알코올 및 H₂O₂ 유도 신경세포 사멸에 대항하여 참취와 초석잠 함유 빵의 뇌신경세포 보호 효과

국문 초록 (Abstract)

본 연구에서는 참취(Aster scaber Thunb., AS), 초석잠(Stachys sieboldii Miq., SS) 그리고 참취와 초석잠을 1:1로 섞은 분말(ASM)의 70% 에탄올 추출물(ASE, SSE, ASM)의 항산화 활성에 의한 신경 보호 효과를 알아보았다. ASE, SSE 및 AS-M은 농도 의존적으로 DPPH 라디칼을 제거하였으며, 이들은 세포 내 활성산소종을 강하게 억제함으로써 사람의 신경모세포종 SK-N-SH에서 H₂O₂ 또는 에탄올에 의해 유도된 세포 독성을 억제하여 세포를 보호하였다. AS-M은 ASE 및 SSE보다 더 높은 항산화 활성 및 뇌신경세포 보호 효과를 나타내었다. 계속되는 연구에서 SKN-SH 세포에서 H₂O₂ 또는 에탄올에 의한 세포 독성에 대한 AS, SS 및 ASM을 함유한 빵 추출물(ASB, SSB 및 AS-MB)의 뇌신경세포 보호 효과를 알아보았다. ASB, SSB 및 ASMB 각각의 처리는 SK-N-SH 세포에서 H₂O₂ 또는 에탄올에 의한 세포 독성을 감소시키고 세포 내 활성산소종 수준을 감소시켰다. 참취로부터 분리된 주요 화합물(astragalin과 isoquercitrin) 처리는 HO-1 항산화 유전자의 발현을 증가시키므로 에탄올에 의해 유도된 세포사멸에 대한 보호 효과를 나타내었다.

The present study evaluated the neuroprotective effects of Aster scaber Thunb. (AS), Stachys sieboldii Miq. (SS), and their mixture at a ratio of 1:1 (ASM) based on antioxidant activity. The 70% ethanol extracts of AS, SS, and ASM (ASE, SSE, and AS-M, respectively) scavenged DPPH radical in a dose-dependent manner and strongly suppressed cellular reactive oxygen species, thereby protecting against H₂O₂- or ethanol-induced cytotoxicity in human brain neuroblastoma SK-N-SH cells. AS-M showed higher antioxidant activity and neuroprotective effects than ASE and SSE. In a continuing study, the neuroprotective effects of extract of bread containing AS, SS, and ASM (ASB, SSB, and AS-MB, respectively) against H₂O₂- or ethanol-induced cytotoxicity in SK-N-SH cells were investigated. Treatment with ASB, SSB, and AS-MB reduced cytotoxicity of H₂O₂ or ethanol in SK-N-SH cells and reduced the intracellular reactive oxygen species level. Treatment with main compounds (astragalin and isoquercitrin) isolated from Aster scaber Thunb. showed protective effects against ethanol-induced cell death by up-regulation of the HO-1 antioxidant gene.
