinoma in situ from 10 weeks to 14 weeks, and early invasive growth and highly differentiated squamous cell carcinoma at 16 weeks. It revealed that group 3 was highly effective on carcinogenesis than that of group 2.
4. Control group (a-carotene and ganoderma lucidum) showed normal appearrance in the most cases, especially little capillary congestion and irregular duct were shown from 12 weeks to. 16 weeks.
