The purpose of this study was to evaluate the effect of beta-aminoproprionitrile (BAPN) intoxication on the periodontium of the rat.
Seventy senven Spraque-Dawley white rats weighing about 100gm were divided into experimental group (55 rats) and control group (22 rats), and experimental group were subdivided into subcutaneous injection group and oral administration group.
The animals of subcutaneous injection group recieved each day, 6 days weekly, subcutaneous injection of 30mg of BAPN per 100gm of body weight and the animals of oral administration group were given each day, 6 days weekly, Ice of 25mg per 1000cc aqueous solution of BAPN by intragastric tube.
Five animals in each experimental groups and 2 animals in each control groups were sacrificed at 3, 5, 7, 9, II, 13 days after subcutaneous injection and- 1,2, 3, 4, 5 weeks after oral administration.
After sacrifice, the mandibles were fixed in 10% neutral formaiin, decalcified in formic acid, embedded in paraffin, sectioned at 5-7 micron, and stained with the following method; hemstoxylin-eosin, periodic acid-Schiff, Van Gieson, Masson's trichrome, Gomori's silver, and toluidine blue stain.
The result were as follows;
1. The characteristic histopathologic finding of experimental group was acellular eosinophilic foci surrounded by palisading fibroblasts; known as "lathyritic bodies", closely resemble the "Verocay bodies" of the neurolemmoma and the pseudorosettes of the neuroblastoma.
2. The earlist lesions were seen 5 days after subcutaneous injection of BAPN and 2 weeks after oral administration. Starting in the center of the periodontal ligament, they extended apically and gingivally, involving the entire periodontal ligament by end of experiment.
3. Alveolar bone was apt to change into immature woven bone during experiment, and root and alveolar bone resorption was found at the end of experiment.
